Poria cocos F.A.Wolf is a Chinese traditional medicine used to treat chronic gastritis, edema, nephrosis, gastric atony, and acute gastroenteric catarrh. Polysaccharides are the main active component of P. cocos. We obtained polysaccharides PCP-1, PCP-2, and PCP-3 from the degradation of P. cocos polysaccharides (PCP) with different concentrations of H2O 2 solution. Molecular weights were determined by high performance size exclusion chromatography. HPLC analysis of monosaccharide composition confirmed that PCP-1, PCP-2, and PCP-3 are heteropolysaccharides composed of glucose and arabinose. IR spectra indicated obvious characteristic peaks of polysaccharides. The antioxidant activities of these polysaccharides were evaluated by established in vitro systems, including scavenging activity of hydroxyl radicals, ABTS radicals, and ferrous ions. The degradation polysaccharides exhibited obvious and concentration-dependent antioxidant properties. In addition, DNA binding analysis showed that PCP-1 had a stronger capacity than other polysaccharides to interact with DNA. However, each polysaccharide had a certain capacity for DNA damage protection. © 2014 Elsevier Ltd. All rights reserved.

Objective: To develop a HPLC method for the determination of synephrine and arecoline contents in Xiao'er Xiaoji Zhike oral liquid. Method: The analysis was performed on a Symmetry C18 column (4. 6 mm x 250 mm, 5 μm) eluted with acetonitrile-methanol-0. 1% acetic acid solution (containing potassium dihydrogen phosphate 0. 6 g and SDS 1. 0 g per 1 000 mL) (15: 30: 55)as mobile phase. The detection wavelength was set at 215 nm. Result: The synephrine and arecoline concentrations had good linear relationship between 0. 281 5-2. 815 and 0. 040 16-0. 401 6 μg (r>0. 999 7), and the average recoveries of the two compounds were 97. 1% (RSD 1. 4%) and 100% (RSD 1. 3%), respectively. Conclusion: The method is simple, accurate and sensitive.

To explore the phylogenetic relationships among medicinal leeches and their closely related species by comparing their ITS gene fragments. 8 different leech species, including Whitmania pigra, Whitmania acranulata, Whitmania laevis, Hirudo nipponia, Poecilobdella manillensis, Poecilobdella javanica, Poecilobdella hubeiensis and Erpobdella octoculata were collected as the materials for study. After the total DNA of all samples was extracted, amplified by PCR and cloned, their ITS sequence, AT content, variable sites and evolutionary relationships were analyzed. The length of ITS was 948-981 bp with 210 variable sites and 43 inserts on/deletion. The average G + C content was high up to 56.0%-57.8%. The aligned ITS sequence demonstrated that their ITS genes were relatively conserved in species. Data analysis showed that the relationship between E. octoculata and P. javanica was closer than others, their genetic distance was only 0.007. Through the phylogenetic tree analysis, three species of Haemopidae and four species of Hirudinidae were obviously clustered into two branches. The results support the taxonomic status of Haemopidae and Hirudinidae, and provide sufficient evidences that Erpobdella octoculata and Poecilobdella javanica should not be assigned to two genera, of which the taxonomic system should be further researched.

In order to discover anticancer agents from natural sources, an ethanol-soluble extract of the root bark of Juglans cathayensis was investigated and showed cytotoxic effects against various human cancer cell lines. A subsequent phytochemical study on the EtOAc-soluble fraction determined 2-methoxyjuglone (1) as one of the main active constituents. Compound 1 was shown to be cytotoxic against HepG2 cells. Morphological features of apoptosis were observed in 1-treated HepG2 cells, including cell shrinkage, membrane blebbing, nuclear condensation, and apoptotic body formation. Cell cycle analysis with propidium iodide staining showed that 1 induced cell cycle arrest at the S phase in HepG2 cells. Flow cytometric analysis with annexin V and propidium iodide staining demonstrated that 1 induced HepG2 cell apoptotic events in a dose-dependent manner (0-8 μg/mL). Western blot analysis of apoptosis-related proteins revealed that 1 induces HepG2 cell apoptosis through mitochondrial cytochrome c-dependent activation of the caspase-9 and caspase-3 cascade pathway (intrinsic pathway). An in vivo experiment using tumor-bearing mice showed that treatment with 1 at 0.5 and 1.0 mg/kg per day decreased the tumor mass by 56% and 67%, respectively. © 2013 The American Chemical Society and American Society of Pharmacognosy.

