Background: The study of the effect of different dialysis methods on cellular immune function of maintenance haemodialysis (MHD) patients should provide theoretical support for deciding on the best method of blood purification that effectively improves cellular immune function of haemodialysis patients. Subjects and Method: Sixty MHD patients were randomly divided into three groups that respectively received treatment of haemodialysis (HD), high flux haemodialysis (HFHD) and haemodiafiltration (HDF). Peripheral blood T lymphocyte subsets [CD4+, CD8+, CD25+ (mIL-2R) and CD4+/CD8+ ratio] and serum interleukin (IL)-2 and soluble IL-2 receptor (sIL-2R) levels were detected before dialysis and 4, 24 and 48 hours after dialysis in all cases. Results: Compared with the HD group, CD4+ and CD25+ cells, CD4+/CD8+ ratio and IL-2 level increased but sIL-2R level decreased in the HFHD and HDF groups at four hours without statistical significance (p > 0.05) and at 24 and 48 hours after dialysis with statistical significance (p < 0.05), while CD8+ cells had no change after dialysis (p > 0.05). Compared with the HFHD group, CD4+ and CD25+ cells, CD4+/CD8+ ratio, and IL-2 level increased but sIL-2R level decreased in the HDF group at four and 24 hours without statistical significance (p > 0.05) and at 48 hours after dialysis with statistical significance (p < 0.05), while CD8+ cells had no change after dialysis (p > 0.05). Conclusion: The results indicate that HD can briefly improve the cellular immune function of MHD patients, while HDF and HFHD can improve it continuously, with HDF having the best effect.

Purpose: Diabetic nephropathy (DN) is one of the most common diabetic microvascular complications. Inflammatory factors participate in each stage of DN, and nuclear factor (NF)-κB and monocyte chemoattractant protein-1(MCP-1) play important mediation roles. The purpose of this study was to investigate the renal protective effects of a diet and exercise intervention in a rat model of Type 2 diabetes mellitus (T2DM). Method: Control rats (Group A, n = 10) were fed a normal diet, while 30 rats were fed a high-glucose, high-fat diet and given an intraperitoneal injection of streptozocin to establish the T2DM model. Model rats (n = 8 per group) were randomized into Groups B, C, and D. Groups C and D were treated with glibenclamide, and Group D received an 8-week diet and exercise intervention. Blood, 12-hr urine, and kidney tissue samples were collected postintervention for detecting blood glucose and lipid levels, expression of MCP-1 and NF-κB, and renal function indices. Results: Postintervention, blood glucose, and lipid levels in Groups C and D were lower than those in Group B, with decreases in Group D significantly greater than in Group C. Every index of renal protection showed greater improvement in Group D than in Group C (p <.05). The expression of NF-κB and MCP-1 was lower in Group D than in Group C (p <.05). Conclusions: The diet and exercise intervention reduced the inflammatory reaction and delayed T2DM and DN progression by inhibiting the activation of NF-κB and downregulating the expression of MCP-1. © 2015, © The Author(s) 2015.

This study was to discuss the clinical diagnosis and treatment features of severe pulmonary infections after renal transplantation, in order to improve the cure rate. Retrospective research was applied to analyze the 52 cases of patients with serve pulmonary infection after renal transplantation in the department of organ transplantation in People's Hospital of Zhengzhou from July, 2008 to December, 2013. The pathogens which caused the pulmonary infection after renal transplantation were bacteria, virus, fungi, mycobacterium tuberculosis, combined infection and unknown pathogens, and the rate were respectively 28.85%, 15.38%, 7.69%, 3.85%, 21.15% and 23.08%. In the 52 cases, there were 41 cases of patients successfully survived, which was 78.85% and 11 cases died, which was 21.15%. The mortality rate of combined infection and unknown pathogens infection was significantly higher than that of single pathogen infection. Pulmonary infection after renal transplantation was of rapid deteriorated progress and high mortality rate. It was significant for patients to receive the comprehensive and integrated treatment.

