Photodynamic therapy (PDT) is a promising cancer treatment modality that uses dye-sensitized photooxidation of biologic matter in target tissue. This study explored effects of the photosensitizer BCPD-17 during PDT for osteosarcoma. LM-8 osteosarcoma cells were treated with BCPD-17 and cell viability after laser irradiation was assessed in vitro with the 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide assay. The effects of BCPD-17 during PDT recurrence were then examined on tumor-bearing mice in vivo. BCPD-17 had dosedependent cytotoxic effects on LM-8 osteosarcoma cells after laser irradiation which also had energy-dependent effects on the cells. The rate of local recurrence was reduced when marginal resection of mice tumors was followed by BCPD-17-mediated PDT. Our results indicated BCPD-17-mediated PDT in combination with marginal resection of tumors is a potentially new effective treatment for osteosarcoma.

Aim: To assess the effect and safety of lanthanum carbonate (LC) for hypophosphatemia in patients with end-stage renal disease (ESRD). Methods: According to the collaborative review group search strategy, we searched MEDLINE (1996 to 2012.12); EBCO (1996 to 2012.12), and CNKI. We searched Chinese journals by hand. We conducted quality assessment and data extraction by two independent investigators. Meta-analysis was conducted by RevMan 5.0. Results were expressed as OR with 95% confidence interval for dichotomous outcomes and WMD with 95% confidence interval for continuous outcomes. Result: We identified 16 reports which might meet the inclusion criteria for our review. The meta-analysis showed that LC was superior to placebo in treating hypophosphatemia of end-stage renal disease patients (OR=5.46, 95% CI: 2.37 to 2.61, p<0.005) and as efficient as conventional therapies (WMD=-0.06, 95% CI: -0.27 to 0.15, p=0.57). The incidence of all adverse events was similar between LC- and placebo-treated patients (OR=1.16, 95% CI: 0.79 to 1.68, p=0.45). Conclusion: Lanthanum carbonate is well effective and tolerated in treating hyperphosphatemia of ESRD patients. Lanthanum carbonate is not likely to cause hypercalcemia compared to calcium-based phosphate binders. © 2013 Informa Healthcare USA, Inc.

Luteolin is a flavone in medicinal plants as well as some vegetables and spices. It is a natural anti-oxidant with less pro-oxidant potential but apparently with a better safety profile. The purpose of this study was to investigate the molecular mechanism of luteolin-mediated apoptosis of MG-63 human osteosarcoma cells. MTT assay kit was employed to evaluate the effects of luteolin on MG-63 cells proliferation. Then, we performed Annexin V-FITC/PI to analyze the apoptotic rate of the cells. Furthermore, the inhibitory effects of luteolin on the expressions of BCL-2, BAX, Caspase-3 and Survivin were detected by Western blotting. As expected, luteolin (0.5, 2.5, 12.5 µg/mL) inhibited the growth of MG-63 cells by inhibiting cell proliferation and inducing cell apoptosis. Western blotting demonstrated that luteolin (0.5, 2.5, 12.5 µg/mL) inhibited the expressions of BCL-2, Caspase-3 and Survivin, and promoted the expression of BAX in MG-63 cells with a concentration dependent way. Luteolin can inhibit osteosarcoma cell proliferation and induce apoptosis effectively in a dose dependent manner through down-regulating the expression of BCL-2, Caspase-3 and Survivin proteins levels and up-regulating the expression of BAX protein level. These findings indicated that luteolin may be used as a novel herbal medicine for the treatment of osteosarcoma. © 2015 by School of Pharmacy

