The complete mitochondrial (mt) genome sequences of Neobenedenia melleni were determined and compared with those of Benedenia seriolae and B. hoshinai. This circular genome comprises 13,270 bp and includes all 36 typical mt genes found in flatworms. Total AT content of N. melleni is 75.9 %. ATG is the most common start codon, while nad4L is initiated by GTG. All protein-coding genes are predicted to terminate with TAG and TAA. N. melleni has the trnR with a TCG anticodon, which is the same to B. seriolae but different from B. hoshinai (ACG). The mt gene arrangement of N. melleni is similar to that of B. seriolae and B. hoshinai with the exception of three translocations (trnF, trnT and trnG). The overlapped region between nad4L and nad4 was found in the N. melleni mt genome, which was also reported for the published Gyrodactylus species, but it was not found in those of B. seriolae and B. hoshinai, which are non-coding regions instead. The present study provides useful molecular characters for species or strain identification and systematic studies of this parasite. © 2014, Springer Science+Business Media Dordrecht.

The first complete mitochondrial (mt) genome of Ancyrocephalidae is reported herein. The mt genome of Tetrancistrum nebulosi was 13,392 bp in length containing 12 protein-coding genes (lacking atp8), 22 tRNA genes and 2 rRNA genes. The longest non-coding region was located between nad5 and trnG, and the A + T content was 72.4%. All tRNAs had the typical clover-leaf secondary structure except for trnS1(AGN), trnR, trnF and trnQ. The rrnL and rrnS subunits were separated by trnC, as documented in the monopisthocotylean groups (Benedenia and Gyrodactylus species), while they were adjacent to each other in the polyopisthocotylean species (Microcotyle sebastis, Polylabris halichoeres and Pseudochauhanea macrorchis). © 2014 Informa UK Ltd.

A HPLC method was developed for the determination of multiple preservatives in fruits and vegetables, including ortho-phenyl phenol, thiabendazole, imazalil, carbendazim, cinnamaldehyde, 2-naphthol, and 2, 4-dichlorobenzene oxygen ethanoic acid. Samples were extracted by alkaline ether. The extract was dissolved in methanol and finally injected into the HPLC system after the extraction solution was evaporated to dry on a rotary evaporator. The separations were effected by the gradient elution mode using a mobile phase composed of methanol-phosphate buffer (pH 6.8) at the flow of 0.8 mL/min. The eight preservatives were separated on an ODS-SP C18 column and then detected using a DAD detector with detection wavelength of 190~400 nm. The correlation coefficient of this method ranged from 0.9991 to 0.9997. The recoveries at spiked levels ranged from 93.4% to 97.4% and the relative standard deviations (RSD) ranged from 0.53 to 1.96. The results demonstrated that this method was simple, sensitive and accurate, which had a promising potential for practical applications in the determination of preservatives.

A novel 4-(2,4-dihydroxybenzylideneamino)benzenesulfonic acid (HL), and its kalium(I), calcium(II) complexes [M(L)n]·2nH2O·Cl n (M = K(1) n = 1, M = Ca(2) n = 2), have been prepared and characterized. The crystal and molecular structures of 1 and 2 were determined by single-crystal X-ray diffraction. The interaction of 1, 2 and ligand (L) with calf thymus DNA was investigated by UV-visible (UV-vis), fluorescence and viscosity measurements. Experimental results indicate that 1, 2 and L could bind to DNA via the intercalation mode, and the binding affinity of 1 is stronger than that of 2 and L. The intrinsic binding constants of 1, 2 and L were 5.60 × 105, 6.53 × 105 and 1.44 × 10 5 M-1, respectively. The cleavage reaction on plasmid DNA has been monitored by agarose gel electrophoresis. The results indicated that 1 and 2 could cleave pBR322 DNA. © 2012 Elsevier B.V. All rights reserved.

A method was established for the determination of vardenafil, sildenafil, and tadalafil in health foods using solid-phase extraction-ultra-performance liquid chromatography-tandem mass spectrometry (SPE-UPLC-MS/MS). The sample was extracted by methanol or hydrochloric acid, and then purified by a hydrophilic/lipophilic balanced (HLB) SPE column. The target compounds were separated on a reversed phase column using a solution of acetonitrile and 0.1 % formic acid in water as the mobile phase, and then detected by positive electrospray ionization under multiple reaction monitoring (MRM) mode. Good linearity of the calibration curves was achieved for three target compounds in the range of 10~200 μg/L, with correlation coefficients (r2) greater than 0.99. The average recoveries of the blank matrix sample were all more than 75%, and the relative standard deviations (RSDs) were less than 8%. The limits of detection (LOD) for vardenafil, sildenafil, and tadalafil in the capsule form were 10 μg/kg, 10 μg/kg, and 20 μg/kg, respectively. The limits of quantitation (LOQs) for vardenafil, sildenafil, and tadalafil in the capsule form were 40 μg/kg, 40 μg/kg, and 80 μg/kg, respectively. For oral samples, the LODs were 3 μg/L, 3 μg/L, and 6 μg/L, respectively; the LOQs were 10 μg/L, 10 μg/L, and 20 μg/L, respectively. The sample matrix effects were also studied, which had a suppressive effect on the response of vardenafil, sildenafil, and tadalafil, and the most significant suppressive effect was found on vardenafil. ©, 2014, South China University of Technology.
