Shoot segments with axillary buds from young branches of Vaccinium ashei 'Powderblue' were culturedin vitro. The axillary buds grew out and formed aseptic plantlets. Then young leaves of aseptic plantlets were used as explants to induce regenerating multiple shoots directly. The multiple shoots became thicker by subculturing and formed the whole plantlets finally. The results show that the optimal regeneration of multiple shoots was achieved on WPM medium supplemented with 0.2 mg·L-1 thidiazuron (TDZ) and 0.1 mg·L-1 1-naphthylacetic acid (NAA). The optimal subculture medium was WPM medium supplemented with 0.5 mg·L-1 zeatin (ZT) and 0.05 mg·L-1 NAA. The optimal rooting medium was 1/2WPM medium supplemented with 0.8 mg·L-1 3-indolebutyric acid (IBA) and 0.1 g·L-1 activated carbon (AC). The tissue culture technology was studied in this research by using semi-lignified shoots from female individual 'Oosterwijk' (Ilex verticillata). The results showed that bud formation on the nodal explants was induced on the WPM medium supplementing with 6-BA 0.50 mg·L-1 and NAA 0.05 mg·L-1; the best medium for proliferation was WPM supplemented with 6-BA 1.00 mg·L-1, NAA 0.10 mg·L-1 and sucrose 20 g·L-1, the proliferation coefficient was 6.1 after subculturing for 25 d; the optimal rooting medium was 1/2WPM supplemented with IBA 0.20 mg·L-1 and NAA 0.40 mg·L-1, with rooting rate up to 95.2% and 5.7 roots per shoot. After rooting culture for 30 days and acclimatization in greenhouse for 10 days, the shoots were transferred to the substrate of perlite and peaty soil (1:1, V/V) and the survival rate was 98%. These results can provide the technical basis for scale production of plantlets of I. verticillata 'Oosterwijk'.
