Mitochondrial fission is predominantly controlled by the activity of dynamin-related protein1 (Drp1), which has been reported to be involved in mitochondria apoptosis pathways. However, the role of Drp1 in a rat model of cardiac arrest remains unknown. In this study, we found that activation of Drp1 in the mitochondria was increased after cardiac arrest and inhibition of Drp1 by 1.2 mg/kg of mitochondrial division inhibitor-1 (Mdivi-1) administration after the restoration of spontaneous circulation (ROSC) significantly protected against cerebral ischemic injury, shown by the increased 72-h survival rate and improved neurological function. Moreover, the increase of the vital neuron and the reduction of cytochrome c (CytC) release, apoptosis-inducing factor (AIF) translocation and caspase-3 activation in the brain indicate that this protection might result from the suppression of neuron apoptosis. Altogether, these results indicated that Drp1 is activated after cardiac arrest and the inhibition of Drp1 is protective against cerebral ischemic injury in a rat of cardiac arrest model via inhibition of the mitochondrial apoptosis pathway.

BACKGROUND: Osteogenic induction has been regarded as an indispensable step for adipose-derived stromal cells (ADSCs) to transform into osteoblasts, thus to secret bone matrix to repair bone defects. However, this conclusion has not been confirmed. OBJECTIVE: Tissue-engineered constructs were prepared from osteogenically induced ADSCs and scaffolds. The constructs were implanted into bone defects and non-bone areas. Whether osteogenically induced ADSCs were transformed into osteoblasts was determined according to bone formation. METHODS: ADSCs were prepared by collagenase I digestion of subcutaneous fat from the back of dogs aged 12 months. Passage 3 ADSCs were harvested and seeded on the biphasic calcium phosphate ceramics to form a cell-scaffold construct. Box-like bone defects, 20 mm in length and 10 mm in height, were created in the mandibular bilaterally in each dog along the alveolar bone where teeth had been extracted. Then the cell-scaffold constructs were implanted in dog mandibular bone defects. An empty defect group was used as control. The tissue-engineered constructs and osteoinductive calcium phosphate ceramics were implanted in muscle. After implantation for 6 and 12 weeks, bone formation was analyzed using histomorphometry. RESULTS AND CONCLUSION: ADSCs-scaffold construct promoted bone formation in bone defect area, but not in muscle area, however, osteoinductive calcium phosphate ceramics promoted bone formation in muscle area. ADSCs were not transformed into osteoblasts after osteogenic induction. The precise mechanism needs further investigation.

BACKGROUND: Some studies have showed that after indirect co-culture, bone marrow mesenchymal stem cells can differentiate into myocardial cells and hepatocypte-like cells. OBJECTIVE: To investigate the possibility of human umbilical cord blood mesenchymal stem cells (UCB-MSCs) to differentiate into hepatocytes by co-culture with human hepatocyte line LO2 in vitro. METHODS: Full-term umbilical cord blood samples were obtained sterilely. The UCB-MSCs were isolated by density gradient centrifugation and directly adherence growth, then passaged with trypsin digestion at 80% cell fusion. By utilizing cell culture plate insets with microporous membrane combined with 6-well plate, the LO2-/UCB-MSCs co-culture system was established. UCB-MSCs were plated into the wells of 6-well plate at a density of 1×107/L. LO2 cells were plated into the cell culture plate insert at a density of 1×10 5/L. UCB-MSCs were plated in both layers in the control group. Surface markers of adhered cells were detected by flow cytometry. Morphological changes of UCB-MSCs were observed by inverted phase contrast microscopy. mRNA expression was detected by reverse transcriptase PCR. RESULTS AND CONCLUSION: HUCB MSCs expressed CD44 and CD29 strongly, but CD34 and CD45 were expressed negatively. After 5 days, fusiform-shaped cells were reduced in the co-culture group; while, the time passing by, cells shaped irregular round or polygonal were increased, which were similar to hepatocytes. At 4 weeks after culture, UCB-MSCs were still fusiform-shaped in the control group. At day 5 after culture, alpha fetoprotein mRNA expressed positively, but other expressed negatively in the co-culture group; at day 14 after culture, cytokeratin-19 mRNA and albumin mRNA expressions were observed; moreover, with the time passing by, the expression of albumin mRNA was increased, but the expression of alpha fetoprotein-19 mRNA was decreased. Antigenic expressions in the control group were negative. This suggested that UCB-MSCs could differentiate into hepatocypte-like cells by co-culture with human hepatocyte line LO2 in vitro.

AIM: To investigate the expression and relationship of Survivin and Fas in primary pterygium. METHODS: SP immunohistochemistry was used to detect the expression of Survivin protein and Fas protein in primary pterygium of 40 cases and nomal conjunctiva of 20 cases. All data were analyzed by SPSS 11.5. RESULTS: The positive expression of Survivin in pterygium (37.5%) was higher than that in normal conjunctiva tissues(10.0%, P<0.05). The positive expression of Fas in primary pterygium was significantly higher (75.0%) than that in normal conjunctiva tissues (15.0%, P<0.05). There was a positive correlation between the expression rate of Survivin and Fas in primary pterygium(P<0.01) and there was consistency between the expression strength of Survivin and Fas in primary pterygium(P>0.05). CONCLUSION: It indicated that Survivin may play an important role in the pathway of development of primary pterygium. There was a close correlation between the expression of Survivin and Fas in primary pterygium. It also indicated that Survivin and Fas may commonly participate in the occurrence and development of primary pterygium.

• AIM: To explore the therapeutic method of treating chronic dacryocystisis with nasal endoscopic dacryocystorhinostomy. • METHODS: A total of 30 patients (32 eyes) of chronic dacryocystitis were submitted to nasal endoseopic dacryocystorhinostomy. They underwent the irrigation of lacrimal passage after the surgery, and were followed up by endoscopy for aperiod range from 1-3 years. • RESULTS: Twenty-six eyes (81%) were cured, 4 eyes (13%) were improved, and the total effective rate reached to 94%. • CONCLUSION: Nasal endoscopic dacryocystorhinostomy for the chronic dacryoeystitis is an ideal way to treat the chronic dacryocystitis , because it possesses the advantages of small tissue injuries, simple and quick operation, fewer complications.

• AIM: To explore the correlated factors between multivariate ocular factors and the development of dry eye. • METHODS: Five hundred and thirty-six cases with dry eye were selected and evaluated with a series of ocular testing of the Schirmer test, the tears film stability, the staining with fluorescein sodium on cornea. A questionnaire about dry eye was surveyed. The multivariate nonconditional Logistic regression analysis was used to analyze the relation between multivariate ocular factors and the development of dry eye. • RESULTS: The Logistic regression predictive equation for dry eye was: Y = -2.21 + 0.459 X3 + 10.572 X4-0.069 X5 + 0.766 X6 + 0.924 X7 + 0.264 X9-1.78 X12 + 7.235 X13 + 4.917 X14 + 5.148 X15; P = exp (Y)/[1 + exp(Y)]. The P value meant the probability of suffering dry eye. • CONCLUSION: Our data showed that multivariate ocular factors might play a role in the development of dry eye. The clinical features of dry eye are significant for the diagnosis, and it can be predicted by using the suitable logistic predictable equation.
