Pseudomonas stutzeri was isolated from earthworm (Eisenia foetida) gut found to produce extracelluar siderophores when grown in succinic acid medium under iron deficiency. The siderophores were found to be pyochelin type. These siderophores were antagonistic to plant pathogenic fungi like Fusarium oxysporum, F.solani, F.moniliformae, F.udum, Macrophomena phaseolina, Rhizoctonia solani, Colletotrichum capsicii, Aspergillus flavus and A.niger. © Global Science Publications.

Biological nitrogen fixation involves the reduction of atmospheric dinitrogen (N2) into ammonia (NH3) by a metalloenzyme nitrogenase. The nitrogenase reductase (Fe-protein) isolated from Rhizobium strain was been purified and characterized. The nitrogenase Fe protein is a α2 dimer containing Fe4S4 cluster. The Fe-protein is similar to other Fe-proteins with respect to its molecular weight. The Fe-protein was eluted with 400mM-Nacl on to a DEAE-cellulose column as a dark-brown band at the top of the column. The molecular weight of the protein is about 60 K .Da. and the protein concentration is about 14.2mg/mL. © Global Science Publications.

N-acetylmuramyl-L-alanyl-D-isoglutamine (MDP) is the peptidoglycans obtained from the Mycobacterium tuberculosis. The isolate Mycobacterium tuberculosis (MTl) was obtained from the sputum of the tuberculosis patients. The MTl isolates undergone for the morphological, biochemical and staining technique for the conformation of Mycobacterium tuberculosis. The mid log phase MTl isolates were used for the extraction of MDP by using HPLC. The muropeptides were eluted from the HPLC with 2 to 30% linear gradient of aceto nitrile containing 0.5% formic acid at 320 L/min. The purified MDP was eluted with in the retention time of standard Muramyldipeptide.

The Nitrogenase Fe- protein was isolated from Rhizobium sps. which was purified and the properties similar to those of the Fe- Protein of other nitrogen fixing organisms with respect to its molecular weight, non-protein groups like Phosphates, Ribose and an adenine like unit are covalently bound to the protein. The nitrogense Fe protein is a α, dimer containing Fe1S1 cluster. The Fe- protein was eluted with 400 mM- Nacl on to a DEAE-Cellulose column as a dark brown band at the top of the column. The molecular weight of the protein is about 61500Da.

Enumeration of nitrifying bacteria from the Earthworm gut (Eisenia foetida) by a microtiter system based on the Most probable number (MPN) method has been discussed in this study. Higher MPNs were obtained with the microtechnique, with increased accuracy as compared to standard MPN tube technique. Our studies have shown that there are higher number of nitrifying bacteria associated with earthworm gut. Further, nitrification was detected by qualitative tests. © Global Science Publications.
