Objective: Insulin-like growth factor-2 (IGF-2) and pleckstrin homology-like domain, family A, member 2 (PHLDA2) appear to play an important role in paracrine interactions at the maternal-fetal interface in human pregnancy. But the potential predictive value of serum IGF-2 and PHLDA2 concentrations in pre-eclampsia remains to be established. The goal of the present study was to explore the expression of IGF2 and PHLDA2 in mid-pregnancy with pre-eclampsia (PE). Methods: Samples of serum were collected from women with normal mid-pregnancy women (control group, n=21), mild pre-eclampsia (M-PE group, n=19) and severe pre-eclampsia women (S-PE, n=19). The expressions of IGF2 and PHLDA2 in the serum were determined by ELISA and western blot. After delivery, the fresh placental tissues were collected. The expression levels of IGF2 and PHLDA2 in placenta were detected by real-time PCR, western blot and immunohistochemistry. Results: It was found that the concentrations of IGF2 and PHLDA2 were remarkably dropped in comparison with control group. And there was significant difference in PHLDA2 expression between mild pre-eclampsia and severe pre-eclampsia in serum. After delivery, the differences in imprinted genes IGF2 and PHLDA2 were found in placental tissues between PE and normal pregnancy. mRNA and protein expression levels were all obviously decreased in PE groups. Conclusion: These data are the first to demonstrate imprinted genes IGF2 and PHLDA2 associated with PE in mid-pregnancy. It illustrates that IGF2 and PHLDA2 may participate in PE pathogenesis. © 2017, E-Century Publishing Corporation. All rights reserved.

BACKGROUND: We have found that Xingnaojing can effectively inhibit mediators of inflammatory mediators and promote fibrinolytic activity in a rabbit model of myocardial ischemia and reperfusion. However, it is not completely clear what is the action of mechanism of fibrinolytic system activity in human umbilical vein endothelial cells. OBJECTIVE: To investigate the effects of Xingnaojing on recombinant human tumor necrosis factor α-mediated tissue-type plasminogen activator (t-PA) and plasminogen activator inhibitor-1 (PAI-1) expression in human umbilical cord vein endothelial cells. METHODS: The passages 3-5 of human umbilical cord vein endothelial cells were obtained, and were induced by adding 10 μg/L recombinant human tumor necrosis factor α in the culture medium. Xingnaojing in different concentrations (5, 10, 20 mL/L) were injected in the Xingnaojing group, and fluvastatin (1 μmol/L) was added as positive control group. A blank control group of simple human umbilical cord vein endothelial cells culture was set up. After 24 hours of cultivation, the production of t-PA and PAI-1 in the cultured medium was determined by ELISA. The mRNA expression of t-PA and PAI-1 was measured by reverse transcript-PCR. RESULTS AND CONCLUSION: Compared with the blank control group, the secretion and mRNA expression of PAI-1 in the group treated with recombinant human tumor necrosis factor α (10 μg/L) were significantly increased (P < 0.05); however, the secretion and mRNA expression of t-PA in the group treated with recombinant human tumor necrosis factor α (10 μg/L) were significantly decreased (P < 0.05). The secretion and mRNA expression of t-PA were significantly higher in the Xingnaojing group at different concentrations than in the recombinant human tumor necrosis factor α group (P < 0.05); however, the secretion and mRNA expression of PAI-1 were significantly lower in the former group than the latter group (P < 0.05), which was in a dose-dependent manner. Xingnaojing can effectively improve the fibrinolytic function of human umbilical cord vein endothelial cells mediated by recombinant human tumor necrosis factor α. © 2015, Journal of Clinical Rehabilitative Tissue Engineering Research. AIM: To investigate the morbidity of dry eye in policeman of Shunde district, and to analyze the related factors and proposed methods of preventing and treating dry eye. METHODS: Totally 650 policemen who came for physical examination in Shunde district recived questionnaire survey. The patients with dye eye disease were finally diagnosed through slit lamp examination, Schirmer test, tear film break-up time (BUT) measurement and keratoconjunctival fluorescent staining. The morbidity among different policemen was further analyzed and stasticed. RESULTS: The results showed that 152 in 650 policemen suffered from dry eye disease, the ratio was 23.4%. Comparied with security police and crimanl police, the ratio in traffic police, patrolman, and back office police was significantly higher. The prevalence was higher when the ages were greater than 40. CONCLUSION: Unsuitable working environment, air contamination are high risk factors of dry eye disease.
