To accurately evaluate the comprehensive trait of new flue-cured tobacco varieties (lines), the data of 12 trait indexes (including natural leaf number, yield, percentage of high grade leaf, grade index, nicotine content, reducing sugar content, potassium content, bacterial wilt disease index, climate spot disease index, appearance quality score, stem content and total score of sensory evaluation) obtained in 2012 Guizhou regional test were analyzed with fuzzy comprehensive evaluation method. GGE biplot analysis was used to analyze comprehensive evaluation results, conduct varieties (lines) evaluation and testing points evaluation, and analyze the adaptable areas of each variety (line). The results showed that Guiyan 2, 2010A4 and MT-2 obtained better and stabler comprehensive evaluation value; YQ-1 obtained better comprehensive evaluation value in District 1 (Longgang and Weining), as did Guiyan 2 in District 2 (Zunyi, Guiding and Kaili) and 6517 in District 3 (Yinjiang). According to evaluation of testing points, those in Guiding and Zunyi were good in representativeness and discrimination; those in Yinjiang, Weining and Longgang were good in discrimination, but not as good in representativeness; those in Kaili were good in representativeness, but not as good in discrimanation. To reveal the rules of distribution and molecular variation of strains of Potato virus Y (PVY) in Qiannan tobacco belt of Guizhou, PVY strains were initially identified by serological method; further, 19 selected PVY isolates were subjected to molecular identification. The results showed that all the cloned fragments of the 19 PVY cDNA were 1074 nts in full-length, containing a complete cp gene of 801 nts and encoding 266 amino acids. The results of sequence comparison showed that the similarity of nucleotide sequence of cp genes in the 19 PVY isolates was 87.0%-99.9%, and the similarity with the cp genes in the 17 PVY strains logged in GenBank was 86.6%-99.9%. Phylogenetic tree analysis indicated that 3 of the 19 PVY isolates from Qiannan had closer relationship with PVYN and PVYNTN, and the other 16 PVY isolates were more closely related to PVYN:O. Source
