The snakehead fish Channa argus is an important food fish in China. We identified six microsatellite loci for C. argus. These six microsatellite loci and four other microsatellite markers were used to analyze genetic diversity in four cultured populations of C. argus (SD, JX, HN, and ZJ) and determine their relationships. A total of 154 alleles were detected at the 10 microsatellite loci. The average expected and observed heterozygosities varied from 0.70-0.84 and 0.69-0.83, respectively, and polymorphism information content ranged between 0.66 and 0.82 in the four populations, indicating high genetic diversity. Population JX deviated from mutation-drift equilibrium and may have experienced a recent bottleneck. Analysis of pairwise genetic differentiation revealed FST values that ranged from 0.028 to 0.100, which indicates a moderate level of genetic differentiation. The largest distances were observedbetween populations HN and SD, whereas the smallest distances were obtained between populations HN and JX. Genetic clustering analysis demonstrated that the ZJ and HN populations probably share the same origin. This information about the genetic diversity within each of the four populations, and their genetic relationships will be useful for future genetic improvement of C. argus through selective breeding. © FUNPEC-RP. Mitochondrial DNA

We sequenced and characterized the complete mitochondrial genome of Channa argus, Channa maculata and their hybrid [C. maculata (♀) and C. argus (♂)]. All the three mitochondrial genomes contained the typical complement of 13 protein-coding genes, 22 transfer RNAs (tRNAs), 2 ribosomal RNAs (rRNAs) and 1 control region. The entire mitochondrial DNA (mtDNA) molecule of C. maculata was 16,559 bp long while the complete mtDNA molecule of C. argus and hybrid snakehead fish was 16,558 bp long. This is the first report on the complete mitogenome sequence of C. maculata and hybrid snakehead fish. © 2013 Informa UK, Ltd. Source

The Chinese soft shelled turtle (Pelodiscus sinensis) has been used as food and medicine for thousands of years in China. In the last 25 years, some large-scale P. sinensis farms have been established. However, little is known about the genetic variations in farmed broodstocks although, they are important in selective breeding. Researchers genotyped 336 individuals from seven stocks using eight polymorphic microsatellites to study genetic diversity and population structure off. sinensis. In the seven stocks, a total of 120 alleles were detected at the eight loci. All seven stocks showed high allelic (A = 9.50-12.25; Ar = 9.25-12.50) and gene diversity (H E = 0.73-0.79). But certain degree of inbreeding (inbreeding index franged from 0.07-0.23) was detected in all seven stocks. Both AMOVA and F ST analysis revealed significant population structuring (F ST = 0.114, p<0.01). The information about genetic diversity and population structure of the seven stocks would supply a basis for future genetic improvement of P. sinensis through selective and cross breeding. © Medwell Journals, 2012.
