Objective: To observe the inactivation effect of β-propiolactone on hepatitis A virus (HAV). Methods: HAV was added with 1/4 000 β-propiolactone and 1/4 000 formaldehyde respectively. The inactivation effect was verified by determination of virus titer, and the time for inactivation was optimized. The influence of the two inactivating agents on titer and immunogenicity of HAV antigen, as well as their toxicities to mice and guinea pigs, were evaluated. Results: No HAV was detected after inactivation with β-propiolactone for 10 h or with formaldehyde for 3 d. HAV was completely inactivated after treatment with β-propiolactone for 24 h, indicating that β-propiolactone was superior to formaldehyde in inactivation of HAV. The HAV antigen titers after inactivation with β-propiolactone and formaldehyde for various times showed no significant difference, which were 1:640 EU/0.1 ml. The positive rate of anti-HAV in mice 28 d after immunization was 100%. Conclusion: β-propiolactone inactivated HAV by direct action on DNA while maintained the immunogenicity. Source
