MicroRNAs (miRNAs) are a class of non-coding RNAs that post-transcriptionally inhibit gene expression. In this study, we discovered that microRNA-370 (miR-370) was down-regulated in endometrioid ovarian cancer cells. In IGROV1 and TOV112D endometrioid ovarian cancer cells, miR-370 suppressed cellular viability and colony formation. miR-370 also enhanced endometrioid ovarian cancer cell chemosensitivity to cDDP. Endoglin (ENG) was directly and negatively regulated by miR-370. In addition, hypermethylation was a potential mechanism of miR-370 epigenetic silencing. We conclude that miR-370 acts as a tumor suppressor in endometrioid ovarian cancer via ENG regulation. © 2014 Elsevier Ireland Ltd.

In this work, an ultrasound-assisted procedure for the extraction of Lingzhi polysaccharides was established. Response surface methodology (RSM) was used to optimize the ultrasound-assisted extraction parameters (ultrasonic frequency (X1), extraction temperature (X2), extraction time (X3), and ratio of water to raw material (X4)) for enhancing the forward extraction efficiency of Lingzhi polysaccharides by implementing a three-level, four-variable Box-Behnken experimental design. The independent variable with the largest effect on response was X1 (ultrasonic frequency), followed by X2 (extraction temperature) and X3 (extraction time). The optimum extraction conditions were found to be ultrasonic frequency 8 kHz, extraction temperature 95 °C, extraction time 3 h and ratio of water to raw material 12. Under these conditions, the forward extraction efficiency of Lingzhi polysaccharides can increase from 42% to 75%. Pharmacological experiment showed that Lingzhi polysaccharides could decrease CyclinB1 mRNA expression in CaSki cells and inhibit CaSki and HeLa cells proliferation. Results indicated that Lingzhi polysaccharides possessed strong antitumor activity. © 2010 Elsevier Ltd. All rights reserved.

Orthogonal array design (OAD) was applied for the first time to optimize ultrasonic-assisted extraction (UE) of polysaccharides from Poria cocos (PCP). An analysis of variance technique was employed as the data analysis strategy in this study. PCP were successfully extracted from P. cocos under the optimum UE conditions: 75 min extraction time, 70 mesh particle size, and 90 °C extraction temperature. An analytical method was developed and validated for the determination of chemical components of PCP by high performance liquid chromatography (HPLC). Wistar rats were fed for 40 days with PCP. The total antioxidant status of organs was assayed by monitoring antioxidant enzyme activities. Results indicated that PCP could increased the level of GSH and activities of antioxidant enzymes in Wistar rats. These results confirm the effectiveness of PCP supplementation in detoxifying free radicals that are produced excessively in rats with cervical cancer. © 2009 Elsevier Ltd. All rights reserved.
