A chemical investigation of marine sponge Jaspis stellifera, collected from South China Sea, led to the isolation of two new compounds, Jaspiferin A and B (1-2), and six known compounds, gibepyrone F (3), p- hydroxy benzaldehyde (4), 3-Indole-3-aldehyde (5), Thymine (6), 24(28)-dehydroaplysterol (7), (25s)-26- methylene-cholest-4-en-3-one (8). Their structures were determined by extensive spectroscopic analysis in association with physical and chemical properties, as well as comparison of their spectral data with these reported in literatures. The biogenetic transformation of compound 2 was also speculated. © 2012 Reproduction is free for scientific studies.

The aim of the current study was to investigate the importance of strain elastography (SE) in the differential diagnosis of benign and malignant soft tissue masses. SE was adopted to examine 61 patients with superficial masses, classify their elastic scores and assess their strain ratios (SRs) between the masses and the surrounding structures. Significantly increased SR values and elastic scores were observed in the malignant masses compared with the benign masses (5.42±3.47 vs. 1.80±2.10, P<0.001; 3.13±0.34 vs. 2.03±0.99, P<0.001). Area under receiver operating characteristic curve values of the SRs and elastic scores were 0.87 (P<0.001) and 0.805 (P=0.001), respectively. With an SR of >2.295 as the optimal threshold value, the sensitivity, specificity and positive and negative predictive values for diagnosing a malignant mass were 93.8, 80.5, 65.2 and 97.1%, respectively; whilst using an elastic score of ≥3 as the optimal threshold value, the sensitivity, specificity and positive and negative predictive values for diagnosing a malignant mass were 100, 51.6, 51.6 and 100%, respectively. SR values and elastic scores were significantly different between the malignant and benign soft tissue masses. Therefore, SE may be used to effectively differentiate between malignant and benign soft tissue masses. © 2017, Spandidos Publications. All rights reserved.

Background: Isoliquiritigenin (ISL) has various biological activities including as antioxidant and an inhibitor of PI3K/AKT signaling pathway. However, both oxidative stress and activated PI3K/AKT signaling contribute to the aberrant proliferation of vascular smooth muscle cells (VSMCs). This study is aimed to explore the effect of ISL on the proliferation of human arterial smooth muscle cells (HASMCs) and to investigate the underlying mechanisms. Methods: BrdU incorporation, cell cycle and reactive oxygen species (ROS) in normal or ISL treated HASMCs were analyzed by flow cytometry. Cell viablity was measured by CCK-8. Protein expression levels were examined by Western blot, and superoxide dismutase (SOD) activity was detected by using commercial kit. Results: We observed that ISL could inhibit the proliferation of HASMCs in a dose and time dependent manner. Cell cycle of ISL treated HASMCs arrested mainly in G1/S phase and accompanied with elevated expression of p27 and decreased expression of CyclinD1 and CyclinE. In addition, ISL could down-regulated the expression of p-PI3K and p-AKT, alleviated oxidative stress and enhanced the SOD activity in HASMCs. Furthermore, H2O2 treatment partly improved cell viability and up regulated p-PI3K and p-AKT in HASMCs. Conclusions: Therefore, we concluded that ISL inhibited the proliferation of HASMCs via attenuating oxidative stress and suppressing PI3K/AKT signaling pathway. The inhibitory effect of ISL on PI3K/AKT signaling pathway, at least partly, was mediated by ROS. © 2017 The Author(s).

Background:Maternal hypoxia induces sustained fetal adaptations associated with changes in gene expression. We hypothesized that intermittent maternal hypoxia has an influence on regional expression of endothelial nitric oxide synthase (eNOS) in fetal arteries of New Zealand White rabbits.Methods:Timed- pregnant New Zealand White rabbits (term = 30 ± 1 d) were randomly assigned to a normoxic control group (n = 5) or a hypoxia group (12% O 2, n = 5) during days 10-29 of pregnancy. At the end of pregnancy (29 d gestation), blood samples were collected from mothers and fetuses. Carotid and femoral arteries of fetuses were extracted for eNOS mRNA and protein concentration and analysis of total NOS activities.Results:Our data demonstrate that chronic intermittent maternal hypoxia significantly increased eNOS mRNA and protein concentrations and total NOS activities in carotid artery segments but decreased eNOS mRNA and protein concentrations and total NOS activities in femoral artery segments in the same fetuses. Vascular endothelial cells, but not smooth muscle cells, of fetal rabbits exhibited positive immunostaining for the eNOS protein.Conclusion:These observations suggest that chronic hypoxia can regulate regional expression of eNOS as an adaptive response to hypoxic stress in fetal arteries. with the wide application of biometric identification technology, the security defects are gradually exposed. The encryption technology which was combined with Biometric cryptography and biometric identification technology is to compensate for lack of biometrics in terms of security caused. Based on the existing research of face biometrics technology, the face biometric encryption algorithm with security and fault tolerance was proposed: fuzzy random cyclic mapping. In each cycle, the use of encryption model is the key of the previous cycle as a random seed to generate a mapping matrix. The facial features of the user was mapped to formate a cyclic random mapping process. In the process of encryption, fault tolerant technology was used to reduce the effect of random noise and the characteristics of the legitimate users face image for recognition rate. The experiment showed that the biological characteristics of face encryption algorithm was in high security based on fuzzy random cyclic mapping.

