Investigated have been the properties of atoxigenic Vibrio cholerae strains, O-antigen producers, after extended storage for 36 months. Determined is the fact that their cultural-morphological characteristics along with physiological ones remain unaffected, and the levels of O-antigen production are fully retained.Worked out is an experimental technology for submerged cultivation of cholera vibrio atoxigenic strains that allows for the obtainment of larger quantities of native O-antigen preparations than in the case with virulent strains-producers. Identified are the conditions that contribute to the maximum O-antigen generation: cultivation temperature — 37 °C; termination of the microorganism cultivation at the stage of transition from growth phase to stationary phase; application of organic and/or mineral defoaming agents. The technology for O-antigen concentration using tangential ultrafiltration has been put to an evaluation test. O-antigen preparations obtained from atoxigenic V. cholerae strains using this experimental technology have similar to the alternative ones, obtained from toxigenic strains in accordance with certified technology, protein and polysaccharide composition. The newly constructed prototypes of the chemical cholera vaccine, manufactured using O-antigens of atoxigenic V. cholera strains, are in compliance with the requirements for the national chemical cholera vaccine. © 2015.
