The progressive, ecologically pure technologies of isolation of sesquiterpene γ-lactones of plant origin were examined in this article. The original phytopreparations were developed on their basis. The pharmacoeconomic characteristics of the obtaining of substances of phytopreparations with application of supercritical CO2-extraction and method of preparative centrifugal partition chromatography in comparison with the classical methods of extraction and column chromatography were presented. © 2009 al-Farabi Kazakh National University. Two sesquiterpene lactones, grosheimin and cynaropicrin, present in the aerial part of the Chartolepis intermedia Boiss. plant can be obtained from the ethyl acetate solid extract of the dried plant. A large-scale purification processing 16 kg of dry plant is presented. The first liquid–liquid extraction step of the 16 kg produced 550 g of dry extract that were divided in two equal portions. A classical low-pressure chromatography method can produce from 275 g of dry extract 13.8 g of grosheimin and 52.3 g of cynaropicrin 95 % pure lactones in 110 h using large amounts of organic solvents and 8 kg of silica gel. Another purification method employing countercurrent chromatography, with a modern hydrostatic column, and reversed phase preparative liquid chromatography were used with the second 275 g dry extract portion. It could produce 17.9 g of 99.4 % pure grosheimin and 68 g of 98.7 % pure cynaropicrin lactones in 95 h, with better yields and minimized losses in chromatographic columns, and using smaller solvent volumes than the classical method. This review shows the data about isolated heteroatom-containing sesquiterpene lactones from various genera of Asteraceae family, natural occurence, isolation methods and their biological activity. Chlorinecontaining sesquiterpene lactones were isolated from genera of Artemisia L., Achillea L., Acroptilon Cass, Centaurea L., Chartolepis Cass, Eupatorium L., Jurinea Cass, Rhaponticum Adams, Saussurea DC. The greatest quantity of chlorine-containing sesquiterpene lactones were isolated from Centaurea genus. 25 new sesquiterpene lactones with chlorine atom in molecule were isolated and characterized. The sesquiterpene lactones, which in their structure contained a sulphur atom as a component of various alkoxyl-, thiohydroxygroups and acid residues, were isolated from genera of Eupatorium L., Petasites Mill, Helenium L., Acritopappus R.M. King and H. Rob., Arctium L., Saussurea DC. Nitrogen atom in the structure of natural sesquiterpene lactones is present in the form of amino groups as a part of ester groups as well as amino acids as Michael adducts. Nitrogen-containing sesquiterpene lactones were isolated from Acanthospermum Schrank, Saussurea DC. Isolated heteroatom-containing sesquiterpene lactones possess bactericidal, antifungal, cytotoxic, antiviral, virucidal, antiulcer and others activities. Another method for obtaining heteroatom-containing sesquiterpene lactones is a directed synthesis of chlorine-, fluorine-, bromine-, sulphur- and nitrogen-containing sesquiterpene lactones. On the basis of sesquiterpene lactone artemisinin known as an effective antimalarial preparation, we synthesized over 200 derivatives, among which fluorine-, chlorine-, bromine-, nitrogen-, sulphur- and azide-containing derivatives of artemisinin were obtained. © 2013 Al-Farabi Kazakh National University.
