The concentration of the endogenous dipeptide carnosine reaches tens of millimoles in human muscle tissue. Vast data on various carnosine functions have accumulated over more than 100 years of research, the anti-aging effect being thought the most important. The effect of carnosine was studied in cultured cells. Apart from the well-known effects of increasing the Hayflick limit and causing morphological rejuvenation of cells, carnosine was found to stimulate cell divisions in the colony formation assay and to inhibit the cell transition to quiescence. A transcriptome analysis showed that carnosine-induced changes are mainly related to the positive regulation of the cell cycle at all of the levels from the onset of DNA synthesis to chromosome condensation. It was assumed that the cell cycle-stimulating potential underlies the rejuvenating effect of carnosine and that high carnosine concentrations in muscle tissue are necessary for the recovery (regeneration) of muscles after intense exercise apart from their role as a physiological buffer. © Pleiades Publishing, Inc., 2014.

AimsThis study aimed to determine the influence of carotid artery surgery on ocular functions and ocular blood flow in patients with ocular ischaemic syndrome (OIS) in the late postoperative period.MethodsOne hundred and eighty patients with OIS were operated on; 104 of them suffered from acute forms of the ischaemic disease and 76 patients had chronic forms of ocular ischaemia. Before surgery and in the course of 6 months and 12 months afterwards, all the patients were examined. Visual acuity, electrophysiological investigations (the threshold of electrical sensitivity (TES) and the level of liability of optic nerve (LON)) and blood flow in orbital vessels were assessed.ResultsAfter surgery visual acuity increased in patients with the acute forms of OIS (P0.05). TES and LON also improved (P0.01). Mean indices of blood-flow velocities in the ophthalmic artery, the central retinal artery and the posterior ciliary arteries increased at 6 and 12 months after surgery (P0.05). There was ocular blood flow acceleration and decrease of vasoresistance in orbital arteries in both groups of patients.ConclusionsCarotid artery surgery effectively improved ocular blood flow in patients with acute and chronic forms of OIS in the late post operative period. © 2012 Macmillan Publishers Limited All rights reserved.

The study investigated the association of the ABCB1 C3435T polymorphism gene with the risk for uveal melanoma (UM). Genotyping was carried out using the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) analysis. All the tumors under study were histologically verified. The patients with UM were found to have a high frequency of predisposing alleles and genotypes of this polymorphism in the patients with UM. The CC genotype of the ABCB1 C3435T polymorphism was shown to be associated with the increased risk of UM (OR=8.5;p=7·10-4). A significant correlation was first found between the frequency of the CC genotype of the ABCB1 C3435T polymorphism and the height of a tumor and degree of its pigmentation (p=0.0391 and p=0.0381, respectively, Fisher's test). A marginal association was first shown between the CC genotype of the ABCB1 C3435T polymorphism and the vascular state and histological type.

Deepithelialization of the cornea (diameter 7 mm) was performed in rabbits and the rate of defect epithelialization was evaluated. Conjunctival ischemia was modeled by application of graduated alkaline burn. Antioxidant activity and content of nitrates and nitrites was measured in the tear fluid before and after burn by chemiluminescence and Griess methods, respectively. Emoxypin and mexidol promoted healing of corneal epithelial defect at the stage of epitheliocyte migration to the defect area and at the stage of their proliferation, respectively. After treatment with both agents, the area of conjunctival ischemia decreased more rapidly, but the efficiency of mexidol was higher. Antioxidant activity and content of products of NO metabolism in tear fluid decreased after burn. Mexidol, but not emoxypin, increased these parameters. Thus, mexidol and emoxypin have different effects on corneal epithelialization and conjunctival ischemia and effects of mexidol are more pronounced. © 2015, Springer Science+Business Media New York.

Human recombinant protein Wnt7a (hrWnt7a) inhibits cell proliferative activity and triggers cell polarization. Although cell polarization process was maintained only over a short time, probably via microenvironmental stimuli, hrWnt7a is involved in the transformation of the retinal pigment epithelium. Analysis of Wnt signaling pathway and its regulation will help to understand the processes in retinal pigment epithelial cells under pathological conditions, which can be useful in developing new generation drugs. © 2015, Springer Science+Business Media New York.

Three different morphological types were identified. The review presents the most common methods of modeling of retinal ischemia in vitro (chemical ischemia with iodoacetic acid, incubation of the retinal pigment epithelium cells with oligomycin, deprivation of oxygen and glucose) and in vivo (a model with increased intraocular pressure, cerebral artery occlusion, chronic ligation of the carotid arteries, photocoagulation of the retinal vessels, occlusion of the central retinal artery, endothelin-1 administration). Modeling ischemic injury in rats is the most frequently used method in studies, because the blood supply of their eyes is similar to blood flow in the human eyes. Each method has its own advantages and disadvantages. Application of methods depends on the purpose of the experimental study. Currently model of ocular ischemic disease can be obtained easily by injecting vasoconstrictive drug endothelin-1. It is the most widely used method of high intraocular pressure induced ocular ischemic damage similar to glaucoma, occlusion of central retinal artery or ophthalmic artery in human. The development of experimental models of ocular ischemic diseases and detailed investigation of mechanisms of impairment of microcirculation are useful for improve the efficiency of diagnostic and treatment of ischemic diseases of retina and optic nerve. © 2014, Izdatel'stvo Meditsina. To analyze the associations of the rs3087243 CTLA4 polymorphism and the rs1800629 TNF polymorphism with endocrine ophthalmopathy (EOP) in ethnic Russian patients with Graves disease (GD).The case-control study enrolled 205 patients with GD. The distribution of alleles and genotypes of the rs3087243 CTLA4 and rs1800629 TNF polymorphisms was studied in 141 patients with GD and EOP (a GD+EOP group) and 64 patients with GD without EOP (a GD-EOP group). The polymorphic alleles were identified by polymerase chain reaction-restriction fragment length analysis.The patients with GD in their history and EOP had significantly higher frequencies of A allele and AA genotype and a lower proportion of G allele and GG genotype of the rs3087243 CTLA4 polymorphism. Comparative analysis revealed no significant differences in the frequency of the alleles and genotypes of the rs1800629 TNF polymorphism.The rs3087243 CTLA4 polymorphism is associated with the risk of EOP in ethnic Russian patients with GD.