Objective: To discuss the clinical effects of Western medicine therapy assisted by Ginkgo biloba tablet (GBT) on patients with vascular cognitive impairment of none dementia (VCIND). Methods: A total of 80 patients with VCIND were divided into two groups randomly: Conventional treatment group (control group) and combined treatment group. Conventional treatment group was given conventional treatment with anti-platelet aggregation. In this group, 75 mg aspirin was given three times a day for 3 months. While in combined treatment group, 19.2 mg GBT was given three times a day for 3 months together with conventional treatment (anti-platelet aggregation drugs). Montreal cognitive assessment (MoCA) and transcranial Doppler (TCD) were used to observe changes of cognitive ability and cerebral blood flow in VCIND patients before and after treatment in both groups. Then the clinical data were analyzed so as to compare the efficacy in two groups. Results: After 3 month-treatment in combined treatment group, the scores of executive ability, attention, abstract, delayed memory, orientation in the MoCA were significantly increased compared with those before treatment and those in control group after treatment. Besides, blood flow velocity of anterior cerebral artery increased significantly than that before treatment and that in control group after treatment. Conclusions: GBT tablet can improve the therapeutic efficacy as well improve cognitive ability and cerebral blood flow supply of patients with VCIND. © 2012 Hainan Medical College.

AIM: To observe the effects of salvianolic acid B (SalB) on in vitro growth inhibition and apoptosis induction of retinoblastom a HXO-RB44 cells. METHODS: The effects of SalB on the HXO-RB44 cells proliferation in vitro were observ ed by MTT colorimetric method. The morphological changes of apoptosis before and after the treatm ent of SalB were observ ed by Hoechst 33258 fluorescent staining m ethod. Apoptosis rate and cell cycle changes of HXO-RB44 cells were detected by flow cytometer at 48 hours after treated by SalB. The expression changes of Caspase-3 protein in HXO-RB44 cells w ere detected by Western Blot. RESULTS: SalB significantly inhibited the growth of HXO-RB44 cells, while the inhibition was in a concentration-and tim e-dependent m anner. The results of fluorescent staining m ethod indicated that HXO-RB44 cells show ed significant phenom enon of apoptosis including kary orrhexis, fragm entation and the form ation of apoptotic bodies, etc. after 24, 48 and 72 hours co-culturing of SalB and HXO-RB44 cells. The results of flow cytom eter show ed that the apoptosis rate and the proportion of cells in Sphase were gradually increased at 48 hours and 72 hours after treated by different concentrations of SalB. Western Blot strip showed that the expression of Caspase-3 protein in HXO-RB44 cells was gradually increased with the increase of the concentration of SalB. CONCLUSION: SalB can significantly affect on HXO-RB44 cells growth inhibition and apoptosis induction which may be achieved through the up-regulation of Caspase-3 expression and the induction of cell cy cle arrest. AIM: To explore the heart protection effects by RNA interference of Caspase-3 on rat acute myocardial infarction (AMI). MATERIALS AND METHODS: 45 SD rats were randomly divided into sham group, AMI group and Caspase-3-siRNA group. Animal model of AMI was established by ligation of anterior descending branch (LAD). Rats of sham operation group and AMI group were administered with lentivirus harboring empty vector into myocardial tissue. Rats of Caspase-3-siRNA group were administered with lentivirus harboring Caspase-3 RNA interference vector. After 72 hours, the heart function were evaluated by echocardiogram analysis. Then the rats were sacrificed and the myocardial tissue were collected. The expression level of Caspase-3 mRNA and protein in each group were analyzed by RT-PCR and Western blot, respectively. Moreover, the infarct size was analyzed by triphenyltetrazolium chloride (TTC) staining and the apoptosis index of myocardial cells were analyzed by TUNEL assay. RESULTS: Compared with the sham operation group, the expression levels of Caspase-3 mRNA and protein were significantly upregulated (p < 0.05) and the Caspase-3 activity was enhanced (p < 0.05) in AMI group and Caspase-3-siRNA group. However, the Caspase-3-siRNA group showed lower expression levels of Caspase-3 mRNA/protein and weaker Caspase-3 activity, compared with AMI group (p < 0.05). Furthermore, the apoptosis index and the infarction range of myocardial cells were enhanced in AMI group and Caspase-3-siRNA group (p < 0.05). The apoptosis index and infarction range of myocardial cells in Caspase-3-siRNA group were decreased (p < 0.05). Interestingly, the Left Ventricular End-Diastolic dimension (LVEDd) and the Left Ventricular End-Systolic diameter (LVESd) in AMI group and Caspase-3-siRNA group were improved (p < 0.05), but the Ejection Fraction (EF) and Fractional Shortening (FS) in AMI group and Caspase-3-siRNA group were reduced (p < 0.05). All the LVEDd, LVESd, EF and FS in Caspase-3-siRNA group significantly improved than that of AMI group (p < 0.05). CONCLUSIONS: The downregulation of Caspase- 3 mediated by lentivirus interference can decrease the infarct size of myocardial tissue and the apoptosis index of myocardial cells. It also can improve heart function in rats with acute myocardial infarction.

