OBJECTIVE: This study assessed the relationship between the p53-dependent apoptosis and recurrent spontaneous abortion (RSA). PATIENTS AND METHODS: Thirty women with recurring miscarriages were enrolled as experimental group, and 30 women with normal reproduction served as control group. Immunohistochemistry was used to evaluate expression of p53 in villous tissue specimens. Further, expressions of CDKN1A and Bax mRNAs were evaluated by qPCR. TUNEL assay was utilized to document cell apoptosis. RESULTS: Expression of p53 was significantly increased in chorionic villi of patients in experimental group (p < 0.05 vs. control group). Further, CDKN1A and Bax mRNA levels were elevated in experimental group (p < 0.05 vs. control group), and the cell apoptosis index was increased as well. CONCLUSIONS: The p53-CDKN1A and p53- Bax signaling pathways appear to be activated in RSA. Thus, the apoptosis pathways controlled by p53 may be involved in the pathogenesis of RSA.

Background: Chromosomal and genomic aberrations are common features of human cancers. However, chromosomal numerical and structural aberrations, breakpoints and disrupted genes have yet to be identified in esophageal squamous cell carcinoma (ESCC).Methods: Using multiplex-fluorescence in situ hybridization (M-FISH) and oligo array-based comparative hybridization (array-CGH), we identified aberrations and breakpoints in six ESCC cell lines. Furthermore, we detected recurrent breakpoints in primary tumors by dual-color FISH.Results: M-FISH and array-CGH results revealed complex numerical and structural aberrations. Frequent gains occurred at 3q26.33-qter, 5p14.1-p11, 7pter-p12.3, 8q24.13-q24.21, 9q31.1-qter, 11p13-p11, 11q11-q13.4, 17q23.3-qter, 18pter-p11, 19 and 20q13.32-qter. Losses were frequent at 18q21.1-qter. Breakpoints that clustered within 1 or 2 Mb were identified, including 9p21.3, 11q13.3-q13.4, 15q25.3 and 3q28. By dual-color FISH, we observed that several recurrent breakpoint regions in cell lines were also present in ESCC tumors. In particular, breakpoints clustered at 11q13.3-q13.4 were identified in 43.3% (58/134) of ESCC tumors. Both 11q13.3-q13.4 splitting and amplification were significantly correlated with lymph node metastasis (LNM) (P = 0.004 and 0.022) and advanced stages (P = 0.004 and 0.039). Multivariate logistic regression analysis revealed that only 11q13.3-q13.4 splitting was an independent predictor for LNM (P = 0.026).Conclusions: The combination of M-FISH and array-CGH helps produce more accurate karyotypes. Our data provide significant, detailed information for appropriate uses of these ESCC cell lines for cytogenetic and molecular biological studies. The aberrations and breakpoints detected in both the cell lines and primary tumors will contribute to identify affected genes involved in the development and progression of ESCC. © 2012 Hao et al.; licensee BioMed Central Ltd.

Objectives: The present study is to evaluate the expression level of enhancer of zeste homolog 2 (EZH2) and vascular endothelial growth factor (VEGF), and analyze their correlations with clinicopathological characteristics and survival in patients with clear cell renal cell carcinoma (CCRCC). The effect of EZH2 on apoptosis and cell proliferation in 786-O renal cancer cell line is investigated.Methods: The expression level of EZH2 and VEGF was detected in 185 primary CCRCC patients’ tissues using tissue microarray and immunohistochemistry. Small interfering RNA or enhanced green fluorescent protein transfection was employed to investigate the effect of EZH2 inhibition or overexpression on VEGF expression, apoptosis and cell proliferation in 786-O cells using flow cytometry, immunofluorescence microscopy, quantitative real-time reverse-transcription polymerase chain reaction and Western blot analysis.Results: High expression level of EZH2 and VEGF was observed in advanced CCRCC and correlated with the TNM stage (p = 0.013, p = 0.001) and distant metastasis (p = 0.011, p = 0.038), respectively. EZH2 was positively correlated with VEGF in CCRCC tissues (correlation coefficient = 0.850, p < 0.001). Kaplan–Meier survival analysis revealed that patients with positive EZH2 expression had a shorter overall survival time compared to patients with negative EZH2 expression (34.3 vs. 67.2, p < 0.001). In 786-O cells, EZH2 silencing inhibited VEGF expression and cell proliferation while increasing apoptosis (p < 0.001). EZH2 overexpression promoted VEGF expression and cell proliferation while inhibiting apoptosis (p < 0.001).Conclusions: EZH2 correlates positively with VEGF and associates with adverse clinicopathologic characteristics and shorter survival time in CCRCC patients. EZH2 accelerates antiapoptosis and cell cycle in 786-O cells. © 2014, Federación de Sociedades Españolas de Oncología (FESEO).

