Objective: The aim of this study was to compare spontaneous and induced rates in mice of both sexes of lines BALB/c, NMRI, OF-1 and C57BL/6/ cenp, by testing chromosomal aberrations in bone marrow cells. Materials and methods: For this purpose, the most efficient line was determined, based on the significant appearance of lower spontaneous rates and higher induced rates to mutagen substances such as cyclophosphamide (CP). Results: The result obtained was that line Balb/c, in both sexes, differs significantly from the other lines tested. The lesser total number of cells with structural aberrations was found in this line and a greater number of induction of chromosomal aberrations when CP was used as a mutagen. Conclusions: This work will give way to the use of the best line of mice as bio model in this genotoxicity test, and will also give this cytogenetic technique greater veracity and robustness.

The aim of this study was to characterize the symptomatology and anatomopathological lesions caused by 5 clinical isolated Leptospira spp. from Nicaragua in a Mesocricetus auratus biomodel. Materials and methods. 50 hamsters were inoculated via i.p with 1mL of the culture of each strain in exponential phase having a cellular concentration of 7.5 x 106 leptospira/mL, (10 animals per strain). Signs of the disease, mortality during 14 days, and macroscopic and microscopic anatomopathological lesions by haematoxylin-eosin and Warthyn Starryn stain technique were evaluated. Results. All the strains presented high mortality, showing clinical lesions of the experimental infection. Death to 100% of the animals was caused between the third and tenth day post-infection. In the anatomopathologic study, the strains of the Ballum and Pomona serogroup produced haemorrhaging specifically in the kidney and lungs. The animals manifested hepatic and renal congestion, while the renal haemorrhage was observed with more frequency in the strain of the Pomona serogroup, differing from the other strains, which presented this lesion less frequently. Conclusions. This work allowed a better characterization of these strains in order to use them as future vaccine candidates for future Leptospirosis epidemics in Nicaragua.

Introduction: the dry aqueous extract from the decoction of the bark of some selected Mangifera indica L. varieties is the active principle of some Vimang® formulations. This natural extract was reported to have antioxidant, antiinflammatory, antimicrobial, spasmolytic, analgesic and antipyretic properties. The main component is mangiferin, a xanthone c-glycosilated, with several reported biological actions. For this reason, mangiferin was chosen as chemical marker for the study of the chemical stability, and the quality control of the different formulations of mango tree bark, commercially used as antioxidant nutritional supplement to treat several chronic diseases. Objective: to study the influence of the suggested storage conditions for Mangifera indica L. bark on the physical, chemical and microbiological properties and the content of mangiferina in the vegeral raw material. Methods: the fresh bark material of Mangifera indica L was grounded and stored in polyethylene bags during 70 days under two storage conditions at 22 ± 1°C and 55 ± 5 % relative humidity. Presence and total counting of micro- organisms was determined in the bark, whereas the pH value and the content of total solids and mangiferin were identified in the aqueous extract. The mangiferin was analyzed by high performance liquid chromatography. Results: there were observed rapid microbial growth and presence of unwanted micro-organisms from the beginning of the study. After 31 day-storage under specified temperature and humidity conditions, a pathogenic micro-organism called Pseudomonas aeruginosa emerged. The content of total solids and mangiferin in the aqueous extract did not significantly change during the study, which indicated the lack of chemical degradation in these components. The pH value of the aqueous extract decreased as a result of the emergence of fungi in the bark. Conclusions: it is recommended to store the fresh bark for 30 days as maximum, in order to prevent deterioration and contamination of the vegetal material caused by pathogenic micro-organisms.

