The present study investigated the anti-obesity and anti-diabetic effects of kaempferol glycoside (KG) fractions which were composed of four kaempferol glycosides and purified from unripe Jindai-soybean (Edamame) leaves in C57BL/6J mice. High fat-fed mice treated with 0.15% dietary KG for 92 days had reduced body weight, adipose tissue and TG levels compared to the high fat-fed control group. KG-treatment also decreased fasting blood glucose, serum HbA1c (hemoglobin A1c) levels and improved insulin resistance. Gene expression analysis of the liver showed that KG decreased peroxisome proliferator-activated receptor (PPAR-γ) and sterol regulatory element-binding protein (SREBP-1c) expression. These results suggest that KG reduced the accumulation of adipose tissue, improving hyperlipidemia as well as diabetes in obese mice by increasing lipid metabolism through the downregulation of PPAR-γ and SREBP-1c. The Northeast sticky bean bun is a popular traditional food in Northeast China with a long history and unique flavor. However, little research has been conducted on the sourdough used to prepare Northeast sticky bean buns. In this study, the polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) technique was employed to study the diversity and dynamic changes of the microflora in homemade sourdough and samples of sourdough used for Northeast sticky bean buns collected from different Northeast provinces. There were six genera and 11 species of bacteria identified from the sourdough of Northeast sticky bean buns, and Lactobacillus fermentum and Lactobacillus plantarum were estimated to be the dominant species during fermentation. Moreover, three genera and four species of fungi were identified from the sourdough of Northeast sticky bean buns, and Saccharomyces cerevisiae was estimated to be the dominant species during fermentation. The microfloral diversity of the homemade sourdough was relatively rich. However, the microfloral diversity and dominant bacterial species continuously changed at different fermentation stages. ©, 2015, South China University of Technology. All right reserved. Source

The aim of this study was to investigate the effect of yeast metallothionein (MT) with known lead-expelling and liver-protecting properties on mice in which, chronic lead poisoning was induced by lead acetate solution gavage for 35 days. Dimercaptosuccinic acid (DMSA) and animal MT (rabbit liver Zn-MT) were used as controls and MT was administered to the mice. Subsequently, the weight, blood lead levels, and liver function indices were measured and a routine blood tests were conducted to observe the severity of pathological changes in the liver tissue. Compared with that of the control group, the weight of MT-treated groups was not significantly different. The blood lead level in high-dose MT-treated group was significantly decreased, while the levels of white blood cells (WBC), mean corpuscular volume (MCV), and hemoglobin (HGB) were significantly increased (p < 0.05). However, there were no significant differences in granulocyte (GR) counts between all groups (p > 0.05). The levels of glutamate pyruvate transaminase (GPT) in all MT-treated groups were significantly decreased (p < 0.05). Meanwhile, the levels of glutamate oxaloacetate transaminase (GOT) in the high-dose MT-treated group were significantly decreased (p < 0.05), pathological changes in liver tissue were ameliorated, and the degree of improvement was higher than that of DMSA group. Two types of yeast metallothionein exhibited lead-expelling and liver-protecting effects on mice with chronic lead poisoning, similar to animal metallothionein. ©, 2015, South China University of Technology. With the extraction liquid of rebaudioside as experimental materials, the influence on the purity and yield of seperation factors of rebaudioside by simulated moving bed chromatography were studied. Used single factor experiment and orthogonal combination experiments to optimize the separation technology parameters of rebaudioside. The optimal process parameters were: switching time was 12 min, the sample concentration was 40 mg/mL, the feed flow rate was 7.5 mL/min, flow rate of first washing stage was 10 mL/min, desorption flow rate was 15.4 mL/min, regeneration flow rate was 7.26 mL/min, flow rate of second washing stage was 20.3 mL/min. The regional optimal allocation and connection were: three parallel and two series connection in the adsorption district, three series connection in the first washing stage, six series connection in the desorption stage, one parallel and two series connection and contrary to the feed direction in the regeneration stage, three series connection in the second washing stage. Under this operating condition, the purity of rebaudioside was 80.69%, yield was 93.48%. © 2016, Chinese Institute of Food Science and Technology. All right reserved. Source
