Using a TV device for studying microcirculation (×40), we analyzed the density of the whole microvascular network and the density of arterioles in the pia mater of the sensorimotor cortex in SHR rats of different ages (3-4 and 12 months) after intracerebral transplantation of human mesenchymal stem cells. We found that the density of pial microvascular network in SHR rats receiving transplantation of human mesenchymal stem cells increased to a level observed in young Wistar-Kyoto rats. © 2017 Springer Science+Business Media, LLC

The safety and tolerance of treatment with autologous pluripotent mesenchymal stem cells (MSC) in multiple sclerosis were assessed in eight patients using autologous MSC harvested from the red bone marrow. MSC growth, immunophenotypes, and karyotypes, sterility, the absence of contaminating hematopoietic cells, chromosomal aberrations, and signs of aging were monitored during cultivation. Introduction of MSC into patients' blood was performed in accordance with a protocol consisting of short intravenous infusions at a dose of 2·106 per kg body weight, once every 30 days. Treatment duration was 4-8 months. Treatment efficacy was evaluated at 4, 8, and 12 months. Repeated intravenous infusions of autologous MSC were tolerated well by all patients and no significant side effects were seen either in the early or the late post-treatment phases. A clear positive effect was seen in some cases within two months from the start of treatment. Five of eight patients showed 0.5-point improvements on the EDSS at four months. At 12 months, observations revealed improvements on the EDSS by 0.5-1 point in six of the eight patients, stabilization in one, and progression in one. These results provide evidence of the safety of this treatment and its moderate clinical efficacy in multiple sclerosis patients who are incurable using traditional approaches. © 2012 Springer Science+Business Media, Inc.

We performed morphological analysis of the structure of rat hippocampus after ablation of the left sensorimotor cortex. Four experimental groups were formed: two control groups (intravenous and intracerebral injections of the culture medium) and two experimental groups (intravenous and intracerebral transplantation of MSC). Ten weeks after surgery, disturbed cytoarchitectonics and great number of dead neurons were found in all zones of the hippocampus in animals of the control groups. In animals receiving cell therapy, no pathological changes in the structure of the hippocampus were found: hyperchromatic neurons were absent and the cells had regular shape and closely adjoined to each other. © 2012 Springer Science+Business Media, Inc.

We studied the effect of platelet lysates from different donors on fi broblast growth in culture. In most samples (40 of 50), the growth-stimulating characteristics were greater than in 10% FCS, but every ninth sample exhibited low mitogenic activity. A weak dependence between platelet concentration and total protein content was noted, but no correlation was found between these parameters and fi broblast growth in culture. © 2011 Springer Science+Business Media, Inc.

We studied the effect of various methods of transplantation of mesenchymal stem cells on neuronal survival in rat brain 1 and 6 weeks after severe traumatic brain injury. It was found that intracerebral and systemic transplantation of mesenchymal stem cells improves neuronal survival in the piriform cortex of the contralateral hemisphere without affecting neuronal survival in the marginal zone of the traumatic cavity and amygdaloid nuclei. Intracerebral transplantation of mesenchymal stem cells increases the content of the astroglial component of the scar in the borderline zone of the traumatic cavity. © 2011 Springer Science+Business Media, Inc.
