Objectives: This study aimed to investigate the expression levels of T-cell immune response cDNA 7 (TIRC7) in immune thrombocytopenia (ITP) patients before and after high-dose dexamethasone (HD-DXM) treatment.Methods: Forty-four patients with ITP were enrolled and received dexamethasone (40 mg/day) for 4 consecutive days. Patients who had platelet counts more than 50 × 109/l or less were defined as responders or non-responders, respectively. Quantitative polymerase chain reaction and enzyme-linked immunosorbent assay were used to measure RNA level and plasma level of TIRC7, respectively.Results: TIRC7 levels (RNA and plasma level) were significantly higher in ITP than that in control (P < 0.0001). However, after treatment, TIRC7 levels were significantly decreased in responders (P < 0.0001) but not in non-responders (P > 0.05).Discussions: TIRC7 might be associated with the pathogenesis of ITP, and TIRC7 levels could be used as an indicator to evaluate patients’ response to HD-DXM treatment. © W. Aims: To evaluate the safety and efficacy of percutaneous closure (PC) using modified double-disc ventricular septal defect (VSD) occluders in patients with ruptured sinus of Valsalva aneurysm (RSVA). Methods and results: Between 2005 and 2012, 25 patients were enrolled in our study for attempted PC. There were five patients (20%) with RSVA combined with VSD and two (8%) with aortic regurgitation (AR). The median age was 45 years (24-74 years). The QP/QS was 2.4±0.6 (1.6-4.0). The median diameter of lesions was 6 mm (4-8 mm) while the median size of occluders was 8 mm (6-12 mm). Twenty-three patients (92%) underwent PC successfully. Occluders were retrieved in two patients within one week. Trivial residual shunts developed in three and mild occluder-related AR occurred in five at discharge. During a median follow-up of 19 months (6-96 months) all trivial residual shunts vanished and mild occluder-related AR disappeared in four out of five patients. The proportion of patients in NYHA Class III/IV was reduced from 72% at baseline to 12% at the time of last follow-up (p<0.001). Conclusions: In selected patients with RSVA, PC using modified double-disc occluders may become a valuable alternative to surgery with encouraging immediate outcomes and midterm results. © Europa Digital & Publishing 2014. Objective: To isolate Toxoplasma gondii (T. gondii) strains from stray cats and explore their prevalence in Xuzhou City. Methods: The sera of 41 stray cats were collected to detect the antibodies of T. gondii by using a commercial enzyme-linked immunosorbent (ELISA) kit. The tissues including the heart, brain and tongue from these cats were digested by acid pepsin solution and inoculated to Kunming mice. These suspicious isolates were subsequently identified by a specific PCR method. Results: A total of 11 strains were isolated from 41 stray cats, which were confirmed by the PCR results. Moreover, 17 cats (41.5%) were found to be positive with the antibodies of T. gondii. Conclusion: A high prevalence of T. gondii infection was found in Xuzhou City, which indicates that the stray cats infected with T. gondii would be an important infection source that may infect humans and other animals in this area. © 2014, Editorial Office of Chinese Journal of Schistosomiasis Control. Schisandra chinensis contains schisandrin, deoxyschisandrin, r-schisandrin, and other active substances, which have many medical functions such as neuroprotective effects, anticancer function, etc. This study, using High-Performance Liquid Chromatography (HPLC) method, determined the contents of three active ingredients, established an Evaluation of Uncertainty factor mathematical model for HPLC method detecting these three active contents in Schisandra, and fully analyzed the uncertainty sources in the measurement process. Then, uncertainty was evaluated and calculated. The results showed that when the sample amount was 2.00 mg the determination of schisandrin and deoxyshisandrin in Schisandra shows a good linear relationship. In conclusion, the method developed using high-performance liquid chromatography in this study is suitable for uncertainty factors’ assessment in the determination of schisandrin, deoxyshisandrin, and r-schisandrin in Schisandra. Ginseng is one of famous herbs, which has many medical functions, such as neuroprotective effects, and anticancer function, because ginseng contains many active substances, and threonine is one important ingredient. In this study, to establish a mathematical model of uncertainty assessment for Threonine content in Ginseng by the amino acid assay, the sources of uncertainty in the measurement process were completely concerned, the main sources of uncertainty were fully analyzed, and evaluated and calculated. The results showed, for 0.2083g sample, the Threonine determination in Ginseng showed a good linear relationship. IN conclusion, this method developed in this study is suitable for the Uncertainty factors assessment of threonine measurement in Ginseng by amino acid assay. We also did animal test. To detect the immune activity of threonine in Ginseng, we detected the CD3+, CD19+ using FACS of mice after oral administration. © Qin et al. Source
