The Nisin Z fermentation supernatant with an antibiotic activity of about 1000IU/mL removed by centrifugal sedimentation was treated as waste water after Nisin Z being absorbed to the production strain cells in industrial production. A protocol to obtain high purity Nisin Z by a combination of macroporous adsorption resin chromatography, acetone-salt aqueous two-phase and medium pressure preparative liquid chromatography from fermentation supernatant was established. The purity of Nisin Z increased about 95 fold from 0.28% to 26.65% after enriching with HZM-3 resin and increased to 55.43% followed by aqueous two-phase decolorization. With separation of the preparative liquid chromatography, the purity of Nisin Z was 94.12%. It is very necessary to isolate capsaicin monomer for studying the relationship between capsaicin and other substances in chili oil. Resin HZ835 was screened from 5 types of macroporous resins due to good adsorption capacity and selectivity of capsaicin by chromatographic separation of chili oil. The optimum operation conditions were as follows: mobile phase was methanol: water (1.5:1, v:v), sample volume was 1.76% and flow rate was 1BV/h. Under these conditions, crystal capsaicin was obtained with the purity of 96.7%.