We observed the effect of Dianxianning, which was used as additive treatment to treat 206 epilepsy patients, on the epilepsy seizure rate. Based on a multicenter, prospective, randomized, and controlled clinical trial design, we used the seizure rate of epilepsy as the main index. For the treatment group comprising 137 patients, we combined Dianxianning with chemical medicine, which is the basic treatment. For the control group with 69 patients, we added placebo. The results showed that 1) Effect on seizure rate: After a three-month treatment, the seizure rate of the treatment group decreased by 37.84% on average, whereas that of the control group decreased by 13.18% on average. Statistically comparing the two groups, there was a significant difference between these groups (P < 0.05). 2) Effect on seizure frequency: As time passed, the frequency in each group gradually decreased. After a three-month treatment, there was a significant difference between the two groups (P < 0.05). 3) Comparison between the before and after treatment of each group: There was a very significant difference between the two groups (P < 0.0001). The results indicated that, as an additive treatment, Dianxianning has a good effect on controlling the epilepsy seizure rate and frequency management. VirG is outer membrane protein of Shigella and affects the spread of Shigella. Recently it has been reported that apyrase influences the location of VirG, although the underlying mechanism remains poorly understood. The site of interaction between apyrase and VirG is the focus of our research. First we constructed recombinant plasmid pHIS-phoN2 and pS-(v1-1102, v53-758, v759-1102, v53-319, v320-507, v507-758) by denaturation-renaturation, the phoN2:kan mutant of Shigella flexneri 5a M90T by a modified version of the lambda red recombination protocol originally described by Datsenko and Wanner and the complemented strain M90TÄphoN2/pET24a(PhisphoN2). Second, the recombinant plasmid pHIS-phoN2 and the pS-(v1-1102, v53-758, v759- 1102, v53-319, v320-507, v507-758) were transformed into E. coli BL21 (DE3) and induced to express the fusion proteins. Third, the fusion proteins were purified and the interaction of VirG and apyrase was identified by pull-down. Fourth, VirG was divided and the interaction site of apyrase and VirG was determined. Finally, how apyrase affects the function of VirG was analyzed by immunofluorescence. Accordingly, the results provided the data supporting the fact that apyrase combines with the α-domain of VirG to influence the function of VirG. © 2014 by the authors licensee MDPI Basel Switzerland.

A novel brachytherapy instrument, β particle axillary odor therapeutic instrument (also known as β particle axillary odor applicator), was successfully developed. The instrument mainly contains six parts including irradiator, supporter, lifter, shield cover, controller and treatment trolley. A full coverage therapy performed by the applicator can eliminate the axillary odor of patient. In this method, the 90Sr/90Y β radioactive source located in the irradiator is used to irradiate apocrine sweat gland under patient's armpit. Preliminary clinical tests indicate that the therapeutic effect is definite whether immediate irradiation, or supplementary irradiation after operation for apocrine sweat gland. This instrument provides another painless, non-surgical and noninvasive treatment. It features a safe, effective, convenient and automatic operation.

A brachytherapy instrument, called β particle hemorrhoid therapeutic instrument or β particle hemorrhoid applicator was developed. It is mainly composed of radiator, locator, lifter, shield sleeve and controller. It can be used to treat internal piles, external piles, combined piles, anal fissure, anal sinusitis, etc. Preliminary clinical tests indicate that the advantages of β particle intracavitary radiotherapy are safe, effective, convenient, painless and non-invasive for curing hemorrhoids comparing with other techniques.
