Glutathione S-transferase (GST) enzyme levels are associated with risk of many cancers, including hematologic tumours. We here aimed to investigate the relationships between GSTM1, GSTT1 and GSTP1 polymorphisms and the risk of AML. Genotyping of GSTs was based upon duplex polymerase-chain-reactions with the confrontingtwo- pair primer (PCR-CTPP) method in 163 cases and 204 controls. Individuals carrying null GSTT1 genotype had a 1.64 fold risk of acute leukemia relative to a non-null genotype (P<0.05). A heavy risk was observed in those carrying combination of null genotypes of GSTM1 and GSTT1 and GSTP1 Val allele genotypes when compared with those carrying wild genotypes, with an OR (95% CI) of 3.39 (1.26-9.26) (P<0.05). These findings indicate that genetic variants of GST and especially the GSTT1 gene have a critical function in the development of AML. Our study offers important insights into the molecular etiology of AML.

Primary thyroid lymphoma (PTL) is a rare malignant thyroid tumor; its pathogenesis is closely related to chronic lymphocytic thyroiditis. The different pathological subtypes and stages of PTL have distinct clinical characteristics and prognosis, but the specific reasons are not clear. Wnt5a is a representative protein of non-canonical Wnt signaling. It plays an important role in many different types of tumors. This study is to explore the changes of Wnt5a and its receptor Ror2 in PTL development process and the clinical significance of their represent. We collected 22 PTL patient tumor specimens and clinical data. We observed the expression of Wnt5a and Ror2 in PTL tumor tissues by immunohistochemistry. Wnt5a was expressed positively in 12 (54.5 %) cases, and Ror2 was expressed positively in 18 (81.8 %) cases. The expression of Wnt5a had a significant difference in different pathological subtypes of PTL (P < 0.05). Wnt5a and Ror2 expression were associated with local invasion and clinical stage, respectively (P < 0.05), and had no significant correlation with age, gender, and tumor size. Although, no significant difference in overall survival was found between positive and negative groups of Wnt5a (P = 0.416) or Ror2 (P = 0.256), respectively. We still consider that Wnt5a and Ror2 play a complex and subtle role in the pathogenesis and progression of PTL and may become potential biomarkers and therapeutic targets of PTL. © 2015 International Society of Oncology and BioMarkers (ISOBM)

OBJECTIVE: To sum the research progress Numb with epithelial mesenchymal transition, and discuss the possible role of Numb in the epithelial-mesenchymal transition mechanism. METHODS: Articles with numb and EMT as key words between 1990-01-2012-10 were retrieved in PubMed and CNKI databases. Inclusion criteria: 1) The relationship of numb and epithelial-mesenchymal transition; 2) The mechanism of numb in the epithelialmesenchymal transition. RESULTS: Numb is involved in an important component of the tumor signal transduction pathways, new research suggests Numb downregulated or missing, EMT occurrence exacerbated. There is gene variation of Notch1 in 10% tumor tissues. The mechanism of action may affect the positioning of E-cadherin and regulate of Notch signaling pathway, that may play a critical role in tumor EMT process. Induced tumor formation and development through participation in EMT may be another important mechanism of Numb affecting cancer development. CONCLUSION: Numb expression level of the tumor and its associated signaling pathway may play an important role in epithelial-mesenchymal transition, but the specific mode of action and mechanisms not yet been elucidated, which needs further investigation.

SIRT2 is a member of the NAD+ dependent deacetylases. In this study, the associations between SIRT2 expression and molecular and clinical characteristics of patients with acute myeloid leukemia (AML) were evaluated by data from The Cancer Genome Atlas. SIRT2 was overexpressed in the intermediate- and poor-risk groups of patients, compared to the favorable-risk group of patients (P=0.002 and 0.004, respectively). High SIRT2 expression was associated with significantly shorter overall survival (OS; P=0.0005) and event-free survival (EFS; P=0.0002) than low SIRT2 expressio in a cohort of 167 patients with AML. Multivariate analyses revealed that high SIRT2 expression was associated with shorter OS (P=0.031) and EFS (P=0.020). Gene-expression profiling showed 259 differential expressed genes including CD4, CD14 and IL10. Gene sets like MAPK signaling pathway, VEGF signaling pathway and acute myeloid leukemia were upregulated in SIRT2(high) patients. We also found different methylation patterns in these two groups. OS and EFS of SIRT2(high) patients who did not undergo transplantation were significantly shorter than those of SIRT2(low) patients (P=0.0120 and P=0.0107, respectively). Taken together, these findings suggest that high SIRT2 expression is a novel and unfavorable prognostic biomarker for AML risk-stratification.

