While designing an optics lab for a conceptual physics course, I came across a "box theater" activity.1 The box theater is a pinhole camera obscura made from a box that students put over their heads and shoulders. I use the activity as a capstone experience to explain optical systems. (Classroom demonstrations of the camera obscura have been described by others.2) First, the students build and experiment with a camera obscura made from a plastic cup and a convex lens with a focal length of 7.5 cm, and then "wear" the box theater. The difficulty with the box theater is the dimness of the image. A cloth drape has to be hung from the bottom of the box around the shoulders of the students to prevent light leakage, and the students have to wait a few minutes for their eyes to adjust to the darkness.

Temperate phages are common, and prophages are abundant residents of sequenced bacterial genomes. Mycobacteriophages are viruses that infect mycobacterial hosts including Mycobacterium tuberculosis and Mycobacterium smegmatis, encompass substantial genetic diversity and are commonly temperate. Characterization of ten Cluster N temperate mycobacteriophages revealed at least five distinct prophage-expressed viral defence systems that interfere with the infection of lytic and temperate phages that are either closely related (homotypic defence) or unrelated (heterotypic defence) to the prophage. Target specificity is unpredictable, ranging from a single target phage to one-third of those tested. The defence systems include a single-subunit restriction system, a heterotypic exclusion system and a predicted (p)ppGpp synthetase, which blocks lytic phage growth, promotes bacterial survival and enables efficient lysogeny. The predicted (p)ppGpp synthetase coded by the Phrann prophage defends against phage Tweety infection, but Tweety codes for a tetrapeptide repeat protein, gp54, which acts as a highly effective counter-defence system. Prophage-mediated viral defence offers an efficient mechanism for bacterial success in host-virus dynamics, and counter-defence promotes phage co-evolution.

Hurricane Katrina created a variety of issues that tested the resilience of families and children who were forced to relocate. This article describes the use of culturally affirming, thematically appropriate bibliotherapy as part of a long-term program to assist a group of elementary school aged African-American children cope with feelings of anxiety, displacement, and loss. This intervention provided a medium through which the participants could explore issues of isolation and the loss of their homes, family structures and a sense of security, while also providing academic and social supports. © 2014, Springer International Publishing.

Threespine stickleback were used to examine phenotypic plasticity of telencephala in relation to inferred ecology. Fish from derived, allopatric, freshwater populations were sampled from three shallow, structurally complex lakes with benthic-foraging stickleback (benthics) and from three deep, structurally simple lakes with planktivores (limnetics). The telencephalon of specimens preserved immediately after capture (field-preserved), field-caught fish held in aquaria for 90 days (lab-held), and lab-bred fish from crosses and raised in aquaria were compared. Field-preserved sea-run (ancestral) stickleback were collected from two separate sites, and parents of lab-bred sea-run stickleback were collected from one of these sites. In field-preserved and lab-held fish, the telencephala of limnetics exhibited triangular dorsal shape, while those of benthics and sea-run fish had rounder shapes. No such pattern was detected in lab-bred fish. Within each treatment type, benthics had larger relative telencephalon sizes, using overall brain size as the covariate, than limnetics. Among field-preserved samples, sea-run fish had smaller telencephalon sizes than lake fish. Intra-population analyses of lake samples showed that field-preserved fish consistently had larger relative telencephalon sizes than lab-bred fish. The opposite was true of the sea-run population. In a separate study using one benthic population and one limnetic population, samples were preserved in the field immediately or held in the lab for 30, 60, and 90 days before they were sacrificed. In both populations, the telencephalon shapes of lab-held fish were similar to those of field-preserved fish but became progressively more like lab-bred ones over 90 days. In contrast, relative telencephalon size decreased dramatically by 30 days after which there was little change. In freshwater threespine stickleback, the telencephalon exhibits considerable phenotypic plasticity, which was probably present in the ancestor. © 2012 Current Zoology.

