The aim of this study was to identify antigens for a vaccine or drug target to control rabbit coccidiosis. A combination of 2-dimensional electrophoresis, immunoblotting, and mass spectrometric analysis were used to identify novel antigens from the sporozoites of Eimeria stiedae. Protein spots were recognized by the sera of New Zealand rabbits infected artificially with E. stiedae. The proteins were characterized by matrix-assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF/TOF-MS) analysis in combination with bioinformatics. Approximately 868 protein spots were detected by silver-staining, and a total of 41 immunoreactive protein spots were recognized by anti-E. stiedae sera. Finally, 23 protein spots were successfully identified. The proteins such as heat shock protein 70 and aspartyl prote-ase may have potential as immunodiagnostic or vaccine antigens. The immunoreactive proteins were found to possess a wide range of biological functions. This study is the first to report the proteins recognized by sera of infected rabbits with E. stiedae, which might be helpful in identifying potential targets for vaccine development to control rabbit coccidiosis. © 2017, Korean Society for Parasitology and Tropical Medicine.

1Asymmetric nanomaterials (Janus nanomaterials) comprising at least two components of different chemistry, functionality, and/or polarity have attracted great scientific interest in a wide range of applications. Additional properties are attributed to the asymmetric spatial distribution of functionalities on a single anisotropic nanoparticle, like amphiphilicity or new catalytic characteristic. In addition, Janus nanomaterials' synergistic potential for multilevel targeting, and combination therapies make them particularly attractive for biomedical applications including biosensing, targeting delivery and bio-detection. In this paper, the advances in properties and applications of Janus nanomaterials are summarized. First, some properties and applications of Janus nanomaterials are described in three different aspects: amphipathicity, catalytic characteristic and biocompatibility. Second, the main biomedical applications are highlighted, which include biosensing, targeting delivery, gene vaccine and antimicrobial. The clone library method using PCR amplification of the 16S ribosomal RNA (rRNA) gene was used to identify pathogens from corneal scrapings of C57BL/6-corneal opacity (B6-Co) mice with bacterial keratitis. All 10 samples from the eyes with bacterial keratitis showed positive PCR results. All 10 samples from the normal cornea showed negative PCR results. In all 10 PCR-positive samples, the predominant and second most predominant species accounted for 20.9 to 40.6% and 14.7 to 26.1%, respectively, of each clone library. The predominant species were Staphylococcus lentus, Pseudomonas aeruginosa, and Staphylococcus epidermidis. The microbiota analysis detected a diverse group of microbiota in the eyes of B6-Co mice with bacterial keratitis and showed that the causative pathogens could be determined based on percentages of bacterial species in the clone libraries. The bacterial species detected in this study were mostly in accordance with results of studies on clinical bacterial keratitis in human eyes. Based on the results of our previous studies and this study, the B6-Co mouse should be considered a favorable model for st udying bacterial keratitis. © 2015 Japanese Association for Laboratory Animal Science.

In the long-term process of evolution, both host plants and herbivorous insects form a series of complex defense and anti-defense strategies. In this paper, these strategies are classified into three levels. First, host plants perceive herbivore-associated molecular patterns (HAMPs) and then lead to HAMP-triggered immunity (HTI). This defense is effective against some populations of the herbivore species that cannot secrete effectors. Second, some populations of the herbivore species secrete special effectors to suppress HTI in plants, thereby resulting in herbivore colonization. Third, the effectors of certain populations of the herbivore species could be recognized by resistance genes of host plants, leading to effective effector-triggered immunity and resistance to the herbivore. The elucidation of the mechanisms underlying molecular interactions between host plants and herbivores will not only deepen the understanding of the coevolution between plants and herbivores, but also provide important technical guidance for the breeding of resistant crop cultivars. © 2015, editorial Board of Chinese Journal of Ecology. The simultaneous determination of fluoride and chloride contents in nickel ore by ion chromatography was established. The sample was decomposed by H 2SO 4. After water steam distillation at 160-180°C, the fluoride and chloride in testing solution would be released and absorbed by NaOH solution, realizing the separation from matrix and other elements. Then, the contents of fluoride and chloride were determined by ion chromatography. The high-capacity IonPac ® AS23 anion separation column (4 mm×250 mm) and IonPac ® AG23 protection column (4 mm×50 mm) were used as chromatographic column and 20 mmol/L NaOH solution was used as eluent. Fluoride and chloride were detected with conductivity detector. The results showed that the peak area of fluoride and chloride had good linear relationship with their contents in the range of 1-25 μg/mL. The correlation coefficients (R) were 1.0000 and 0.9996 for fluoride and chloride respectively. The determination results of fluoride and chloride in sample showed high precision. The recoveries of standard addition were all greater than 95%.

