Molecular beacons carrying JOE dye (4,5-dichloro-2,7-dimethoxy-6-carboxyfluorescein) on a rigid or flexible linker and one or two BHQ1 quenchers have been prepared and tested in real-time PCR using Fusarium avenaceum elongation factor 1 DNA template. The probes were different in their structures (loop size and stem length), linkers for dye attachment (6-aminohexanol or trans-4-aminocyclohexanol), quencher composition (single and double BHQ1) to elucidate the influence of all these features. Fluorogenic properties of the probes were studied and compared to those of FAM (fluorescein)-based probes. All the factors - stem length, JOE vs FAM, rigid vs flexible linker, single vs double quencher - appeared to play a considerable role in the probes fluorescent properties and determine the usability of the probe at two different temperatures of fluorescence detection (55 and 64).

Successful drug delivery using implantable pumps may be found in over 12,500 published articles. Their versatility in delivering continuous infusion, intermittent or complex infusion protocols acutely or chronically has made them ubiquitous in drug discovery and basic research. The recent availability of iPRECIO®, a programmable, refillable, and implantable infusion pump has made it possible to carry out quantitative pharmacology (PKPD) in single animals. When combined with specialized catheters, specific administration sites have been selected. When combined with radiotelemetry, the physiologic gold standard, more sensitive and powerful means of detecting drug induced therapeutic, and/or adverse effects has been possible. Numerous application examples are cited from iPRECIO® use in Japan, United States, and Europe with iPRECIO® as an enabling drug delivery device where the refillable and programmability functionality were key benefits. The ability to start/stop drug delivery and to have control periods prior dosing made it possible to have equivalent effects at a much lower dose than previously studied. Five different iPRECIO® applications are described in detail with references to the original work where the implantable, refillable, and programmable benefits are demonstrated with their different end-points. © 2011 Tan, Watts and Davis.

There are cases where a fluid transport apparatus cannot accurately discharge a set discharge amount of fluid, due to individual differences in components of the fluid transport apparatus, such as the diameter of an opening of an elastic tube. Therefore, in a fluid transport apparatus including a fluid transport cartridge and a fluid transport driving unit that are separable from each other, the fluid transport driving unit comprises a driving section that causes a drive mechanism to operate to transport fluid filling a transport path of an attached fluid transport cartridge to a discharge opening of the transport path; a receiving section that receives a characteristic value measured in advance for each of a plurality of the fluid transport cartridges; and a control section that controls the driving section based on the characteristic values received by the receiving sections.

In a fluid transport apparatus, there are cases where a set fluid discharge amount cannot be accurately discharged, due to wear and tear on an elastic tube. Therefore, in a fluid transport apparatus including a fluid transport cartridge and a fluid transport driving unit that are separable from each other, the fluid transport cartridge comprises an elastic tube that transports fluid, a pressing portion that moves between a first position in which a transport path of the fluid is constricted by pressing on the elastic tube from outside and a second position in which the transport path of the fluid is not constricted, and a housing portion that houses a drive mechanism for moving the pressing portion.

The connecting section is provided at a linear portion of the chassis, which has an ovular shape when seen from a top surface direction of the reservoir. The connecting section is provided within a concave portion formed in the linear portion. An internal elastic tube that delivers the fluid flowing therethrough and is pressed out by the fluid delivery mechanism, from the reservoir to the connector, but is not pressed until the fluid delivery device is used by being embedded, is arranged along an inner circumference of the ovular shape. The connecting section and the fluid injection port that receives the fluid to be replenished in the reservoir are arranged in a line along a replenishing direction of the fluid in the chassis.