In all, more than 30 colleges are being recognized for offering affordable, flexible online education options in the state. "There are over 1.2 million students enrolling in New York college and university programs annually,” said Dan Schuessler, CEO and Founder of AffordableCollegesOnline.org. "As more students apply to these schools, more also seek information on non-traditional and remote learning. Prostate cancer (PCa) is the second leading cause of cancerrelated death in American men and many PCa patients develop skeletal metastasis. Current treatment modalities for metastatic PCa are mostly palliative with poor prognosis. Epidemiological studies indicated that patients receiving the diabetic drug metformin have lower PCa risk and better prognosis, suggesting that metformin may have antineoplastic effects. The mechanism by which metformin acts as chemopreventive agent to impede PCa initiation and progression is unknown. The amplification of c-MYC oncogene plays a key role in early prostate epithelia cell transformation and PCa growth. The purpose of this study is to investigate the effect of metformin on c-myc expression and PCa progression. Our results demonstrated that (i) in Hi-Myc mice that display murine prostate neoplasia and highly resemble the progression of human prostate tumors, metformin attenuated the development of prostate intraepithelial neoplasia (PIN, the precancerous lesion of prostate) and PCa lesions. (ii) Metformin reduced c-myc protein levels in vivo and in vitro. In Myc-CaP mouse PCa cells, metformin decreased c-myc protein levels by at least 50%. (iii) Metformin selectively inhibited the growth of PCa cells by stimulating cell cycle arrest and apoptosis without affecting the growth of normal prostatic epithelial cells (RWPE-1). (iv) Reduced PIN formation by metformin was associated with reduced levels of androgen receptor and proliferation marker Ki-67 in Hi-Myc mouse prostate glands. Our novel findings suggest that by downregulating c-myc, metformin can act as a chemopreventive agent to restrict prostatic neoplasia initiation and transformation. © The Author 2013. Diabetes mellitus is an enormous menace to public health globally. This chronic disease of metabolism will adversely affect the skeleton if not controlled. Both type 1 diabetes mellitus (T1DM) and type 2 diabetes mellitus (T2DM) are associated with an increased risk of osteoporosis and fragility fractures. Bone mineral density is reduced in T1DM, whereas patients with T2DM have normal or slightly higher bone density, suggesting impaired bone quality is involved. Detrimental effects of T1DM on the skeleton are more severe than T2DM, probably because of the lack of osteo-anabolic effects of insulin and other pancreatic hormones. In both T1DM and T2DM, low bone quality could be caused by various means, including but not limited to hyperglycemia, accumulation of advanced glycosylation end products (AGEs), decreased serum levels of osteocalcin and parathyroid hormone. Risk for osteoarthritis is also elevated in diabetic population. How diabetes accelerates the deterioration of cartilage remains largely unknown. Hyperglycemia and glucose derived AGEs could contribute to the development of osteoarthritis. Moreover, it is recognized that oral antidiabetic medicines affect bone metabolism and turnover as well. Insulin is shown to have anabolic effects on bone and hyperinsulinemia may help to explain the slightly higher bone density in patients with T2DM. Thiazolidinediones can promote bone loss and osteoporotic fractures by suppressing osteoblastogenesis and enhancing osteoclastogenesis. Metformin favors bone formation by stimulating osteoblast differentiation and protecting them against diabetic conditions such as hyperglycemia. Better knowledge of how diabetic conditions and its treatments influence skeletal tissues is in great need in view of the growing and aging population of patients with diabetes mellitus. © 2013 Higher Education Press and Springer-Verlag Berlin Heidelberg.

Background: Spatial learning is the ability to learn and use features in space to navigate within an environment. In fishes, it is associated with residence in structurally complex habitat, but very little is known about its ancestral condition in adaptive radiations. Goal: To investigate the relationship between foraging mode and spatial learning in derived populations of a well-studied evolutionary model fish species, and to test the importance of experience on spatial learning in an ancestral analogue of this species. Organism: Threespine stickleback (Gasterosteus aculeatus) occur as sea-run (ancestral) and divergent freshwater (derived) forms. Freshwater populations occupy an ecological continuum, with benthic stickleback at one extreme, residing and foraging in shallow, structurally complex lakes, and limnetics at the other, living and feeding in the open water of deep lakes that lack structure. Methods: I used a T-maze to measure spatial learning. In Experiment 1, I compared five benthic and five limnetic field-caught stickleback populations to explore ecological divergence of spatial learning. In Experiment 2, I used a sea-run population to infer the ancestral condition of spatial learning; I studied laboratory-reared sea-run fish raised in spatially complex or simple aquaria because adequate samples of field-caught sea-run adults proved difficult to attain. Results: In Experiment 1, benthics exhibited better spatial learning than limnetics. These differences were independent of differences in boldness, exploratory behaviour, activity level, or other performance variables that are independent of spatial learning. In Experiment 2, no differences were detected between rearing treatments, but a number of fish from either group still solved the maze, indicating that even fish reared in spatially simple conditions were capable of spatial learning. Progesterone receptor membrane component 1 (PGRMC1) is a 25 kDa protein with an N-terminal transmembrane domain and a putative C-terminal cytochrome b5 domain. Heme-binding activity of PGRMC1 has been shown in various homologues of PGRMC1. Although the general definition of PGRMC1 is as a progesterone receptor, progesterone-binding activity has not been directly demonstrated in any of the purified PGRMC1 proteins fully loaded with heme. Here, we show that the human homologue of PGRMC1 (hPGRMC1) binds heme in a five-coordinate (5C) high-spin (HS) configuration, with an axial tyrosinate ligand, likely Y95. The negatively charged tyrosinate ligand leads to a relatively low redox potential of approximately -331 mV. The Y95C or Y95F mutation dramatically reduces the ability of the protein to bind heme, supporting the assignment of the axial heme ligand to Y95. On the other hand, the Y95H mutation retains ∼90% of the heme-binding activity. The heme in Y95H is also 5CHS, but it has a hydroxide axial ligand, conceivably stabilized by the engineered-in H95 via an H-bond; CO binding to the distal ligand-binding site leads to an exchange of the axial ligand to a histidine, possibly H95. We show that progesterone binds to hPGRMC1 and introduces spectral changes that manifest conformational changes to the heme. Our data offer the first direct evidence supporting progesterone-binding activity of PGRMC1. © 2015 American Chemical Society.
