The aim of the present study was to investigate the periodontitis-associated changes in the number, proliferation and differentiation potential of human periodontal ligament stem cells (PDLSCs). Cultures of human periodontal ligament cells (PDLCs) were established from healthy donors and donors with periodontitis. The numbers of stem cell were characterized using flow cytometry. PDLSCs were isolated from the PDLCs by immunomagnetic bead selection. Colonyforming abilities, osteogenic and adipogenic potential, gene expression of cementoblast phenotype, alkaline phosphatase activity and invivo differentiation capacities were then evaluated. Periodontitis caused an increase in the proliferation of PDLSCs and a decrease in the commitment to the osteoblast lineage. This is reflected by changes in the expression of osteoblast markers. When transplanted into immunocompromised mice, PDLSCs from the healthy donors exhibited the capacity to produce cementum PDLlike structures, whereas, the inflammatory PDLSCs transplants predominantly formed connective tissues. In conclusion, the data from the present study suggest that periodontitis affects the proliferation and differentiation potential of human PDLSCs invitro and invivo.

The present study aimed to investigate the expression and significance of p16 in the occurrence, development and regression of infantile hemangioma (IH). The expression of p16 was examined in proliferating, involuting hemangioma and normal tissues using immunohistochemical techniques. The expression of p16 was significantly lower in proliferating hemangioma than in involuting hemangioma, and was significantly lower in the involuting hemangioma than in normal tissues. Significant differences were found between the three groups (P<0.05). The results indicate that p16 may be important in the regression of IH endothelial cells and in anti‑angiogenesis. There is a certain association between p16 expression and the regression of hemangioma. This provides a theoretical basis for the further study of the pathological mechanisms of p16 in hemangioma and potential gene therapies that may treat this disease. © 2015, Spandidos Publications. Thrombosis and hemocyte damage are the main problems of applied non-coated biomaterials to cardiac surgery that remain unsolved. The present study is aimed at the chemical modification of polyvinyl chloride (PVC) for applications in cardiac surgery and the biological property assessment of modified PVC. Sodium alginate (SA)/ heparin (HEP) composites were covalently immobilized onto the surface of the PVC pipeline. The surface grafting density and protein adsorption were determined by ultraviolet spectrophotometry. The surface contact angles were evaluated by contact-angle measurement, whereas the surface characteristics were evaluated by Fouriertransform infrared spectroscopy. Blood coagulation time and platelet adhesion were measured using an automated blood coagulation analyzer and a hemocytometer, respectively. Surface morphologies of the thrombus and platelets were evaluated by scanning electron microscopy. The immobilization of SA/HEP reduced the contact angles of the coated surface. Protein adsorption was reduced by the immobilization of SA. The activated partial thrombin time and thrombin time of the coated PVC were significantly prolonged as compared with the non-coated PVC. Platelet adhesion and thrombus formation were all reduced by the immobilization of HEP. The results revealed that the SA/ HEP coating can improve the antithrombogenicity of the PVC pipeline, as well as improve its biocompatibility and hemocompatibility, which are essential for cardiac pulmonary bypass surgery.

AIM: To reveal the mechanism by which acupuncture treatment promotes gastrointestinal function recovery in acute abdomen disease. METHODS: Forty SD rats were randomly divided into five groups. Rats in the normal group were not given any surgery or treatment, and rats in other four groups (1- and 2-day model groups and 1- and 2-day treatment groups) were given an operation to obstruct the ileum by thread ligation. The two treatment groups were given acupuncture at Zusanli and Yanglingquan acupoints. Serum levels of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) were compared between different groups, and the rate of small intestinal transit was also compared. RESULTS: Serum concentrations of TNF-α and IL-6 differed significantly between the 1-day acupuncture group and 1-day model group, as well as between the 2-day acupuncture group and 2-day model group, although there was no significant difference between the 1- and 2-day acupuncture groups. The rate of small intestinal transit differed significantly between the 1-day acupuncture group and 1-day model group, between the 2-day acupuncture group and 2-day model group, as well as between the 1- and 2-day acupuncture groups. CONCLUSION: Acupuncture at Zusanli and Yanglingquan acupoints can increase serum levels of TNF-α and IL-6 and the rate of small intestinal transit in rats with incomplete intestinal obstruction. © 2015 Baishideng Publishing Group Inc. Solitary fibrous tumors (SFTs) are a rare type of neoplasm that originate from the pleura. Although SFTs may occur in a variety of extrathoracic regions, they are considered to be rare in the submandibular region. The current study presents the case of a 39-year-old female with a 3x4-cm, fibro-elastic, movable, painless nodule in the right side of the submandibular region. The patient exhibited no other clinical manifestations in the head and neck region. A computed tomography scan demonstrated the presence of a well-defined, slightly low-density nodular shadow measuring 2.6x3.3x3.8 cm, in proximity to the right submandibular gland, with mild contrast enhancement and no association with the adjacent lymph nodes. The lesion was surgically excised, and following histopathological and immunohistochemical analysis, immunohistochemical staining determined that the lesion was positive for cluster of differentiation (CD)34, CD99 and vimentin, and negative for desmin, CD31 and S-100; therefore, a diagnosis of an SFT was determined. The patient has so far been followed up for 22 months, with no signs of recurrence or metastases. The present study also discusses the clinical, histopathological and immunohistochemical features, treatment strategies and potential clinical outcomes of SFTs. The study proposes that, although rare, SFTs of the submandibular region should be included in the differential diagnosis of soft-tissue tumors in the submandibular region. © 2014, Spandidos Publications. Chronic hepatitis C is a globally serious medical problem, which has more rapid progresses into cirrhosis and hepatocellulor carcinoma without effective and less toxic novel treatments. Silymarin has been used to treat a range of liver disorders, and its main effects include antioxidant, antifibrotic, anti-inflammatory, immumoregulatory and hepatocellulor regeneration-promoting effects. Currently, investigators found that silymarin has anti-hepatitis C viral effects via indirectly inhibiting NK-κB-induced inflammation and immune responses, and direct antiviral effects against HCV infection. In some clinical trails, the intravenous formulation of silymarin has been found to decrease serum HCV-RNA titers and enhance IFN anti-HCV efficacy. Therefore, in the future, large scale clinical studies focusing on the antiviral effects of silymarin should be required.
