Ubiquitin which has the function of selective protein degradation may play an important role in the regulation of insect growth and development. The coding sequence of an ubiquitin gene from the larvae of the rice stem borer, Chilo suppressalis Walker (Lepidoptera: Pyralidae) named CsUB (GenBank Accession No. GU238420) was cloned by RT-PCR and sequenced in this study, with primers according to the sequences of ubiquitin genes from Homo sapiens, Drosophila melanogaster and Lepidopteran insects. Sequence analysis showed that the length of the coding sequence is 228 bp, encoding 76 amino acids with calculated molecular weight of 8.50 kDa and the theoretical isoeletric point of 5.26. Signal sequence and transmembrane domain had not been found. Multiple sequence alignment indicated that CsUB gene sequence with other known gene sequences of invertebrates and vertebrates had a high degree of homology (more than 72% similarity) and a shorter genetic distance (lower than 0.360). During the genetic diversity analysis, the total of 104 polymorphic sites was detected from 18 ubiquitin gene sequences and 18 haplotypes were sorted. Abundant genetic diversity and strong codon usage bias were found by the haplotype diversity (1.000), average number of nucleotide differences (47.475), nucleotide diversity (0.20866), effective number of codons (44.526), codon bias index (0.559) and scaled Chi-square (0.779). The predicated secondary structure composition of CsUB protein had about 32.89% extended strands, 36.84% random colis, 15.79% alpha helixes and 14.47% beta turns. Subcellular localization analysis showed that CsUB protein of cytoplasm, cell nucleus, mitochondrion, cell skeleton and plasma membrane occupied about 47.80, 26.10, 17.40, 4.30 and 4.30%, respectively. Sequence, homology and structural analysis confirmed that CsUB gene was highly conserved during evolution and belonged to ubiquitin gene family. The results might provide some fundamental data for further studies on expressed characteristics and physiological functions of CsUB gene. © 2011 Academic Journals.

Based on the published sequences of MT genes from A. bisporus, G. lucidum, T. camphoratus and P. involutus in NCBI database, their molecular structures, physiological functions and evolutionary relationship were analyzed using the bioinformatics methods. Results showed that AbMT, GlMT, TcMT and PiMT genes contained complete ORF and their theoretical points of the last three encoding proteins were higher than 7.0. AbMT, GlMT, TcMT and PiMT were relatively rich in random coil and extended strand but transmembrane helices and signal peptides were not found. 4 MTs were mainly localized in cell nucleus (over 60%) and their cellular functions might have some relation to central intermediary metabolism. Multiple sequence alignment indicated relatively high identity (more than 52%) and short genetic distances (lower than 0.900) among 4 MT nucleotide sequences. Abundant genetic diversity and strong codon bias were found based on the halotype diversity, average number of nucleotide differences, nucleotide diversity, effective number of codons, codon bias index and scaled chi-square. Simultaneously, we deduced that 4 MT genes during molecular evolution were under positive selection. The present study might provide basis for further investigation of MTs molecular mechanisms and genetic laws.

TiN coatings were deposited by Magnetron Sputter Ion Plating (MSIP) on the surface of high-speed steel W18Cr4V.The microstructure,coating thickness, micro-hardness and wear resistance of surface modification sample were investigated by the D/MAX-IIIB X-ray diffraction tester, the coating thickness tester, the HV-1000 micro-sclerometer,the LKDM-2000 friction and wear tester,the S-3000N scaning electron microscopy.The influence of sputtering time,N2 partial pressures and sputtering current on the microstructure,coating thickness, micro-hardness and wear resistance of TiN coatings were also analysed. The composite electroslag remelting technology was prepared with Cr12Mo1V1 steel as matrix, and the WC particles as hard phase steel bonded carbide. By means of metallographic analysis and scanning electron microscopy and other methods to study the microstructure and grinding crack morphology of the composites, and rockwell hardness and friction and wear experiment was carried out. The results show that WC particles had no obvious slant phenomenon, partly dissolved in the steel matrix, Fe3W3C multiple carbides precipitated after cooling. And the external hardness and abrasion resistance fo the roll were higher than the core, combined with good WC particles and steel matrix, and wear surface grinding marks was small, shallow furrowed, improved the roll wear resistance and service life. © (2014) Trans Tech Publications, Switzerland.