The ethanol extract of the whole plants of Viburnum mongolicum afforded six new nor-dammarane triterpenoids: 3β,12β-dihydroxy-25,26,27- trinordammara-22-en -24,20-olide (1), 3β,12β-dihydroxy-24α- methoxy-25,26,27-trinordammara-20,24-epoxy (2), 3β-O-acetyl-12β- hydroxy-23,24,25,26,27-hexanordammarane-20-one (3), 12β-Oacetyl- 15α-hydroxy-17β-methoxy-3-oxo-20,21,22-23,24,25,26, 27-octanordammanrane (4), 12β-O-acetyl-15α,17β-dihydroxy-3-oxo- 20,21,22-23,24,25,26,27-octanordammanrane (5), and 12β,15α-dihydroxy- 3-oxo-17-en-20,21,22-23,24,25,26,27-octanordammanrane (6), together with two known nor-dammarane triterpenoids, 12β-hydroxy-3-oxo-24α-methoxy- 25,26,27-trinordammara-20,24-epoxy (7) and 3β,12β-dihydroxy-23,24,25, 26,27- hexanordammarane-20-one (8). The structures of the isolated compounds were established based on 1D and 2D (1H-1H COSY, HMQC, and HMBC) NMR spectroscopy, in addition to high resolution mass spectrometry. The isolated compounds were tested in vitro for cytotoxic potential against seven tumor cell lines and radical scavenging activities. Compounds 4-6 exhibited significant cytotoxic activities against all tested tumor cell lines and radical scavenging activities against ABTS+ radicals comparable with the standard drug Trolox. © 2013 by the authors.

AIM: To observe the clinical significance of travoprost for prevention of high intraocular pressure (IOP) within 24 hours after diabetic retinopathy (DR) vitrectomy. METHODS: A total of 102 patients (102 eyes) with DR who underwent vitrectomy and related literatures were analyzed and compared. All patients were randomly assigned to group A (vitrectomy combined with intravitreal silicone oil, C3F8 or balanced salt solution tamponade group, 51 patients 51 eyes) or group B (surgical therapy is the same as the group A, while instilling one drop of 0.04g/L travoprost eyedrops in the inferior cul-de-sac about 60 minutes before the surgery, 51 patients 51 eyes). The changes of IOP, best-corrected visual acuity (BCVA) and ophthalmoscope observations within 24 hours respectively before and after the surgery were counted and analyzed. RESULTS: The mean IOP of each group within 24 hours after the surgery was higher than the contralateral eyes (t=2.17 and 2.09, P<0.05) and the mean IOP within 24 hours before the surgery (t=2.41 and 2.28, P<0.05). The incidence of high IOP (the high IOP was defined as mean IOP≥25mmHg) and decreased BCVA in group A transcended group B (χ2=4.86 and 3.99, P<0.05). Meanwhile, the mean BCVA of group A within 24 hours after the surgery was slightly worse than that of group B (t=1.43, P>0.05). CONCLUSION: The preliminary result in our scrutiny showed that the preoperative treatment of 0.04g/L travoprost eyedrops was well tolerated with the significant reduction in the incidence of high IOP and decrease in BCVA within 24 hours after DR vitrectomy. It has clinical significance about the prevention of visual impairment owing to high IOP within 24 hours after DR vitrectomy.

To construct a mouse model of highly metastatic gastric lymphoma with orthotopic transplantation of human primary gastric lymphoma specimen. A fresh surgical specimen of primary gastric lymphoma was obtained intraoperatively and implanted into the submucosa of stomach in nude mice. Tumor formation, invasion, metastasis, morphological characteristics under light microscopy and electron microscopy, immunohistochemistry,and the karyotype of orthotopically transplanted tumor cells were studied. An orthotopic highly metastatic model of human primary gastric lymphoma in nude mice(HGBL-0305) was successfully established. Histopathology of transplanted tumors showed primary gastric diffuse large B cell lymphoma. CD19, CD20, CD22 and CD79alpha were positive, while CD3 and CD7 were negative. The number of chromosome ranged from 56 to 69. DNA index(DI) was 1.47+/-0.12(i.e. heteroploid). Until now, HGBL-0305 model has been maintained for 45 generations by orthotopic passage for almost 4 years in nude mice. A total of 156 nude mice were used for transplantation. The growth rate and resuscitation rate of liquid nitrogen cryopreservation of transplanted tumor cells were both 100%. The autonomic growth of the transplanted tumor cells invaded and destructed all the layers of the nude mice stomach. The metastasis rates of liver, spleen, lymph node, and peritoneal seeding were 69.5%, 55.6%, 45.7%, and 30.5%, respectively. An orthotopic highly metastatic model of human primary gastric lymphoma in nude mice is successfully established. HGBL-0305 model may simulate the natural course of primary gastric lymphoma in human and provides an ideal animal model for studies on pathogenesis, metastasis biology and anti-metastatic therapies of primary gastric lymphoma.
