Objectives: To explore the general physiological and molecular changes occurring as a result of acute hypothyroidism. Materials and Methods: Hypothyroidism was developed by thyroidectomy in wistar rats. After surgery, animals were observed for 14 days in order to determine changes in body weight, feed consumption, rectal temperature, heart rate, and blood pressure, clinical pathological and hormonal alteration. In addition, relative changes in weight, histopathology and MHC-α and β gene expression of heart was also evaluated. Results: Thyroidectomised rats showed lethargy, piloerection and decreased locomotors activity. Day dependent significantly decreased body weight and feed consumption were seen in hypothyroid rats. Rectal temperature was significantly reduced at day 7 and 14 after surgery. Heart rate and blood pressure were significantly decreased at day 14 in thyroidectomized rats in comparison with euthyroid rats. Haematological parameters shown high WBC count. Serum LDL and phosphorous levels were high where as triglycerides; total protein, creatinine kinase and globulin were low. Heart weight was significantly high. Histopathology of heart tissue showed myocardial segmental degeneration. Downregulation of MHC-α and upregulation of MHC-β were seen in hypothyroid rats in comparison with euthyroid rats. Conclusion: This finding suggests that deficiency of thyroid hormone (TH) in hypothyroidism is associated to a cardiac dysfunction and acute changes in body homoeostasis as result of sudden arrest of thyroid hormone.

Background: Preparation of highly standardized herbal products with respect to chemical composition and biological activity is considered to be a valuable approach in this field. SJT-DI-02 polyherbal formulation was successfully developed at our institute and filed for patent at Mumbai patent office. Objective: The present work was marker based standardization of patented, novel and efficacious polyherbal formulation namely SJT-DI-02 for the treatment of diabetes. The SJT-DI-02 was comprised of dried extracts of rhizomes of Acorus calamus, leaves of Aegle marmelose, fruits of Benincasa hispida, roots of Chlorophytum arendinaceum, seeds of Eugenia jambolana, leaves of Ocimum sanctum, pericarp of Punica granatum, seeds of Tamarindus indica. Selected plants were collected, dried and extracted with suitable solvents. The formulation was prepared by mixing different fractions of extracts. Materials and Methods: For successful and best standardization, first of all selection and procurement was carried out. Selection is done on the basis of therapeutic efficacy and amount of the marker present in the particular plant part. At the time of procurement side by side phytochemical screening and estimation of phytoconstituents was carried out. After completion of preliminary screening using characterized markers, we tried to develop best TLC systems using selected solvent composition. Finally well-developed TLC systems were applied in HPTLC. In the present study polyherbal formulation was standardized by using different four markers. TLC Densitometric methods were developed using HPTLC for the quantification of these marker compounds. Solvent systems were optimized to achieve best resolution of the marker compounds from other components of the sample extract. The identity of the bands in the sample extracts were confirmed by comparing the Rf and the absorption spectra by overlaying their UV absorption spectra with those of their respective standards. The purity of the bands due to marker compounds in the sample extracts were confirmed by overlaying the absorption spectra recorded at start, middle and end position of the band in the sample tracks. After conforming all these things fingerprints were developed for all three formulations which will be act as authentification and quality control tool. Results: % w/w of asarones is 3.61, % w/w of marmelosin is 4.60, % w/w of gallic acid is 10.80 and % w/w of lupeol is 4.13.The method was validated in terms of linearity, precision, repeatability, limit of detection, limit of quantification and accuracy. In well-developed mobile phase system linearity was found to be in the range of 0.983-0.995, % recovery was found to be in the range of 97.48-99.63, % RSD for intraday and interday was found to be 0.13- 0.70 and 0.32 -1.41 and LOD and LOQ was found to be in the range of 0.15- 0.61 and 0.45 -1.83 microgram per ml. Conclusion: Thus High performance thin layer chromatography (HPTLC) methods were developed and validated in terms of linearity, precision, repeatability, limit of detection, limit of quantification and accuracy. The methods were rapid, sensitive, reproducible and economical. It does not suffer any positive or negative interference due to common other component present in the formulation and would also serve as a tool for authentication of herbal products containing marmelosin, gallic acid, lupeol and asarones. Thus this work provides standardized and therapeutically active polyherbal formulations for the different ailments.

MicroRNAs (miRNAs) are a novel group of universally present small noncoding endogenous RNAs that regulate gene expression and protein coding by base pairing with the 3 untranslated region (UTR) of target mRNAs. So they have been associated with several physiological processes and play an important role in the manifestation of diverse diseases. miRNAs expression is associated with the normal and diverse pathophysiological state including cardiac hypertrophy, neurodegenerative diseases, diabetes and its complication, and cancer because individual miRNAs are associated with the regulation of the expression of multiple target genes. Modulating the expression of a single miRNA can influence an entire gene network and thereby modify complex disease phenotypes. From recent studies, it has been confirmed that miRNA has a potential physiological role in various body systems. But in some specialized condition over expression of miRNA within the cytoplasm also leads to some pathological condition in the body. Here, we summarize the roles of miRNAs in various pathological conditions and consider the advantages and potential challenges of miRNA-based therapeutic approaches compared to conventional drug-based therapies.

Q-Ratio method for simultaneous estimation of Ebastine and Montelukast Sodium in combined tablet dosage form have been developed. The UV spectrophotometric method is the Q - analysis (absorption ratio) method, which involves the formation of absorbance equation at 261.34 nm (isoabsorptive point) and at 253.00 nm the maximum absorption of Ebastine. The linearity ranges for Ebastine and Montelukast sodium were 5-45 μg/ml and 5-45 μg/ml respectively. The accuracy of the method was assessed by recovery studies was found to be and 100.52 ± 0.1893 and 100.13 ± 0.1883 for Q analysis (absorption ratio) method for Ebastine and Montelukast Sodium respectively. These methods are simple, accurate and rapid; those require no preliminary separation and can therefore be used for routine analysis of both drugs in quality control laboratories.

Cancer is multipathological disease that is being considered immortal in today's global medicine world. Therefore, the present study was aimed to develop and evaluate polyherbal formulation for its anticancer activity. Four polyherbal formulations namely SJT ONC (1-4) were prepared by mixing of different proportion of extracts of stem bark of Tecomella undulata, Bauhinia variegata, Oroxylum indicum and leaves of Indigofera tinctoria. All the four formulations were evaluated for their cytotoxic effect using MTT (3-(4,5-dimethylthiazolyl-2)-2, 5-diphenyltetrazolium bromide) assay on cell lines of human colon adenocarcinoma (Caco-2) and human breast adenocarcinoma (MCF-7). Amongst all four polyherbal formulations, SJT ONC-1 (1000 μg mL -1) showed significant cytotoxicity (pO.OOl) on the Caco-2 (63.82%) and MCF-7 (74.18%) cell lines as compared to control. Further, SJT ONC-1 was evaluated for its apoptotic activity using cell death detection ELISA PLUS assay and it showed significant increase (p<0.01) in DNA fragmentation of Caco-2 (14.83 fold) and MCF-7 (3.22 fold) compared to control group. Moreover the antitumor activity of SJT ONC-1 (300 mg kg -1, p.o.) was evaluated against Dimethylbenz (a) anthracene induced mammary tumors in rats fed with high fat diet. The result suggested the ST ONC-1 significantly decrease the tumor volume and the effect was comparable to the standard drug 5-fluro uracil. Thus, the SJT ONC-1 could render prospective candidate for the therapy of cancer. © 2012 Academic Journals Inc.