The rice stem borer, Chilo suppressalis Walker, is an economically important pest distributed in rice producing regions of China. To understand its molecular adaptation, the authors cloned the cDNA sequence of CcsHsp90 which is essential for dealing with many environmental stresses in all organisms and measured the mRNA expression in the diapausing larvae exposed to temperature stress. The results showed that CcsHsp90 (GenBank accession no. FJ866608) is potentially encoded to 312 amino acids with calculated molecular weight of 35.59KDa and the theoretical isoeletric point of 4.72. The deduced amino acid sequence of CcsHsp90 is displayed over 80% of the degree of conservation to other Lepidopteran insects. The relative mRNA expression levels of CcsHsp90 in the diapausing larvae which were exposed to different temperatures from 28 °C (CK) to -14 °C with a gradient of 7 °C for 4h increased first and decreased later and peaked at O "C. Then the larvae were exposed to O 7deg;C for 0-8h, the change of the levels was the same as the above and peaked at 4h treatment. Therefore, it could be concluded that O °C for 4h treatment might induce the maximal expression of CcsHsp90 in the diapausing larvae of the rice stem borer.

To understand the physiological mechanisms of temperature stress on the diapause larvae of rice stem borer Chilo suppressalis Walker at physiological and biochemical levels, determinations were made on the contents of water, lipid, total sugar and low molecular mass carbohydrates and the activities of SOD, POD, CAT in the larvae after series temperature stress (STS) and gradient temperature stress (GTS). With the decrease of temperature, the water content in the larvae decreased, and the decrement below 0°C was significantly larger in treatment GTS than in treatment STS. The lipid content in the larvae decreased gradually, but no significant difference was observed between treatments STS and GTS. The total sugar content in the larvae in treatment STS increased after an initial decrease, but that in treatment GTS continued to decline. Four species of low molecular carbohydrates, i. e., trehalose, glucose, glycerol and fructose were detected in the larvae. In treatment STS, the contents of glycose, glycerol and fructose in the larvae decreased after an initial increase, while the trehalose content was in adverse. In treatment GTS, the trehalose content decreased first and increased then, the glucose and glycerol were in adverse, but the fructose content had little change. In the range from 14 to -14°C, the SOD and POD activities in the larvae in treatment STS were significantly lower than those in treatment GTS, but the CAT activity was in adverse. The changes of these indices reflected the physiological responses of C. suppressalis diapause larvae to different temperature stress.

Decorin (DCN), a component of the extracellular matrix (ECM), participates in ECM assembly and influences cell proliferation and apoptosis in many mammalian tissues and cells. However, expression and function of DCN in the ovary remain unclear. This study cloned the full-length cDNA of goat DCN obtained from the ovary of an adult goat. Sequence analysis revealed that the putative DCN protein shared a highly conserved amino acid sequence with known mammalian homologs. The tissue distribution of DCN mRNA expression was evaluated by real-time PCR, and the results showed that DCN was widely expressed in the tissues of adult goat. Immunohistochemistry results suggested that DCN protein existed in the granulosa cells and oocytes from all types of follicles and theca cells of antral follicles. Moreover, hCG-induced DCN mRNA expression was significantly reduced by the inhibitors of protein kinase A, PI3K, or p38 kinase (P < 0.05), which are key mediators involved in hCG-induced DCN expression. Overexpression of DCN significantly increased apoptosis and blocked cell cycle progression in cultured granulosa cells (P < 0.05). Western blot analysis also showed that overexpression of DCN upregulated the expression levels of p21 protein (P < 0.05), whereas no effects were observed on the expression of Bax and Bcl-2 and on Bcl-2/Bax ratio (P > 0.05). These findings suggested that DCN regulates the apoptosis and cell cycle of granulosa cells. © 2016 Elsevier Inc.

Cutting tooth especially due to the poor quality of the shearer brazing, caused enormous economic losses each year in our country, which caused great pick consumption and delayed hours, and then resulting in reduced efficiency. The tooth body wear of the whole shearer picks accounts for half of its overall failure. In this experiment, the heat treatment which included heating temperature, soaking time, and cooling, etc. was designed by service conditions and performance requirements of the body material 40Cr. The change of microstrcture and mechanical property was analysed by quenching and tempering treatment, thus prolong the service life of shearer picks by improving the comprehensive performance of the tooth body material. © (2013) Trans Tech Publications, Switzerland.

The hydraulic cylinder is the essential component in the construction machinery, and the quality and safety of construction machinery products depends on its performance. As the main part of the hydraulic cylinder rod. the quality and reliability are the key indicators of the hydraulic cylinder, hi this paper, the hard chrome plating layer of hydraulic cylinder rod was tested by metallographic analysis, surface morphology observation, hardness test, corrosion resistance experiment, in order to evaluate the anticorrosion, economy and technology of the electroplating technology, and discuss the anticorrosion mechanism, so the experimental and theoretical basis will be provided in the field of production. The contribution will be made by improving the corrosion resistance and wear resistance, increasing the operational reliability, extendeding service life of the hydraulic cylinder rod. © (2013) Trans Tech Publications, Switzerland.
