The present study was conducted to study the crenellation patterns of semen and their relation with different seminal characteristics and post-thaw motility. Dried semen samples were classified into 6 grades (A1, A2, B1, B2, C1 and C2) based on the crenellation patterns. The study showed that breeds/strains had highly significant (P< 0.01) effect on different seminal traits. Significant (P< 0.05) effect of month of collection on sperm concentration and post-thaw motility was also observed. Crenellation patterns had highly significant (P< 0.01) effect on sperm concentration, initial motility and post-thaw motility. Sperm concentration, Initial motility and post-thaw motility was superior in A1, A2 and B1 grade and inferior in C1 and C2. B2 grade was of intermediate quality semen. A1, A2 and B1 grade ejaculates gave best post-thaw motility after cryopreservation. Poor post-thaw motility was found in C1, C2 and B2 grade ejaculates. Thus the crenellation can be effectively used as an indicator for simple and quick evaluation of semen quality in any semen laboratory as well as, at field level. Source

In this study, the standard high speed cell sorter was used for sorting of Sahiwal bull's sperm. Reasonable conception rate was achieved after insemination of sorted frozen semen to heifers and cows which was significantly lower than the unsorted frozen semen. After faster sorting it was possible to get average 71% female calf by AI with sorted frozen Xsperm which was much higher than the male calf. However it is expected that optimization of this current methodology and some modification of cell sorter may increase the purity of sorted sperm at high speed which may reduce damage of the sperm and improve post thaw motility, hypo-osmotic swelling response and acrosome integrity of sorted sperm. Nevertheless more research is required to develop a method which can minimize the damaging effect of sorting sperm and increase the purity and conception rates in artificial insemination programme at field level. It is inferred from this study that presence of XX and XY cells in heterosexual dizygotic twins produce chimerism.Chimerism may be produced due to vascular anastomoses of foetal membranes during early embryonic life before the critical period of sex differentiation. In this study 60 to 64% pure cell lines were observed in both sexes. With this pure cell lines abnormal development of reproductive organs was found in freemartin heifer. Source
