Ex situ conservation efforts are the last resort for many critically endangered species, and captive breeding centers are thought to provide a safe environment for producing individuals for eventual re-introduction to the wild. The giant panda (Ailuropoda melanoleuca) is one of the world's most endangered animals and is a widely recognized symbol for conservation. Here, we report that captive pandas in China experience environmental and dietary exposures to high concentrations of persistent organic pollutants (polychlorinated dibenzo-p-dioxins, dibenzofurans, and biphenyls) and heavy metals (arsenic, cadmium, chromium, and lead). In the short term, those animals exhibiting elevated levels of such toxins should be relocated to breeding centers in less contaminated areas. Ensuring the long-term survival of both captive and wild pandas depends in part on reducing atmospheric emissions of toxic pollutants throughout China. © The Ecological Society of America

Various protocols for recombinant Interleukin-10 (IL-10) purification in wound healing have been reported previously. However, the therapeutic effect was not obvious. Thus, it is of great importance to find new and effective approaches for therapy. In this study, we propose that IL-10 and Arginine-Glycine-Aspartic (RGD) peptide would be a valuable therapeutic for wound healing. To explore a high-efficiency and cost-effective approach for the production of IL-10 and RGD peptide with bioactivity, a synthetic gene was cloned into a recombinant pTWIN1 vector. As a consequence, rhIL-10-RGD and the pH-induced self-cleavable Ssp DnaB mini-intein as a fusion protein was highly expressed by IPTG induction in Escherichia coli Rosetta without extra residues in a bioreactor. After Ni affinity chromatographic purification, rhIL-10-RGD was released by the Ssp DnaB intein-mediated self-cleavage that is triggered by pH shift. SDS-PAGE and silver staining showed a major band with an estimated molecular mass of 19.3kDa. Cell proliferation assay confirmed its potent proliferation activity on MC/9 murine mast cells. In conclusion, we report a novel strategy to produce rhIL-10-RGD mediated by the pH-induced self-cleavable Ssp DnaB mini-intein, and show that rhIL-10-RGD could play an effective role in wound healing of BALB/c mice.

In this paper, we researched the technology of the shrink-resist finishing of wool fabrics with the immobilized neutral proteinase, prcessed the single-actor experiment. The application technological parameter of the immobilized neutral proteinase was optimized by the orthogonal experimental design. The result showed that the amount of the immobilized neutral proteinase was 5% ( owf ) , the treating temperature was 50 °C , the treating pH value was 7. 0 and the treating time was 60 min. The results finishing the wool fabrics with the free and immobilized neutral proteinase were compared. The result showed that the shrinkage effect was increased 37. 6% , the weight loss percentage was descended 45. 2% , the strength loss percentage was descended 52. 7% , the whiteness was increased 23. 2% and the anti-pill rate was raised by level 2. This will establish the basis of the industrializing enzvme on shrink-resist finishing of wool knitted fabrics.

This study demonstrates that the type of nitrogen sources has an important influence on the growth of Streptococcus thermophilus.at the same time, viable counts was studied by in the medium containing various nitrogen sources (peptone, yeast extract, meat extract, tryptone, soya peptone and casein hydrolysate). The results indicated that soya peptone was the most efficient nitrogen source and and the influence of different concentrations of soya peptone on growth was determined. The results indicated that viable bacteria were stimulated by the high soya peptone concentration (30g/L). © (2012) Trans Tech Publications, Switzerland.

Effect of the extract of Fructus tribuli (EFT) on growth of Lactobacillus acidophilus LA04, LA05, LA06 and LA09 was studied by measuring optical density at 600nm (OD 600) and pH using MRS culture media as the control. The addition of EFT (v/v) was 1%, 2%, 3%, 4% and 5%. Results were as follows: addition of EFT could promote the growth of Lactobacillus acidophilus LA09, The optimum concentration of EFT in MRS media was 1% at incubation 24h, but addition of EFT on growth of Lactobacillus acidophilus LA04, LA05 and LA06 has no significant effect. © (2011) Trans Tech Publications, Switzerland.