This study discussed about the influence of local application of glaucoma medications -- travoprost eye drops to patients' tear film function. We selected 24 patients, 45 eyes with primary open-angle glaucoma or intraocular hypertension. All of the patients topically used the travoprost eye drops for one time every night. After and before the pharmacy, we proceeded 1, 2, 3 mo lines symptom score and Schirmer's test (St), corneal fluorescein staining (FL), breakup time of tear film (BUT). Average value of symptom score and FL of all the patients before pharmacy were 1.32 ± 1. 55, 0.42 ± 0.68, and 1, 2, 3mo after pharmacy were respectively 2.68 ± 1.59, 0.96 ± 0.81; 4.97 ± 1.62, 1.46 ± 0.62; 6.21 ± 1.33, 1.88 ± 0.44. Symptom score and FL of 1, 2, 3 mo patients after pharmacy were all prominent higher than it before pharmacy (P=0.00), and it gradually increased. The average value of patients symptom BUT and St before pharmacy were (7.71 ± 0.87s), (8.32 ± 2.63mm /5min) and 1, 2, 3 mo after pharmacy were respectively (6.93 ± 1.17s), (7.69 ± 3. 33mm /5min); (5.48 ± 1.29s), (6.79 ± 2.94mm /5min); (4.33 ± 1.83s), (5.98 ± 3.11mm/5min). BUT and St value after pharmacy were prominent all lower than the level before pharmacy (P=0.00). And it gradually reduced. Short-term use of travoprost eye drops would aggravate the corneal irritation of patients, and decrease the tear film stability and tear secretion.

After the mir-155-5p over expressed in gastric cancer cells, the expression profile chip was adopted to screen its target genes. Some of the intersection of target genes were selected based on the bioinformatics prediction, in order to study the mechanism of its function and role of research. Affymetrix eukaryotic gene expression spectrum was conducted to screen mir-155-5p regulated genetic experiment. Western blot technique was employed to detect and screen the protein expression of target genes. Mimics was transfected in BGC-823 of gastric cancer cells. Compared with mimics-nc group and mock group, the mRNA expression quantities of SMAD1, STAT1, CAB39, CXCR4 and CA9 were significantly lower. After the gastric cancer cells BGC-823 and MKN-45 had been transfected by mimics, compared with mimics-nc (MNC) group and mock (MOCK) group, it was decreased for the protein expression of SMAD1, STAT1 and CAB39 in mimics (MIMICS) group. The verification of qRT-PCR demonstrated that SMAD1, STAT1, CAB39, CXCR4 and CA9 were the predicted target genes and target proteins of mir-155-5p, the over expression of mir-155-5p could enable the decreasing of its expression level in gastric cancer cells MKN-45 and BGC-823.

Objective: To probe into the influences of Chinese herbal compound on the growth and metabolism of weight-bearing bones of tail-suspended rats. Methods: Twenty-four male SD rats were randomly divided into blank control group (eight), tail-suspended control group (eight) and Chinese medicine treatment group (eight) according to their weights. No treatment was done for the blank control group. Double distilled water lavage was performed daily for the tail-suspended control group. On the basis of the tail-suspended rat model, the rats were given Chinese herbal compound lavage every day in Chinese medicine treatment group. This compound includes mulberry, Poria cocos and barbary wolfberry, etc. The test cycle was four weeks. The rats were killed after the experiment. The right femoral bone was taken out for the physical measurements, and the left femoral bone was for the three-point bending test. The influences of Chinese herbal compound on femoral bone growth and biomechanical properties of simulated weightlessness rats were observed. Results: (1) After simulated weightlessness (tail-suspension), compared with the blank control group, all the physiological indexes of rat femoral bone decreased in tail-suspended group and Chinese medicine treatment group (P<0.05). The strength and rigidity of rat femoral bone decreased in tail-suspended group (P<0.01). The maximum load and rigidity coefficient also decreased with the increasing toughness coefficient in the control group (P<0.01). (2) After the countermeasure of Chinese herbal compound, each biomechanical indexes showed the tendency of increasing in Chinese medicine treatment group, and theses indexes were close to those of the blank control group (P<0.05), which indicated that the bone loss caused by simulated weightlessness was improved. Conclusions: Chinese herbal compound for tonifying kidney could effectively prevent the bone loss and have some enhancements on the bone biomechanical properties. © 2013 Hainan Medical College.