Epithelial ovarian cancer is the most common tumor of ovary. PTK7 plays an important role in tumors. Till now, there is no report about the role of PTK7 in ovarian epithelial neoplasms. This study aims to investigate the expression of PTK7 protein in epithelial ovarian tumors, and its relationship with clinical pathological characteristics. In this study, immunohistochemical staining was used to detect the expression of PTK7 protein in 14 samples of normal fallopian tube epithelium and 204 cases of epithelial ovarian tumor. The relationship between the expression of PTK7 and pathological indicators was statistically analyzed. Kaplan-Meier survival function was used to analyze the prognosis. The expression of PTK7 was found in 92.86% (13/14) of normal fallopian tube epithelium and 45.10% (92/204) of epithelial ovarian tumor tissues. The expression level of PTK7 was significantly decreased from the benign, the intermediate type, to malignant ovarian epithelial tumors (P < 0.001), and decreased from the normal control group to serous carcinomas (P < 0.001). The expression of PTK7 was significantly different in type I and type II epithelial ovarian carcinomas (P < 0.001). PTK7 protein expression was associated with clinical stages (P = 0.038) and metastases (P = 0.038) in ovarian borderline serous tumors. PTK7 protein expression was associated with clinical stages (P = 0.011), WHO grading (P = 0.004), and MDACC grading (P < 0.001) in ovarian serous carcinomas. The survival analysis showed that patients with negative expression of PTK7 protein had a poorer outcome than those with positive expression (P = 0.017). These results indicate that PTK7 protein may be a tumor suppressor and a potential prognostic marker in ovarian serous carcinomas.

Aseptic loosening secondary to periprosthetic inflammatory osteolysis results from the biological response to wear particles and is a leading cause of arthroplasty failure. The origin of this inflammatory response remains unclear. We aim to validate the definite link between endoplasmic reticulum (ER) stress and particle-induced inflammatory signaling pathways in periprosthetic osteolysis. We examine the histopathologic changes of osteolysis and the expression of specific biomarkers for ER-stress-mediated inflammatory signaling pathways (IRE1, GRP78/Bip, c-Fos, NF-B, ROS and Ca(2+)). Moreover, pro-inflammatory cytokines (TNF-, IL-1 and IL-6) and osteoclastogenic molecules (VEGF, OPG, RANKL and M-CSF) were assessed in clinical interface membranes and murine periosteum tissues. We found wear particles to be capable of inducing ER stress in macrophages within clinical osteolytic interface membranes and murine osteolytic periosteum tissues and to be associated with the inflammatory response and osteoclastogenesis. Blocking ER stress with sodium 4-phenylbutyrate (4-PBA) results in a dramatic amelioration of particle-induced osteolysis and a significant reduction of ER-stress intensity. Simultaneously, this ER-stress blocker also lessens inflammatory cell infiltration, diminishes the capability of osteoclastogenesis and reduces the inflammatory response by lowering IRE1, GRP78/Bip, c-Fos, NF-B, ROS and Ca(2+) levels. Thus, ER stress plays an important role in particle-induced inflammatory osteolysis and osteoclastogenic reactions. The pharmacological targeting of ER-stress-mediated inflammatory signaling pathways might be an appealing approach for alleviating or preventing particle-induced osteolysis in at-risk patients.