In the present study, the experiments were investigated with the effecting of NaHCO 3, KH 2PO 4, K 2HPO 4 and Sodium ascorbate on survival of Lactobacillus acidophilus during freeze-drying. Designing different concentrations of protective agents, respectively. The survival ratio of Lactobacillus acidophilus and the number of viable cells will be measured after completing drying. In the process, there were major differences between the different concentrations of protective agents in survival during freeze-drying. As the result, the protective agent was playing a key factor affecting the survival of Lactobacillus acidophilus. At present in the study of the experiments, the highest survival rate was NaHCO 3 as the protective agent, and when the protective agent as K 2HPO 4 the number of viable cells was largest. © (2012) Trans Tech Publications, Switzerland.

Four different prebiotics including fructo-oligosaccharide isomalto-oligosaccharide, inulin and xylo-oligosaccharide were screened for their effects on survival of Bifidobacterium bifidum during freeze-drying. The additions of prebiotics (v/v) were all 4%, 8%, 12%, 16% and 20%. The results showed that prebiotics could act on Bifidobacterium Bifidum. The optimum concentration of fructo-oligosaccharides (FOS) in samples for surival of Bifidobacterium bifidum was 12% and for viable count was 20%. Addition of xylo-oligosaccharides (XOS) played a significant role in promoting cell viablity when it was in low level. The optimum concentration of isomalto- oligosaccharides (IMO) for surival of Bifidobacterium bifidum was 16% and for viable count was 12%, while inulin in samples for surival and viable cell of Bifidobacterium bifidum both was 4%.

In the present study, the experiments were investigated with effecting of sucrose, lactose, glucose, maltose on survival of Lactobacillus acidophilus during freeze-drying. In order to reach sufficient numbers viable cells, so different concentrations of protective agents were designed in the experiment, which as 3%, 6%, 9%, 12% and 15%, respectively. Measuring the survival ratio of Lactobacillus acidophilus and number of viable cells after completed drying. There were significant differences in survival during freeze-drying. Results showed that the protective agent was a critical factor affecting the survival of Lactobacillus acidophilus. In the experiments, the highest survival rate was soluble starch as the protective agent, and when the protective agent as galactose the number of viable cells was largest.

Lactobacillus acidophilus not only improves the intestinal flora balance but also inhabits the growth of undesirable microorganisms in intestine, which is benefit to the health of humans and animals. Plackett-Burman and steepest ascent experiment are the rapid and concise ways of screening the main effective factors. This study is aimed to select the main influence factors and optimize the medium for Lactobacillus acidophilus by Plackett-Burman experiment and steepest ascent experiment.The ideal carbon source was screened among glucose, maltose, lactose and whey powder, and the ideal nitrogen source was screened among casein hydrolysate, peptone, yeast extract powder, fish meal, carbamide, ammonium sulfate and sodium nitrate by single factor experiment. Plackett-Burman and steepest ascent experiment were applied to screen the main effective factors of Lactobacillus acidophilus among peptone, beef extract, yeast extract powder, glucose, K2HPO4, C6H14O7N2, CH3COONa, MgSO4 and Tween-80. Result. The results indicated that glucose (p = 0.01510) as negative factor and K2HPO4 (p = 0.02017) as positive effect were the significant growth factors of Lactobacillus acidophilus, CH3COONa (p = 0.09273) as positive effect was an important factor, and the optimized medium was as follows: glucose - 21 g/L, K2HPO4 - 3.5 g/L, CH3COONa - 6.5 g/L, peptone - 10 g/L, beef extract - 8 g/L, yeast extract pow.nd. Thus, Plackett-Burman and steepest ascent experiment improve the veracity of optimization the medium for Lactobacillus acidophilus compared with the previous research.
