Cyprinid herpesvirus 3 (CyHV-3) is the cause of a lethal disease named as koi herpesvirus disease (KHVD) in carp worldwide. Even though CyHV-3 has been found in China, little is known about the variety of CyHV-3 genotypes distributing in this area. In the present study, to improve the sensitivity of current genotyping approaches, a nested-PCR method was constructed based on the duplex PCR reported previously. The nested-PCR targeting the markers I/II loci was used in genotype identification of CyHV-3 infected fish in Guangdong Province in 2010-2015 and Henan Province in 2013. We detected 3 genotypes including an Asian dominant type (I++ II+) and two emerging genotypes in East Asian and South-East Asian regions (I++ II- and I++ II+Δ). I++ II- was the predominant genotype in this study. Two CyHV-3 variants, ZS410 and HZ419 isolates, were successfully isolated in cell culture and exhibited the genotype of I++ II- and I++ II+Δ, respectively. This is the first report of both I++ II- and I++ II+Δ genotypes isolated in China. These data can contribute consideration and exploration of mode of CyHV-3 spread in China. © 2016 The Japanese Society of Fish Pathology.

Eimeria tenella, an apicomplexan parasite in chickens, possesses an apicoplast and its associated metabolic pathways including the Type II fatty acid synthesis (FAS II). Malonyl-CoA:acyl-carry protein transacylase (MCAT) encoded by the fabD gene is one of the essential enzymes in the FAS II system. In the present study, the entire E. tenella MCAT gene (EtfabD) was cloned and sequenced. Immunolabeling located this protein in the apicoplast organelle in coccidial sporozoites. Functional replacement of the fabD gene with amber mutation of E. coli temperature-sensitive LA2-89 strain by E. tenella EtMCAT demonstrated that EcFabD and EtMCAT perform the same biochemical function. The recombinant EtMCAT protein was expressed and its general biochemical features were also determined. An alkaloid natural product corytuberine (CAS: 517-56-6) could specifically inhibit the EtMCAT activity (IC 50 = 16.47 μM), but the inhibition of parasite growth in vitro by corytuberine was very weak (the predicted MIC 50 = 0.65 mM). © 2012 Elsevier B.V.