Regenerating gene family member 4 (REG4), a secreted protein, is overexpressed in several cancers, including gastric cancer. The present study was undertaken to determine the roles of REG4 in the growth of gastric cancer in the nude mice and in the proliferation and migration in human gastric cancer cell line and its downstream signaling pathway. Gastric cancer models were elicited by intraperitoneally injecting MKN45 human gastric cancer cells and the tumor size was measured every other day. The expressions of REG4 mRNA and protein were increased in the gastric cancer tissues from gastric cancer patients. REG4 increased the gastric tumor weight and size in the nude mice, and promoted the proliferation and migration of gastric cancer cells MKN45. Adeno-associated viral (AAV)-mediated knockdown of REG4 decreased the gastric tumor weight and size in the nude mice, and suppressed the proliferation and migration of MKN45 cells. REG4 increased the expression of phosphorylated protein kinase B (Akt). Triciribine hydrate (TCN), the inhibitor of Akt, decreased the gastric tumor weight and size in the nude mice and abolished REG4-induced weight and size increase of the tumor. TCN also inhibited proliferation and migration and abolished REG4-induced proliferation and migration increase of human gastric cell line MKN45. These results indicate that REG4 promotes the growth, proliferation and migration of gastric cancer through Akt pathway. © 2014, International Journal of Clinical and Experimental Medicine. Existing in Ranunculaceae Aconitum and tomato, with the chemical name 1‑phydroxybenzyl‑1,2,3,4‑tetrahydroisoquinoline, higenamine is widely distributed in China. Higenamine's anti‑inflammatory, antioxidant and anti‑apoptotic effects have been identified in previous studies. The present study attempted to determine the protective effect of higenamine against collagen‑induced arthritis through heme oxygenase‑1 (HO‑1) and PI3K/Akt/Nrf‑2 signaling pathways. A type II collagen (CII)‑induced arthritis (CIA) model was established and clinical arthritis scores were used to appraise the curative effect of higenamine. Inflammatory reactions, oxidative damage and caspase‑3/9 activation were detected using specific ELISA kits. In addition, western blotting was used to evaluate the expression of HO‑1, Akt and Nrf‑2 protein in CII‑induced CIA mice. In CII‑induced CIA mice, the clinical arthritis scores, inflammatory reactions, oxidation damage and caspase‑3/9 activation were increased and activated. The results demonstrated that treatment with higenamine significantly reduced the elevation of clinical arthritis scores (P<0.01), and suppressed the promotion of inflammatory reactions, oxidation damage and caspase‑3/9 activation. Furthermore, higenamine significantly increased HO‑1 protein expression (P<0.01) and upregulated the PI3K/Akt/Nrf‑2 signal pathway in CII‑induced CIA mice. Collectively, it is concluded that higenamine protects against CII‑induced CIA through the induction of HO‑1 and the upregulation of the PI3K/Akt/Nrf‑2 signaling pathway. In conclusion, higenamine may be a beneficial drug for protecting against CIA. © 2016, Spandidos Publications. All rights reserved.

This study aimed to detect micrometastatic tumor cells in the lymph nodes of patients with pN0 lung adenocarcinoma using a combination of thyroid transcription factor-1 (TTF-1) expression and cytokeratin 7 (CK7) expression and to investigate the association of lymph node micrometastasis with the clinicopathological characteristics of patients with lung adenocarcinoma. A total of 54 patients with pN0 lung adenocarcinoma and whose primary tumors were positive for both TTF-1 and CK7 expression were included in this study. In total, 893 lymph nodes were obtained from these 54 patients and were analyzed for micrometastasis by immunohistochemical staining with anti-CK7 and anti-TTF-1 antibodies. CK7- and TTF-1-positive cells were found in the lymph nodes of 9 (16.7%) of 54 patients, and 21 (2.4%) of 893 lymph nodes exhibited positivity for these factors. No cells positive for both CK7 and TTF-1 were detected in the 5 lymph nodes obtained from patients with benign lung tumors. Lymph node micrometastasis was found to be associated with the differentiation grade and primary tumor position (P < 0.05). The detection of lymph node micrometastasis by a combination of CK7 and TTF-1 immunohistochemical staining provides a more accurate assessment of tumor staging for pN0 lung adenocarcinoma.

The nucleus accumbens (NAcc) has been proven to be associated with drug and food craving. NAcc ablative neurosurgery has been suggested to modulate the balance of the brain reward system and thus alleviate drug dependence in patients. It has been hypothesized that it would also alleviate food craving in patients as well as altering their nutritional status.This study aimed to estimate the effect of NAcc neurosurgery on drug craving and nutritional status in patients with drug dependence at 5 years postoperatively.The study included 100 patients with NAcc surgery and 92 patients without surgery. Body mass index (BMI) and body fat percentage (BF%) were examined to assess nutritional status, and questionnaires were administered to assess drug craving.Compared with the nonsurgery group and the relapse patients from the surgery group, the nonrelapse patients from the surgery group had higher BMI and BF% but lower drug craving. There were no significant differences between the nonsurgery group and the relapse patients in BMI, but the relapse patients had higher drug craving than the nonsurgery group.Long-term follow-up suggested that NAcc ablative neurosurgery would alleviate drug craving and yield a better nutritional status if individuals sustained abstinence. It would increase drug craving but would not ruin the nutritional status of patients even when individuals relapsed postoperatively.
