NDM-1 (New Delhi metallo-beta-lactamase) gene encodes a metallo-beta-lactamase (MBL) with high carbapenemase activity, which makes the host bacterial strain easily dispatch the last-resort antibiotics known as carbapenems and cause global concern. Here we present the bioinformatics data showing an unexpected similarity between NDM-1 and beta-lactamase II from Erythrobacter litoralis, a marine microbial isolate. We have further expressed these two mature proteins in E. coli cells, both of which present as a monomer with a molecular mass of 25 kDa. Antimicrobial susceptibility assay reveals that they share similar substrate specificities and are sensitive to aztreonam and tigecycline. The conformational change accompanied with the zinc binding visualized by nuclear magnetic resonance, Zn2+-bound NDM-1, adopts at least some stable tertiary structure in contrast to the metal-free protein. Our work implies a close evolutionary relationship between antibiotic resistance genes in environmental reservoir and in the clinic, challenging the antimicrobial resistance monitoring. © Higher Education Press and Springer-Verlag Berlin Heidelberg 2011. AIM: To observe retinal ultrastructural features and retinal cell apoptosis of defocus induced myopia in guinea pigs.METHODS: Thirty guinea pigs (3-wk old) were received -10.00D rigid contact lenses (RGP) on the right eye and without wearing glasses on the left eye. Diopter change with cycloplegic retinoscopy, ultrastructural features of the retina with transmission electron microscopy were observed, the apoptosis of retinal cell by TUNEL immunofluorescence staining after 3wk were detected.RESULTS: Eye diopter difference was 3.28±0.21D in the right eye and 1.55±0.23D in the left eye before and after induction. There was significantly statistical difference (P<0.01); Retinal ultrastructural features were showed with transmission electron microscopy (TEM) the retinal disk membranes edema, some shedding, mitochondrial swelling and deformation, small amount of vacuolar changes in the inner and outer nuclear layer cells. Some characteristics of apoptosis were also be seen such as cell membrane irregular contractions, irregular nuclear chromatin aggregation. The incidence of retinal cell apoptosis was (2.42±1.24)% in the right eye and (0.29±0.08)% in the left eye after induction. There was significantly statistical difference (P<0.01).CONCLUSION: Accompanied by abnormal apoptosis of retinal cells during the formation of defocus-induced myopia in guinea pigs. Source

Background: Deep vein thrombosis after total knee arthroplasty has attracted increasing attention in recent years, but how to detect deep vein thrombosis in the early time in clinical practice remains unclear. Whether it is necessary to perform type-B ultrasonic or other invasive examination in lower limbs has become a hot issue. Objective: To explore the significance of D-Dimer and fibrin degradation products in the prediction of deep vein thrombosis after total knee arthroplasty. Methods: 56 patients received total knee arthroplasty were collected from Department of Orthopedics, The 306th Hospital of Chinese PLA, between December 2012 and February 2014. The D-Dimer and fibrin degradation products were dynamically monitored before operation and at 1, 3, 5, 7, 10 days post-operation. All the patients received type-B ultrasonic examination in double lower limbs at 10 days post-operation, and divided into thrombus group and non-thrombus group. The D-Dimer and fibrin degradation products in the two groups were compared. Results and Conclusion: Deep vein thrombosis was found in 13 cases by ultrasonic-B postoperation, D-Dimer and fibrin degradation products showed no significant difference between the two groups at 1 week after operation (P > 0.05), but the difference was significant at 10 days (P < 0.01). D-Dimer and fibrin degradation products index should be monitored dynamically for at least 10 days after operation, which is helpful for the early diagnosis of thrombosis. © 2015 Journal of Clinical Rehabilitative Tissue Engineering Research.
