Epithelial cells of the mammalian oviduct play an important role in reproductive and developmental events. Therefore, the objectives of the present study was to establish a method for isolation and culture of White yak (Bos grunniens) oviduct epithelial cells in vitro and to determine the effect of Fetal Calf Serum (FCS), Epidermal Growth Factor (EGF) and Transforming Growth Factor-β1 (TGF-β1) and their combination on White yak oviduct epithelial cells. The results showed and 100 ng mL-1 of EGFmTCM 199 with 10% FCS could significantly increased cell numbers of yak oviduct epithelial. Alone no addition, EGF (10 ng mL-1) and TGF-β1 (1 ng mL-1) had no effect on cell number. However, when included with 10% FCS in medium yak oviduct epithelial cell numbers significantly increased on day 6 of culture. The combination of FCS (10%) and EGF (10 ng mL-1) is the ideal formula to culture yak oviduct epithelial cells. Co-culture with yak oviduct epithelial cells the developmental rate of blastocyst was 8.9%. The results support the culture environment that oviduct epithelial cell may have direct effects on yak embryo developmental block in vitro production. © Medwell Journals, 2013. Source

A study was conducted to evaluate the potential of immature oocytes from the ovaries of yak. The effects of maturation times (23-30 h), hormones (FSH, LH, E2), sera (fetal calf serum, FCS, or oestrous cow serum, ECS) and culture system (co-culture with granulosa cell, GC, or with bovine oviduct epithelia cells, BOEC, tissue culture medium, TCM, 199 absence co-culture cell, and synthetic oviduct fluid with amino acids, SOFaa) on the in vitro development of in vitro matured and fertilised yak oocytes were examined. Immature oocytes surrounded with compacted cumulus cell were cultured for 23-30 h in TCM 199 supplemented with 10% FCS and hormones. In vitro fertilisation (IVF) was performed with frozen-thawed, caffeine and heparin treated spermatozoa from Datong yak. Oocytes were incubated with 1 × 106/ml spermatozoa for 16-18 h and then cultured in co-culture system with GC or BOEC and/or SOFaa for 7-8 days, respectively. Cleavage and development to blastocysts were recorded on days 2 and 8, respectively, after the start of culture. 27-28 h was superior to other culture times and addition hormones (FSH, LH, E2) were superior to absence either of them for oocyte maturation. The best maturation of oocyte in TCM 199 with 5.0 mg/L LH, 0.5 mg/L FSH, 1 mg/L E 2. FCS was superior to ECS for development to blastocysts. Co-culture with GC was superior to SOFaa and TCM 199 absence co-culture cell for cleavage, development to blastocysts. The results show that choice of culture conditions has marked effect on the development of in vitro matured and fertilised yak oocytes. The present results indicate that the co-culture with GCs is the most important factor for IVF to development into blastocysts of yak oocytes matured in vitro. © 2012 Taylor & Francis. Source

Fatty acid synthase (FASN) is a key enzyme in fatty acid anabolism that plays an important role in the fat deposit of eukaryotic cells. Therefore, in this study, we detected 2 novel single-nucleotide polymorphisms (SNPs) in the FASN gene in 313 adult individuals of Datong yak using polymerase chain reaction-single strand conformation polymorphism and DNA sequencing techniques. SNP g.5477C>T is located in intron 3 of FASN, and 3 genotypes, HH, HG, and GG, were detected in this mutation site. SNP g.16930T>A is located in exon 37 of FASN, and 2 genotypes, EE and EF, were detected in this site. Association analysis of these 2 SNPs with meat quality traits showed that in SNP g.5477C>T, yaks with the HH genotype and HG genotypehad significantly higher intramuscular fat content than individuals with the GG genotype (P < 0.01). In SNP g.16930T>A, yaks with the EE genotype also had significantly higher IMF content than individuals with the EF genotype (P < 0.01). The results indicate that FASN may be used as a candidate gene affecting intramuscular fat content in Datong yaks. © FUNPEC-RP. Source

The purpose of this study was to uncover differential patterns of mRNA expression for genes related to different development stages of yak (Bos grunniens) oocytes and preimplantation embiyos. Researchers identified differentially expressed bands by mRNA differential display technology and evaluated the relative expression level of differentially expressed genes in oocytes, two cells, four cells and eight cells embiyos and in morulae and blastulae. Researchers identified four differentially expressed bands of which one gene that was expressed during morula stage showed no homology to any other known genes in GenBank. Three other genes identified showed high identity with zinc finger MYND11 (ZMYND11), Nucleosome Assembly Protein 1-Like 1 (NAP1L1) and Ribosomal Protein L27A (RPL27A), respectively. The expression of ZMYND11 increased in mature oocytes, two cells and eight cells embiyos and in the blastula with no significant differences between these stages (p>0.05). NAP1L1 was expressed at a relatively low level in mature oocytes, two cells embiyos and in the blastula with a sharp increase in eight cells embiyos (p<0.05). The expression of RPL27A decreased during development from mature oocytes to two cells embiyos and then to eight cells stage embiyos and increased significantly in cells of the blastula (p<0.05). ZMYND11, RPL27A and NAPlLl, the mRNA expression levels of which showed differences in different development stages of yak oocytes and early embiyos might be related to normal development of early embiyos and may be involved in maintaining normal reproductive function in yak. © Medwell Journals, 2014.
