To investigate the role and potential molecular mechanism of Galectin-3 (Gal-3) in the etiology of endometriosis-associated infertility.We detected Gal-3 expression in eutopic endometrium from women with endometriosis-associated infertility and healthy women without endometriosis or infertility. We then evaluated Gal-3 expression in endometrial glandular epithelial cells (EECs) and endometrial stromal cells (ESCs) and investigated its response to hormone stimulation in EECs and ESCs from both groups of women.Results of real-time PCR and western blot analysis showed Gal-3 expression in both proliferative and secretory stages of the menstrual cycle decreased significantly in women with endometriosis-associated infertility compared to healthy women. The changes in expression of Gal-3 were more dramatic in EECs than ESCs. Moreover, estrogen (E2) and progesterone (P4) induced Gal-3 expression in EECs of healthy groups, and P4 was more significant than E2 and combined E2 and P4 (E2P4). However, in the endometriosis group, P4 failed to induce a similar increase in Gal-3 expression.Our results suggest that aberrant expression of Gal-3 might contribute to infertility in patients with endometriosis due to progesterone resistance.

Background and Objectives MicroRNA-203 (miR-203) serves as a tumor suppressor or a tumor promoter in different human malignancies. However, its involvement in human gliomas is still unclear. The aim of this study was to investigate the clinical significance of miR-203 expression in gliomas. Methods Real-time quantitative PCR was employed to measure the expression level of miR-203 in clinical glioma tissues. Results The expression of miR-203 was reduced in high WHO grade glioma tissues compared with that in low WHO grade and normal brain tissues, and decreased with ascending tumor WHO grades (P < 0.001). The reduced miR-203 expression in gliomas was significantly associated with higher WHO grade (P < 0.001), lower KPS score (P = 0.008) and poorer disease-specific survival of patients (P = 0.001). More importantly, subgroup analyses according to tumor WHO grade revealed that the disease-specific survival of patients with low miR-203 expression in high WHO grades (III-IV) subgroup was significantly shorter than those with high miR-203 expression (P < 0.001), but no significant difference was found for patients in WHO grades I-II subgroup (P = 0.08). Conclusion Our data validate an important clinical significance of miR-203 in gliomas, and reveal that it might be an intrinsic regulator of tumor progression and a potential prognostic factor for this dismal disease. The primary function of white adipose tissues is to store excess energy. The current study aimed to investigate the roles of Gremlin 2 (Grem2), a glycoprotein in adipogenesis. Using polymerase chain reaction-based microarrays, it was determined that Grem2 was markedly downregulated in adipose tissues from obese animals and humans. In addition, 3T3-L1 cells were used to investigate the details of the mechanisms underlying the anti-adipogenic effects of Grem2. Grem2 expression was markedly decreased upon the induction of adipocyte differentiation, as demonstrated by reverse transcription-quantitative polymerase chain reaction and western blot analysis. Notably, Grem2 overexpression inhibited adipogenesis, while knockdown of Grem2 led to an increase in adipogenesis. At the molecular level, Grem2 promotes nuclear translocation of β-catenin, an integral Wnt signaling component. Consistently, inhibition of Wnt/β-catenin signaling using a retrovirus targeting the β-catenin coding region attenuated the anti-adipogenic effects of Grem2. Therefore, to the best of our knowledge, the current study shows for the first time that Grem2 may be an important regulator of adipocyte differentiation.

Background: The reference value for the urine albumin/creatinine ratio (ACR) varies between races and has not been previously validated in children. We assessed the ACR reference values and the factors that affect them in a population of healthy Chinese children. Methods: A total of 1986 healthy children (1078 males, 908 females) aged 6-19. y were enrolled. The 95th percentile of ACR was used as the normal upper limit. The associations between ACR and gender, age, body mass index (BMI), systolic blood pressure (SBP), preterm birth, intake of fruits, smoking, and geographical area were examined. Results: The normal upper limit of ACR was 14.7. mg/g for male children and 19.8. mg/g for female children. The ACR value for girls was significantly higher than that for boys (P. <. 0.001). ACR was inversely correlated with age (P. <. 0.001) and positively correlated with BMI, SBP, and smoking (all P. <. 0.01). Conclusions: The ACR reference value for healthy children in southwest China is approximately the same as the value for adults, but lower than that for the Western population. Age, SBP, BMI, and smoking in children influence ACR. © 2014 Elsevier B.V.

A case of AIDS complicated by Penicillium marneffei and Candida albicans infection was reported. Before the scattered papular and contagious molluscum-like lesions appeared, the 21-year-old man had been recurrent fever and weight loss for six months. White cheese-like plaques were found on the surface of the buccal mucosa and tongue. Histopathology of the skin biopsy demonstrated numerous intracellular yeast-like organisms. PAS stain was positive. Most spores aggregated like grape or mulberry, less spore being like sausage with clear septation. The mycological culture of cutaneous lesion revealed Penicillium marneffei growth. Penicillium marneffei and C. albicans were identified by fungal culture of oral secretion. Anti-human immunodeficiency virus (HIV) antibody detection was positive.

To study the role of lipoxin A4 (LXA4) in rat liver transplant rejection.An acute rejection model of liver transplantation was established in inbred rats DA to LEW that were randomly divided into a control group and a LXA4 group. Liver morphologic changes were examined using hematoxylin/eosin staining. Alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were quantified to measure liver injury. Intragraft mRNA and protein expressions of interferon (IFN)- interleukin and (IL)-10 were detected by real-time polymerase chain reaction and Western blots, respectively. The serum levels of IFN- and IL-10 were assayed by enzyme-linked immunosorbent assay.LXA4 treatment improved hepatic tissue injury as indicated by morphologic analysis. Serum ALT and AST levels were significantly decreased at day 7 post-transplantation (P< .05). Concurrently, expression of IFN- was downregulated (P< .05) and secretion of IL-10 was enhanced (P< .05).LXA4 attenuated acute rejection with a parallel shift from Th1 to Th2 responses in rat liver transplantation.
