The discussion of applicability of existent mathematical models of ion exchange chromatographic process was continued in this article. It was shown in experiments that the column's geometry introduces an essential value into the ion exchange chromatographic process: the performance of the protein separation decreases with gel bed height increasing. An optimal gel bed configuration for this type of LC is a cylinder with height-diameter ratio from 1:1 to 0,1:1. Simultaneously buffer flow rate was proven to be one of the critical parameters for the calculated mathematical model approaching the real separation process. The reliability of the proteins (with close properties) separation performance from column geometry and elution buffer flow rate was demonstrated on Gloydius snake venom protein purification.
