The paraffin-degrading strain, isolated from waxy oil production wells in Daqing oilfield, were obtained by separation and purification experiment. The paraffin-degrading strain P1 was identified through 16S rDNA sequence analysis, which exhibited the highest similarities to Bacillus anthracis strain XFB-BN. The biosurfactant-producing strain was obtained by laboratory saved. After paraffin-degrading and biosurfactant-producing strain treatment, the paraffin degradation rate could reach to 68.2 %, the paraffin prevention rate could reach to 80.7 %, the reduction rate of crude oil viscosity was 62.4 %. By analyzing the data, the paraffin-degrading and biosurfactant-producing strain had better effect on microbial prevention of wax deposition. Source

The aim of the study was to increase the production of biosurfactant by using the inexpensive medium materials and the optimized fermentation conditions. Five biosurfactant producers were isolated from oilfield wastewater. The most efficient strain S2 was identified through 16S rDNA sequence analysis, which exhibitted the highest similarities to Bacillus subtilis strains CCTCC M201162. The optimal medium composition of this strain were beer wastewater 40 g/L, diammonium phosphate (NH4H2PO4) 4 g/L, MgSO4 0.02 g/L, NaCl 5 g/L. The emulsification index (E24) was optimized using a box-behnken experimental design and response surface methodology(RSM). The RSM revealed that when pH was 7.2, temperature was 42.1°C, inoculum concentration was 5.2% (v/v) and rotate speed was 163 r/min, the optimal condition was obtained. Under this condition, the optimal E24 of the biosurfactant produced by Bacillus subtilis strains CCTCC M201162 was 81.20%, the production of crude biosurfactant increased from 0.72 g/L to 1.26 g/L and the surface tension of fermentation broth reduced from 72.0 mN/m to 22.8 mN/m. A quadratic response model was constructed through RSM designs, leading to a 75.0% increase of biosurfactant production by Bacillus subtilis strains CCTCC M201162. © 2014 American Institute of Chemical Engineers Environ Prog. In the light of test wells with pump, the integrated technology of perforating, oil production and testing is taken, which can reduce working procedure, avoid pollution of drilling fluid to the formation effectively, save the operation cost, improve the work efficiency of the oil well. It is vital for the study of fracturing parameters optimal design and fluids rate forecast to calculate fracture propagation parameters accurately. In order to calculate fracture propagation accurately, in this paper, the model for calculating the value of energy release when the rock body collapses was established basing on catastrophe theory, considering that the collapse of fracturing rock body was caused by the accumulation of rock body deformation energy and releasing suddenly. And making the assumption that the energy that had released can be used for the propagation and extension of the cracks, the new model for calculating the parameters of cracks were established. The characteristics of sandstone rock mass were selected to study and analyze the fracturing parameters through laboratory experiment in daqing oilfield, the results show the maximum error between theoretical and the actual result is 9.71%. Field practices have shown that hydraulic particles as plugging agent, satisfy the need for permanent plugging of formation. Ultra-fine particle cement plays an important role in sealing and plugging formations of medium permeability. To seal and plug formations of low permeability and satisfy other special needs, a nano-meter hydraulic particle plugging agent was developed. This plugging agent has particle sizes of less than 1μm, with average particle size of less than 400 nm, and ratio of particles finer than 100 nm exceeding 21%. It is made by a high energy nano impact mill. The plugging agent is easy to disperse in water. Slurries treated with this plugging agent expand after solidification without the need for expansion agent, and have adjustable thickening time. The plugging agent has satisfactory compressive strength and low air permeability. It has been successfully used in an exploration well of five-interval in the 3rd structure, Nanpu, Jidong Oilfield.
