A new HILIC-ELSD method was developed for compositional analysis of both xylo-oligosaccharides (XOS) with degree of polymerization (DP) from 2 to 8 and acetylated XOS with DP from 3 to 8. The method was carried out on a zwitterionic HILIC column using ELSD as a detector. The influences of mobile phase composition, column temperature and flow rate on the retention time and resolution of XOS were investigated. An excellent separation result was achieved with a linear gradient elution of 75%−50% acetonitrile in 30 min, at a flow rate of 1 mL/min and the column temperature at 35 °C. In addition, LC–ESI–MS was employed to determine the structural information of X7, X8 and acetylated XOS. The proposed method was simple, reliable, and no derivatization procedure was needed. It is suitable for compositional analysis and quality control of XOS. © 2017

In this study, Beagle dogs were administered xylooligosaccharide (XOS, CAS # 87099-0) at doses of 0, 1250, 2500, and 5000 mg/kg/day by oral gavage for 26 weeks. A 4-week recovery period was added to observe delayed or reversible toxicity. Measurements included body weight, food consumption, clinical observations, temperature, electrocardiogram (ECG), urinalysis, blood chemistry, hematology, organ weight, gross necropsy, and histopathological examination. Except for transient diarrhea or vomiting, no treatment-related adverse effects were noted. In the mid-dose groups, transitional diarrhea was observed in the initial 1–2 weeks. In the high-dose groups, diarrhea and/or vomiting were observed episodically over the duration of treatment. However, they disappeared after XOS was withdrawn in the recovery period. Although there was a tendency toward less weight gain in the high-dose group animal group, this is typical in animals and humans fed non-digestible carbohydrates. This chronic toxicity study demonstrated that the no observed adverse effect level (NOAEL) of XOS is 2500 mg/kg body weight (BW)/day. Based on body surface area (conversion factor of 0.54 for dogs to human), this corresponds to daily doses of 1350 mg/kg BW or 81–108 g XOS in human adults weighing 60–80 kg. © 2017

This study use Autohydrolysis explosion liquor of corncob as raw material inspected the process conditions that prepare soluble dietary fiber (SDF) by enzymatic methods and ultrasonic-assisted enzymatic methods. And the key performance of the SDF under different preparation conditions was measured. The optimal conditions of the two kinds preparation were determined by the response surface method and orthogonal experiment, in which enzymatic method: liquid ratio of 1, 27, cellulose 2.7 U/mL, 2.5 h, 55℃, alcohol precipitation time 6.4 h, the extraction rate of SDF under this condition was 18.11%. Ultrasonic-assisted enzymatic methods: cellulose 2.5 U/mL, ultrasonic frequency 50 kHz, ultrasonic time 60 min, ultrasonic power 20 W, the extraction rate of SDF was 24.12% (compared to pure enzymatic methods improved 33.2%). In addition, the results show that the SDF of ultrasonic-assisted enzymatic methods was significantly better than the SDF of pure enzymatic methods in water holding capacity, swelling capacity and soluble substances content. © 2016, Editorial Office of Journal of CIFST. Single factor preliminary and response surface methodology (Box-Behnken center combination examination) was applied to optimize conditions if soluble dietary fiber (SDF) extracted by enzymatic method from corncob. Results showed the optimal conditions were: liquid ratio of 1:20, cellulose 2.1 U/mL, xylanase 1.5 U/mL, 2.5 h, alcohol precipitation time 6 h, 55°C. Under these conditions, the yield of soluble dietary fiber rate was 21.10%. In this study, water holding capacity, swelling capacity and soluble substances content were also researched, which was 6.84 g/g, 7.47 mL/g, 1.21 g/100 mL water.

Xylo-oligosaccharide (XOS) is sugar oligomers composed of a β-1,4-linked xylopyranosyl backbone that are obtained by either chemical or, more commonly, enzymatic hydrolysis of xylan polysaccharides extracted from plant cell wall. In this study, acute and subchronic toxicity of XOS in mice and rats have been evaluated, respectively. In the acute study, no obvious clinical signs of toxicity or mortality were observed in mice at the dosage of 32g/kg BW XOS, excepting transient unformed stools were observed. In the subchronic study, XOS was evaluated in rats with dietary administration at concentrations of 0 (control), 0.9, 2.9, 8.8 and 10% for 13 weeks. Measurements included clinical observations, body weight, food consumption, food conversion efficiency, hematology, blood chemistry, gross necropsy, organ weight and histopathology. Under the conditions, no treatment-related changes were noted in behavior or appearance of the rats and no mortalities occurred. No toxicological findings were found in food consumption, food conversion efficiency, hematology, clinical biochemistry or organ weights in either sex. It is concluded, therefore, that the high dose level, at which the female and male rats consumed about 11.51 and 14.95g XOS/kg bw/d, respectively, is the no observed adverse effect level (NOAEL) of this 13-week toxicity study. © 2012 Informa Healthcare USA, Inc.

The culture conditions of a thermophilic fungus F1208 with high yield of xylanase activity were optimized by single-factor experiment and Box-Behnken design matrix of response surface methodology. The influences of medium composition and fermentation conditions in the cultivation were investigated. A high xylanase activity of 3 026.65 U/mL was obtained in 7 days, which was 3.15 fold of the initial enzyme activity (960.39 U/mL). The results indicated optimum conditions were corncob, mesh number 24~40,5%; soya peptone and yeast cream (ratio 1:3) 2%; glycerine 0.2%; pH 6.4, and the fermentation conditions was 60 mL, 48℃ and 200 r/min. ©, 2015, Chinese Institute of Food Science and Technology.