Cerebral ischemia is a major cause of death, for this reason animal models of cerebral ischemia are widely used to study both the pathophysiology of ischemic phenomenon and the evaluation of possible therapeutic agents with protective or regenerative properties. The objectives of this study were to examine the presence of neuronal damage in different brain areas following the ischemic event, and assess consequences of such activities on the processes of memory and learning. The study group included an experimental group ischemic animals (30 rats with permanent bilateral occlusion of the carotids), and a control group. Was evaluated gene expression and inflammatory ischemic by qPCR techniques 24h post injury, brain tissue morphology in areas of cortex, striatum and hippocampus seven days post injury and processes of memory and learning, 12 days post injury. The morphological studies showed that the procedure induces death of cell populations in cortex, striatum and hippocampus, ischemia modified gfap gene expression and ho, il-6, il-17 and ifn-γ, which can be used as a marker of early ischemic process. Additionally, the ischemic injury caused spatial memory decline. This characterization gives us an experimental model to develop future studies on the pathophysiology of ischemic events and assessing therapeutic strategies. © 2015, Universidad Nacional de Colombia. Whole cell Leptospirosis vaccines have been developed and used from the 20's of last century up today, most of them are not adjuvated and do not confer a long-lasting immunity. These vaccines are mainly against different serogroups of Leptospira interrogans contained in the preparation. Though several attempts to obtain a vaccinal formulation more effective, purer, with wider spectrum and longer protection than the bacterines of inactivated whole cells have been made no vaccine with these characteristics have been registered yet. This paper deals with the obtainment of outer membrane antigens from a Cuban autochthonous strain (Strain 87, L. interrogans serovar Canicola), by a modification of the technology for the production of outer membrane vesicles, patented by researchers from Finlay institute These antigens with nanoproteoliposomic structure were formulated/adjuvated by different strategies, obtaining five preparations with cochlear structures which are nanoparticles of approximately 100 to 150 nm of lenght and from 15 to 30 nm of diameter. Two doses of the immunogens are inoculated in the biomodel Mesocrisetus aureatus, with an interval of six weeks. The challenge was carried out with 100.000 DL 50. Results showed that the new vaccinal formulations confer protection against the homologous challenge and were able to eliminate the carrier status. This, together with the robutness of the preparation method, the higher level of purity, compared to the bacterines, and the no need of aluminium hydroxide make the formulations an alternative of interest for continuing their development.

Objective: This article has aimed to compare Balb/c, NMRI, OF-1 and C57BL/6/cenp mice as a biomodel in the assay of the sperm head basal cavity morphology, keeping in mind the frequency of anomalous forms of the basal head and cyclophosphamide-induced (CF) abnormalities. Materials and methods: Twenty animals/group/mice lines were used, administered for 35 days. A negative control group (not administered), two substance-vehicle controls and a positive control administered with 50. mg/kg FG intraperitoneally for five consecutive days were used. Results and discussion: The best experimental biomodel was with the Balb/c mice, with significant difference with the results obtained in the other lines, keeping in mind the spontaneous and induced values in the sperm concentration and frequency of anomalous epididymal sperms. This study will make it possible to use this mice species more efficiently in genotoxic and preclinical toxicology fertility evaluation of drugs, vaccines and other products. © 2011 Sociedad Española de Andrología.

The aim of this study was to evaluate the sensibility of different murine lines to the cyclophosphamide (CF) measure through the micronucleus assay in bone marrow cells. Being used mice of both sexes of the Balb/c, NMRI, OF-1 and C57BL/6/cenp lines. The use of the best line was suggested in this assay on the base of significant appearance of low indexes spontaneous and high induced indexes to the CF. It was obtained as a result that the Balb/c line in both sexes differs significantly of the other evaluated lines. In this line the lowest results in cytotoxicity index (relationship among young erythrocytes/mature erythrocytes) and genotoxicity indexes (erythrocytes percent in bone marrow with micronucleus) were observed, at the same time of responding from an acceptable form to the CF. Also the line C57/BL6/cenp was the less efficient and more sensitive to the mutagen, being obtained the highest spontaneous and induced results. These results allow to suggest that the Balb/c mice, is the most appropriate line to use as biomodel in the micronucleus assay, when it is used the CF as positive control.
