Koi sleepy disease (KSD), also known as carp edema virus (CEV) disease, is an important, largely acute infection of common carp and ornamental koi (Cyprinus carpio). Since the first report of KSD in Japan in the 1970s, the disease has spread to many countries including England, France, the Netherlands, Germany, Italy, the Czech Republic, Austria, Poland, and India. In the spring of 2015, a koi farm in Hangzhou experienced an outbreak of an infectious disease. In this study we conducted clinical, histopathological, and molecular diagnostic analyses to determine the cause of the outbreak. CEV was found to be the causative agent of the disease. Moreover, PCR sequencing and phylogenetic analyses revealed that CEV was the extremely likely etiological agent of the mortality in this outbreak, which was further confirmed by sequence analysis of the PCR products obtained after nested PCR of the degenerate PCR. We also found that temperature is an important factor affecting carp morbidity and mortality in CEV infection. This is the first report of the detection of CEV in relation to KSD outbreaks in China. Our findings highlight the need for research on the CEV genome to better characterize the spread of CEV. © 2017 Elsevier B.V.

Solanum nigrum fruits have been conventionally used in beverages due to their nutritional substances such as minerals, vitamins, amino acids, proteins, sugars, polyphenols, and anthocyanins. The characterization of components and regulatory mechanism of anthocyanins in S. nigrum fruits have rarely been reported. In this study, we determined that the peel and flesh of S. nigrum fruits shared similar HPLC profiles but different contents and total antioxidant activities for anthocyanins. After an efficient purification method, mainly including extraction with pH 1.0 distilled water and then desorption with pH 1.0 95% ethanol after a DM-130 resin adsorption step to obtain more pure anthocyanin extracts, the purity of anthocyanins extracted from S. nigrum fruits reached 56.1%. Moreover, eight anthocyanins from S. nigrum fruit were identified with HPLC-MS/MS for the first time. A typical R2R3-MYB transcription factor gene, SnMYB, was also cloned for the first time by rapid amplification of cDNA ends (RACE)-PCR from S. nigrum. Moreover, the contents of anthocyanins were shown to correlate well (r = 0.93) with the expression levels of SnMYB gene during the fruit's developmental stages. Most significantly, SnMYB gene successfully produced high anthocyanin content (1.03 mg/g) when SnMYB gene was transiently expressed in tobacco leaves. Taken together, S. nigrum fruits are a promising resource for anthocyanin extraction, and SnMYB gene is an activator that positively regulates anthocyanin biosynthesis in S. nigrum.

An analytical method for the determination of glucose, fructose, isomaltose, isomaltotriose, maltose, panose and maltotriose was developed by high performance anion exchange chromatography coupled with integrated pulsed amperometric detection. The analysis was performed on a CarboPac™10 column with gradient elution of NaOH-NaOAc as mobile phases. The seven sugars were well separated in less than 26 min and showed good linear correlation coefficients between 0.5 and 50 mg/L. The detection limits (LODs) were 0.1 g/L. The recoveries ranged from 76.5% to 108.4% at different spiked levels. The relative standard deviations were between 3.02% and 8.23%. The established method was applied in determining the seven sugars in different batches of Shaoxing Jiafan rice wines from different manufacturers. The standard fingerprints were established by median values using the obtained results. Similarities of the fingerprints of the samples to the standard were calculated by using angle cosine. The results showed that the similarities of wines from different manufacturers were significantly different, so the method could be used for the identification of rice wines from different manufacturers.

Influence of different washing time on the fuzz and pilling performance of wool/polyester fabrics was studied in this paper. The fuzz and pilling performance of washed fabrics were tested and the number of pilling was counted and analyzed. The results showed that after the fabrics were washed with different time, the property of anti-pilling deteriorates to some extent with the increase of washing time. The blended ratio and density of warp and weft of wool/polyester fabrics were important factors affected the properties of anti - pilling. For pure wool fabrics, the number of pilling of the fabrics with small density of warp and weft increased with the increase of washing time. For wool/polyester fabrics, the number of pilling of with high wool content decreased with the increase of washing time.

A method was developed for the determination of eight pesticide residues (fipronil, imidacloprid, acetamiprid, buprofezin, triadimefon, triadimenol, profenofos, pyridaben) in tea by liquid chromatography-tandem mass spectrometry. The sample was extracted by accelerated solvent extraction with acetone-dichloromethane (1:1, v/v) as solvent, and the extract was then cleaned-up with a Carb/NH2 solid phase extraction (SPE) column. The separation was performed on a Hypersil Gold C18 column (150 mm ×2.1 mm, 5 μm) and with the gradient elution of acetonitrile and 0.1% formic acid. The eight pesticides were determined in the modes of electrospray ionization (ESI) and multiple reaction monitoring (MRM). The analytes were quantified by matrix-matched internal standard method for imidacloprid and acetamiprid, by matrix-matched external standard method for the other pesticides. The calibration curves showed good linearity in 1 -100 μg/L for fipronil, and in 5 -200 μg/L for the other pesticides. The limits of quantification (LOQs, S/N >10) were 2 μg/kg for fipronil and 10 μg/kg for the other pesticides. The average recoveries ranged from 75.5% to 115.0% with the relative standard deviations of 2.7% -7.7% at the spiked levels of 2, 5, 50 μg/kg for fipronil and 10, 50, 100 μg/kg for the other pesticides. The uncertainty evaluation for the results was carried out according to JJF 1059-1999 "Evaluation and Expression of Uncertainty in Measurement". Items constituting measurement uncertainty involved standard solution, weighing of sample, sample pre-treatment, and the measurement repeatability of the equipment were evaluated. The results showed that the measurement uncertainty is mainly due to sample pre-treatment, standard curves and measurement repeatability of the equipment. The method developed is suitable for the conformation and quantification of the pesticides in tea.

Fabric formability is not only one of the important factors of visual art, but also a research topic of fabric wearability. To further study the impact of fabric elements on formability, a new approach to the relationship between woven formability and yarn twist factor based on discriminant equation is proposed. Firstly we weave 10 kinds of worsted fabrics which are the same in thickness of yarn but different in twist factor of weft yarn, then test the basic mechanics index and calculate the discriminate value and suitable style. Secondly the relationship between fabric formability variation degree and yarn twist factor is obtained by curve fitting and fuzzy dynamic clustering. Lastly the relationship between fabric formability variation direction and yarn twist factor is got by the constant weights.

This paper presents an approach to identify apparel silhouettes. A feature region of the human face was first proposed for conducting face detection in fashion pictures with the Ada-Boost method, and the head was then located with its positional relation to the facial feature region. The linear relationship between the ratio of the body height to head length and the length of the lower body was ensured by restricting the RBH to a specific range. Under this condition, the apparelled body was divided into several parts, and the boundary of apparel on the lower body was determined considering the influence of the hemline. Based on the widths of the body parts and the apparel on the lower body, shape factors were established to express the extent to which the apparel silhouette approached a certain shape. A computer program was developed for implementation and demonstrated high accuracy in silhouette identification of an appareled body. © 2016, Institute of Biopolymers and Chemical Fibres.