The sample was decomposed with hydrochloric acid-nitric acid-hydrofluoric acid-perchloric acid. If the sample was not completely dissolved, 5 mL of nitric acid was added and heated until the brown red fume disappeared. The residue was dissolved in water under slight boiling conditions. Then, the pH value of solution was adjusted with perchloric acid to 0.5. The content of manganese was determined by flame atomic absorption spectrometry (FAAS). Thus, a determination method of manganese in slag of nickel-based materials was established. The mass concentration of manganese showed good linearity to the absorbance under the selected instrumental conditions. The characteristic concentration and precision could meet the detection requirements. The linear regression equation was Y=0.170 1X+0.015 33 with correlation coefficient of R=0.999 3. The detection limit of method was 0.003 4 mg/L. The interference test indicated that the coexisting elements in sample had no interference with the determination. The proposed method was applied to the analysis of five actual samples for slay of nickel matrix material with different manganese content, and the results were consistent with those obtained by standard method GB 11906-1989. The relative standard deviations (RSD, n=11) were in range of 0.9%-1.5%, The determination results were verified by Blas inspection. It indicated that there were no abnormal value for eleven parallel determination. The recoveries were between 97% and 98%. © 2015, Central Iron and Steel Research Institute. All right reserved.

The equipment based on Field Asymmetric Waveform Ion Mobility Spectrometry (FAIMS) chip had been established to detect melamine in diary successfully. Sample was heated and evaporated into detect core with air as the carrier gas through metal diffuser tube and headspace sampler. Signals were collected and transformed into spectrograph. Melamine in diary was confirmed and detected by comparing spectrographs of samples containing and without melamine. The results showed the precision of as-established equipment is lower than LC-MS's and its signal value proportions to the concentration of melamine. The equipment had been scaled with sample containing melamine and set the alarming concentration, so it can be used for detecting melamine quickly.

A method for the determination of melamine in cosmetics was developed by hydrophilic interaction chromatography-tandem mass spectrometry (HILIC-MS/MS). The sample was extracted in turn by aqueous solution of trichloro acetic acid and hexane,and then cleanedup by the mixed-mode cation exchange ( MCX) solid-phase extraction ( SPE) cartridge. A ZICHILIC column was used for the separation by gradient elution, and an electrospray ion trap mass spectrometer was used in the positive ion mode. The linear range of melamine was from 0.02 mg /L to 0.5 mg /L with a correlation coefficient of 0.9985. The limit of detection ( LOD, S/N >3 ) and limit of quantification (LOQ,S /N >10) were 5.0 μg /kg and 20.0 μg/kg ,respectively. The average recoveries and the relative standard deviations ranged from 84.7% to 93.4% and from 4.5% to 8.4% , respectively, in spiked samples at the concentrations from 0.01 mg/kg to 0.1 mg/kg. The method is suitable for the determination of melamine in cosmetics.

Wall-like Ti60 alloy and TiCP (mass fraction 5%)/Ti60 titanium matrix composites are fabricated by laser direct deposition process. Their microstructure and tensile properties under 600°C are investigated. The results indicate that the deposited Ti60 alloy presents a widmanstatten structure with some spherical rare earth phases distributing along the sub-grain boundaries and the α/β interface. For the deposited TiCP/Ti60 composites, there are some un-melted TiC particles and re-solidified TiC particles appear as discontinuous chain-like. The TiC particles distribute uniformly within the matrix with good interface bonding. Under 600°C, the ultimate tensile strength (UTS) of TiCP/Ti60 composites is 65 MPa higher than that of Ti60 alloy, following with decreased elongation. Ti60 alloy exhibit ductile fracture characteristics, while TiCP/Ti60 composite exhibit some complicated fracture characteristics including intergranular fracture, quasi-cleavage crack and local ductile rupture.
