To clarify the function of geranylgeranyl pyrophosphate synthase gene (NtGGPPS4) in tobacco, the coding sequence of NtGGPPS4 gene was cloned from Nicotiana tabacum. The over-expression vector pCAMBIA1300-NtGGPPS4 was constructed and transferred into Nicotiana tabacum by Agrobacterium tumefaciens mediated method. The contents of α-and β-cembrenediol and plastid pigments in transgenic tobacco plants were measured. The results showed that four transgenic tobacco plants which showed high levels of NtGGPPS4 were obtained. Comparing with the control, the contents of α-and β-cembrenediol, neoxanthin, violaxanthin, lutein, chlorophyll a, chlorophyll b and β-carotene increased significantly in transgenic plants. It indicated that NtGGPPS4 was involved in the biosynthesis of α-and β-cembrenediol and carotenoids in tobacco. © 2017, Editorial Office of Tobacco Science and Technology. All right reserved.

In order to study the imidacloprid residue and the influences of curing methods on its residue in tobacco, a method was developed for determining the imidacloprid residues in tobacco by QuEChERS-HPLC. A pre-treatment approach known as QuEChERS was followed: leaf samples were extracted with acetonitrile, and cleaned up with N-propyl ethylenediamine sorbent before analysis by HPLC. The results showed that: 1)The detection limit(LOD)of imidacloprid was 0.02 mg/kg, its average recoveries at three fortified levels(0.1, 0.5, 1.0 mg/kg) ranged from 88.6% to 96.6% with relative standard deviation of 2.05%-3.42%. 2)When the application rate of imidacloprid was 4 mg(a.i)/plant, the persistent period for controlling pests with piercing-sucking mouthparts was about 15 days. 3)After eliminating the influences of dry matter (matrix) consumption via determining the calcium mass fraction before and after tobacco curing, the imidacloprid residues in cured leaves were calibrated, and the calculated results indicated that the imidacloprid residues reduced by 47.96% and 48.66% after flue-curing and air-curing, respectively. © 2016, Editorial Office of Tobacco Science & Technology. In order to investigate the influences of reaction conditions on the formation of volatile compounds from a model Maillard reaction of glucose and glutamic acid in aqueous solutions, the changes of pH and UV-visible absorbance at different reaction conditions were analyzed. Furthermore, the generated volatile compounds under different reactant ratios, reaction times and solvents were identified by GC/MS. The results indicated that: 1) The UV-visible absorbance A294 nm and A420 nm increased with the increases of glutamic acid content in the model system, propanediol in solvent and reaction time, while A294 nm/A420 nm decreased. 2) The total amount of volatile compounds, and the yields of pyridines, pyrazines, pyrroles, furans, furanones, organic acids and carbocyclic compounds increased, while the yields of pyranones, esters and aliphatic compounds increased at first and then decreased with the prolonged reaction time. When the content of glutamic acid increased in glucose/glutamic acid model system, the total amount of volatile compounds and the generation of pyridines, pyrroles, furans, furanones, esters, carbocyclic compounds and pyranones increased. To further study the function of geranylgeranyl pyrophosphate synthase (GGPPS) gene from Nicotiana tabacum, NtGGPPS1 was successfully cloned from tobacco cultivar K326 using gene-specific primers which were designed according to the coding DNA sequence of GGPPS1 (GenBank: GQ911583.1) from Nicotiana tabacum. Fluorescent expression vector of NtGGPPS1 (PFF19-NtGGPPS1-GFP) was constructed for subcellular localization in tobacco. After treated with different phytohormones, the expression level of NtGGPPS1 in K326 was studied by real-time PCR. RNAi vector pHellsgate2-NtGGPPS1 was introduced into K326 via Agrobacterium-mediated transformation and the contents of plastid pigments in NtGGPPS1 down-regulated tobacco plants were detected. The results showed that green fluorescence protein NtGGPPS1-GFP fusion was distributed in the plastids of transgenic BY-2 cells, suggesting that NtGGPPS1 located in plastids. The expression level of NtGGPPS1 in K326 was significantly increased after MeJA treatment, while it was contrary to IAA treatment. Comparing with the control, the contents of plastid pigments significantly decreased in NtGGPPS1 down-regulated plants, which indicated that NtGGPPS1 was involved in the biosynthesis of β-carotene in Nicotiana tabacum. © 2016, Editorial Office of Tobacco Science and Technology. To improve the accuracy and stability in hardness testing of cigarette and filter rod, a novel hardness tester unit in test station was designed. The unit was mainly composed of a test platform, air cushion shaft, a mechanism for rod sample rotation, weights and sensors. The pressure was vertically loaded and the whole loading process was controlled by an air flotation device, and the rod hardness was measured by a laser meter displacement. A step motor was used in order to control loading speed accurately. The hardness of five standard articles and fifteen cigarette samples of different brands was measured by the novel test station and compared with two test stations in which the pressure was loaded horizontally. The results showed that: 1) Both pre-pressure and full pressure of the novel unit met standard's requirements. 2) The RSDs of hardness of the five standard articles were 0.005%-0.015% and 0.020%-0.035%, the measurement accuracies were 0.001-0.009 mm and 0.003-0.009 mm by vertical loading unit and horizontal loading unit, respectively; it indicated that vertical loading was superior to horizontal loading obviously. 3) The RSDs of hardness of the fifteen cigarette brands ranged from 3.25% to 5.85%, the results showed good stability and consistency. 