Background: The purpose of this study was to evaluate the effects of prehypertensive losartan and amlodipine administration on left ventricular (LV) remodeling and function in spontaneously hypertensive rats-stroke prone (SHRSP). Methods: Spontaneously hypertensive rats-stroke prone were prehypertensively administered losartan, amlodipine, or vehicle. Wistar-Kyoto ratswere used as a control. Blood pressure of the ratswas determined by tail-cuffmethod, and LV structure and function were measured by echocardiography and LV cannulation. Collagen volume fraction was analyzed by picrosirius red staining. Protein expressions of brain natriuretic peptide (BNP) and angiotensin II type 1 (AT1R) and type 2 (AT2R) receptors were determined by use of the Western blotting method. Results: Although both drugs downregulated BNP protein expression, the LV remodeling and function were more improved with losartan than with amlodipine treatment. Losartan upregulated AT1R and downregulated AT2R protein expression. Conclusions: Both drugs inhibited LV remodeling and improved LV function in prehypertensively treated SHRSP. Losartan provided better continued heart protection, potentially due to its persistent inhibition of AT1R and activation of AT2R in the myocardium. Source

Prader-Willi syndrome (PWS) is a genetic disease characterized by persistent hunger and hyperphagia. The lack of the Snord116 small nucleolar RNA cluster has been identified as the major contributor to PWS symptoms. The Snord116 deletion (Snord116del) mouse model manifested a subset of PWS symptoms including hyperphagia and hyperghrelinemia. In this study, male Snord116del mice were characterized and tested for their acute and chronic responses to anorexic substances related to the ghrelin pathway. In comparison with their wild-type littermates, the food intake rate of Snord116del mice was 14% higher when fed ad libitum, and 32% to 49% higher within 12 hours after fasting. Fasted Snord116del mice were less sensitive to the acute anorexic effect of competitive antagonist [D-Lys3]-GHRP6, YIL-781, and reverse agonist [D-Arg 1,D-Phe5,D-Trp7,9,Leu11]-substance P (SPA) of ghrelin receptor GHS-R. All 3 GHS-R inhibitors failed to inhibit chronic food intake of either Snord116del or wild-type mice due to rapid adaptation. Although fasted Snord116del mice had normal sensitivity to the acute anorexic effect of glucagon-like peptide 1 receptor agonist exenatide, those fed ad libitum required a higher dose and more frequent delivery to achieve ∼15% suppression of long-term food intake in comparison with wild-type mice. Ghrelin, however, is unlikely to be essential for the anorexic effect of exenatide in fed mice, as shown by the fact that exenatide did not reduce ghrelin levels in fed mice and food intake of ghrelin-/- mice fed ad libitum could be suppressed by exenatide. In conclusion, this study suggests that GHS-R may not be an effective therapeutic target, and in contrast, exenatide may produce anorexic effect in PWS individuals. Six pieces of concrete-filled cold-formed square steel tube beans subjected to law-cycle revelled loading was tested, and nonlinear finiete element anaysis was also conducted to explore the effects of span to height ratios and with to thiekness ratbs of cross-sectbn on the hysteretic behavior ductility and ability of energy disspation Sane important concbsions can be drawn from the experiments and the nonlinear analyst and listed as follows (1) The con crete-filled cold-fomed square steel tube beams exhbit stable hysteresis behavbr up to local buckling and then shaw considerable good ductility and good ability of energy disspatbn The piich-crinkle phenomena do not occur in the hysteresis loops of load-displaccment (2) The hysteresis behavior and ductility of concrete-filled cold-fomed square steel tube beans depend on fie span to height ratios and with to thickness ratios (3) The proposed nonlhear FE model could accurately predict the overall seismic behavior and the inelastic perfoimance of the concrete-filled cold-fomed square steel tube beams The hysteresis loops from nonlinear FE models under law-cycle reversed bading agree well with those from the law-cyclr reversed loading tests The proposed nonlinear FE model could also be used to conduct the nonlinear parameter analysis of the cold-formed canposite beams.