Gene expression of phenylalanine ammonia-lyase (PAL) in allelopathic rice PI312777 was inhibited by RNA interference (RNAi). Transgenic rice showed lower levels of PAL gene expression and PAL activity than wild type rice (WT). The concentrations of phenolic compounds were lower in the root tissues and root exudates of transgenic rice than in those of wild type plants. When barndyardgrass (BYG) was used as the receiver plant, the allelopathic potential of transgenic rice was reduced. The sizes of the bacterial and fungal populations in rice rhizospheric soil at the 3-, 5-, and 7-leaf stages were estimated by using quantitative PCR (qPCR), which showed a decrease in both populations at all stages of leaf development analyzed. However, PI312777 had a larger microbial population than transgenic rice. In addition, in T-RFLP studies, 14 different groups of bacteria were detected in WT and only 6 were detected in transgenic rice. This indicates that there was less rhizospheric bacterial diversity associated with transgenic rice than with WT. These findings collectively suggest that PAL functions as a positive regulator of rice allelopathic potential. © 2013 Springer Science+Business Media New York.

Hyperproduction of mucin in the nasal epithelium is an important feature of nasal inflammatory diseases. We investigated the mechanism of lipopolysaccharides (LPS) involvement in mucin 5 subtype AC (MUC5AC) expression in human nasal epithelial cells. The primary human nasal epithelial cells were cultured in vitro, which were treated with LPS (10 nM/ml or 1 μM/ml) for 12 and 24 h. LPS-induced MUC5AC protein was determined in nasal epithelial cells. The levels of nuclear factor kappa B p65 (NF-κBp65) and its inhibitor kappa Bα (IκBα) protein were also detected, and interleukin-1β (IL-1β) mRNA was detected by real-time PCR. LPS up-regulated MUC5AC protein in human nasal epithelial cells, and we determined that the up-regulation of MUC5AC expression was due to a time- and dose-dependent degradation of IκBα protein, which resulted in the increase of NF-κBp65 nuclear translocation. Subsequently, we also determined that LPS can induce IL-1β mRNA in a time- and dose-dependent manner. These data show that LPS treatment activated NF-κB by promoting the degradation of IκBα and the nuclear localization of NF-κBp65, which induced MUC5AC overproduction. © 2012 Springer-Verlag.

The synthesis and in vitro photodynamic anticancer activity of a new photosensitizer, tetra(trifluoroethoxy) germanium phthalocyanine (GePcF), were studied. GePcF was characterized by UV-Vis, IR, MS and elemental analysis. The in vitro photodynamic activity of GePcF was studied by MTT. IC50 of GePcF for SW480 cells of human colonic adenocarcinoma and HeLa cells of cervical cancer were 36.53 and 45.78 μmol/L, respectively. GePcF as a photosensitizer may be used to treat cancers due to its photodynamic anticancer activity.

To study the expression of P57(kip2) and Maspin in the pathological scar and their possible role in the pathogenesis of abnormal scars. Immunohistochemistry integrated image analysis and reverse transcription polymerase chain reaction (RT-RCR) were performed to detect the expression of P57(kip2) and Maspin in hypertrophic scar, keloid, mature scar and normal skin. Statistics was used to analyze the datas. The expression of P57(kip2) protein was fixed to fibroblast intranuclear in abnormal scar, and the expression of P57(kip2) protein and P57(kip2) mRNA decreased (P < 0.05). The expression of Maspin protein was fixed to fibroblast cytoplasm and intranuclear in abnormal scar, and the expression of Maspin protein and Maspin mRNA decrease, compared with that in normal group (P < 0.05). There was positive correlation between P57(kip2) protein and Maspin protein expression (P < 0.01). The decreased expression of P57(kip2) and Maspin in abnormal scar shows that they are cicatrix-related genes. There is a positive relationship between the two genes. It may be one of the mechanisms of pathogenesis of abnormal scar. It makes effect through fibroblasts.