4) The RSDs of novel and two other test stations were 2.93%-3.06%, 3.94%-4.33% and 3.70%-4.18%, respectively; novel test station was superior to the two horizontal loading testers. For analyzing the relationships between chemical components and sensory quality of tobacco leaves, an identification method for nonlinear relationship was developed based on generalized additive models. The linear and nonlinear relationships between four routine chemical components (total nitrogen, nicotine, total sugar and reducing sugar) and sensory evaluation indexes of 497 samples of middle leaves of flue-cured tobacco from main tobacco producing areas in China were studied. The results showed that: 1) At the significant level of 0.05, total nitrogen significantly negatively linearly correlated to aroma quality and quantity, offensive odor, irritancy and aftertaste; nicotine possessed a significantly negatively linear correlation with aftertaste and a significantly positively linear correlation with aroma quantity; total sugar and reducing sugar significantly positively linearly correlated to aroma quality and quantity, offensive odor, irritancy and aftertaste. 2) There were significantly nonlinear relationships between total nitrogen and aroma quality, offensive odor, irritancy, aftertaste; and between nicotine, total sugar, reducing sugar and aroma quality, aroma quantity, offensive odor, irritancy, aftertaste; all of them presented an obvious "∩"-shaped curve relationship; while the nonlinear relationship between total nitrogen and aroma quantity was not significant at the significant level of 0.05. 3) The appropriate ranges for total nitrogen, nicotine, total sugar and reducing sugar contents in flue-cured tobacco leaves were 1.5%-2.8%, 2.0%-3.5%, 15%-28%, and 15%-25%, respectively. ©, 2014, Editorial Office of Tobacco Science and Technology. Aiming at the problems of high breakage and poor processing durability of expanded cut tobacco by traditional CO2 expanded cut tobacco processing, a steaming preconditioning discharger was added next to the expansion tower, which moistened the tobacco as it left the tower. Steam was injected into the compartments in the airlock discharger through uniformly distributed nozzles, to allow the tobacco entering into the airlock being evenly humidified and loosed. The results of application showed that after improvement, the percentage of long strands and cut tobacco yield increased by 1.5 and 0.75 percentage points, respectively. Water stained tobacco was avoided in the process of humidifying. The processing durability of expanded cut tobacco was effectively improved, while the breakage of cut tobacco was reduced.

Long running time, high energy consumption and high running cost are the problems associated with the air conditioning system with primary recycling air in cigarette enterprises running under coordinate regulation mode in summer. Therefore a temperature-humidity separate control method for air conditioning system based on single cold source was developed by taking cut tobacco storing room as an example,meantime, the control units of the air conditioning system was improved as following: 1) Installing another surface cooler for dehumidification control and the original surface cooler was used as the secondary cooler for cooling control. The two surface coolers were connected in series, the input cold water of the secondary cooler come from the first cooler with 3-5℃ temperature rise to implement the separate temperature-humidity control by single cold source. 2) Adding an independent fresh air dehumidifying unit in the fresh air section to change the control mode from temperature and humidity coordinate regulation to separate control in summer. The results showed that the separate temperature-humidity control method effectively avoided the cold-heat offset and energy waste in temperature and humidity coordinate regulation mode in summer, the energy consumption of air conditioning unit in dehumidification decreased by 25.5%, and the running efficiency of air conditioning system was promoted. ©, 2015, Editorial Office of Tobacco Science and Technology. In order to reduce the difference of moisture content in output strips from the strip casing machine between batches, the casing machine was improved. To reduce the temperature fluctuation of and the casing droplets in circulated hot air, the inserted exhaust screen was replaced by a screen belt supported by side chains, a self-cleaning device for the screen belt was installed, and a set of casing droplets collecting and cleaning device was added. The results showed that the difference between moisture contents in cased strips of different batches, the extreme difference between batches decreased from 1.47 to 0.46 and was mostly controlled within ±0.5; the extreme difference of input cut tobacco to thin plate dryer between batches reduced from 1.98 to 0.58 and was mostly controlled within ±0.6. As a solution to the dithering of turnover body and high temperatures of hydraulic oil and driving motor, the positioning means of belt conveyor was optimized by a prior knowledge based LS-SVM (Least Square Support Vector Machine) theory. The control system for turnover body was improved by PLC programming; motor and hydraulic oil were cooled by axial flow fan and compressor refrigeration technology respectively; and the working pressure of hydraulic system was readjusted. The results showed that after improvement, the leakage of hydraulic oil and the dithering of turnover body were eliminated, the cracking in the supporting area of turnover body to the piston rod of oil cylinder decreased from 3-4 times/year to 0, the temperature of motor lowered from 65 to 44 ℃ and that of hydraulic oil lowered from 55 to 37 ℃. The failure rate of equipment was effectively reduced, while the reliability and safety of equipment running were promoted. © 2015, Editorial Office of Tobacco Science and Technology.