Component control is a key step of the quality control process in gentamicin (GM) production. The near infrared (NIR) method allows the rapid analysis of various components for the on-line control in the production process. In this study, we selected specific NIR spectral regions and eliminated solvent interference in constructing the predictive models of GM content in aqueous solution. We found that two factors could lead to better NIR predictions: (i) the combined use of specific NIR spectral regions related to both structural characteristics and content; and (ii) the analysis of sample spectra based on solvent reference spectra to identify and extract spectral information of testing components. We constructed predictive models for total GM C components, GM C1a (C1a), micronomicin (MCR) and sisomicin (SISO) in aqueous solution, respectively. These models can be used for the quick content analysis of different components in various types of GM injection samples. © 2010 Elsevier B.V. All rights reserved.

Response surface method (RSM) was employed to optimize the extraction conditions of polysaccharides from Rhizoma Panacis Majoris (the rhizomes of Panax japonicus C. A. Mey. var. major (Burk.) C. Y. Wu et K. M. Feng) (RPMP), a well-known Chinese traditional medicine. In order to obtain the optimal processing parameters, a three-variable Box-Behnken designs (BBD) were applied for experimental designs. RSM analysis indicated the good correspondence between experimental and predicted values, the optimal conditions for the yield of polysaccharides were as follows: the ultrasound time is 31.15 min, extraction temperature is 92.50 C, and the ratio of water to raw material is 40 mL/g. The maximum value (13.87 0.16%) of the yield of polysaccharides was obtained under these optimal conditions. The molecular weight (MW) was determined to be 1.48 10(5)(0.39%)Da by HPSEC-MALLS-RID chromatography system. FT-IR spectra demonstrated obvious characteristic peaks of polysaccharides. The antioxidant activities of RPMP were investigated including scavenging activity of hydrogen radicals, ABTS radicals, and free radicals of superoxide anion in vitro, and the results exhibited that RPMP had a good potential for antioxidant.

Snake bile, a precious traditional Chinese medicine (TCM), was used as the major ingredient of some Chinese patent drugs, such as Shedan Chuanbei powder and Shedan Chenpi powder for hundred years. However, there is still requirement for the comprehensive and definite composition of bile acids in snake bile. In order to rapidly identify the bile acids constituents in snake bile to avoid the adulteration, ultrahigh-performance liquid chromatography with electrospray ionization quadrupole time-of-flight tandem mass spectrometry (UPLC/ESI-QTOF-MS/MS) has been applied to conduct a qualitative analysis on snake bile acids. ESI ion source was used for mass spectra, and data were collected in both positive and negative ion mode. 16 kinds of reference standards, attributed to free bile acids, taurine- and glycine- conjugated bile acids, were detected and their MS behaviors were summarized. Objective: To establish an assay with high sensitivity and specificity for identification and quantification of protein components in commercial thrombin lyophilized powder with UPLC/Q-TOF-MS system. Methods: Lyophilized thrombin samples were digested with trypsin and then separated and analyzed with ultra-performance liquid instrument coupled with a quadrupole time-of-flight mass spectrometer. Both peptide parent ions and fragments were collected simultaneously by alternation between low and elevated energy of the collision cell(MSE methodology). The MSE data was processed by ProteinLynx Global Server (PLGS 2.4), to identify and quantify protein components in the samples. Quantization was achieved with a proven label-free quantification technique("Hi3" method). Results: Twenty-one porcine protein groups and human albumin were identified from the pig-blood-source thrombin lyophilized powders, with thrombin content ranging from 1.9% to 43.7% in different manufacturers' products. Seventeen bovine protein groups were identified from the bovine-blood-source thrombin lyophilized powders, with thrombin content ranging from 49.4% to 77.6%. Conclusion: This assay provides valuable information to understand the origins of the samples, protein components and percentages of lyophilized thrombin powders, as well as some process-related information. The assay potentially can be used as a generic method for the quality research of similar biologics.
