A green process for enantioselective hydrogenation of dehydroamino acid derivatives and dimethyl itaconate with a rhodium catalyst modified by a new phosphine-phosphoramidite has been developed providing 97.7-99.8% enantioselectivity in green solvents such as ethylene carbonate and propylene carbonate. The l-DOPA precursor was obtained by simple filtration in 71% yield with 99.5% ee. Dimethyl itaconate was hydrogenated under solvent-free condition at 50 °C with 98.7% ee. The new rhodium complex was heterogenized on a mesoporous Al 2O 3 support using phosphotungstic acid (PTA) as an anchoring agent and tested in heterogeneous batch and flow reaction modes. The supported catalyst was reused eight times in the batch mode with over 97% ee and used over 12 hours in the flow reaction mode with an average of 97% ee in the asymmetric hydrogenation reaction of (Z)-α-acetamidocinnamic acid methyl ester. © 2012 The Royal Society of Chemistry.

Kinetic resolutions of cyclic racemic secondary alcohols (2-methylenecyclopentan-1-ol rac-1a, 2-methylenecyclohexan-1-ol rac-1b, 2-methylenecycloheptan-1-ol rac-1c, 6-methylene-[1,3]dioxepan-5-ol rac-1d, 2,2-dimethyl-6-methylene-[1,3]dioxepan-5-ol rac-1e and trans-2-bromocyclohexan-1-ol rac-3) catalyzed by different (commercial and in-house-made) lipases were performed using vinyl acetate in THF-hexane. In the most typical cases (rac-1b, rac-1d and rac-3), the immobilized Candida antarctica lipase B (CaLB, for rac-1b and rac-3)- or sol-gel immobilized Pseudomonas fluorescens lipase (sol-gel LAK, for rac-1d)-catalyzed batch mode reactions were compared to the continuous mode reactions carried out in an enzyme-filled stainless steel bioreactor. The effect of temperature (20-60 °C) and flow rate (0.1-0.3 ml min-1) on the continuous-flow acetylation of rac-1b, rac-1d and rac-3 were investigated. In the kinetic resolutions of rac-1b, rac-1d and rac-3, the enantiomeric selectivities (E) were similar in the continuous-flow and batch (shake flask) modes. However, the productivities (specific reaction rate: r), were significantly higher in the continuous-flow mode biotransformations of rac-1b, rac-1d and rac-3. © 2010 Elsevier Ltd. 3D shape- or volume-based virtual screening is a broadly used approach in drug discovery. In recent years a large number of publications have appeared in which these tools were compared not only to competitive methods but to docking studies as well. Studies often showed that the effectiveness of docking could be highly variable due to a large number of possible confounding factors, while ligand-based, shape-based approaches were more consistent. Here, we describe a novel, fully flexible shape-based virtual screening algorithm that does not require previous 3D conformation or conformer generation. Due to its solid consistency it can easily be used on desktop computers by non-expert scientists. The algorithm is demonstrated in a study for the investigation of β-secretase inhibitors and benchmarked on the Directory of Useful Decoys data set. © 2014 American Chemical Society.

Various ketones were efficiently transformed into the corresponding amines using ammonium formate in the presence of Zn dust or 10% Pd/C. The low-cost Zn dust method proved to be effective in amine formation from carbonyl groups at the benzylic side-chain position of aromatic systems, whereas 10% Pd/C was an efficient catalyst in the reductive aminations of carbonyl groups non-conjugated with any π-system. The 10% Pd/C-catalyzed reductions were performed more effectively in a continuous-flow X-Cube reactor than in the batch system. © 2011 Elsevier Ltd. All rights reserved.

While memorials of all kinds and sizes have been presented since his death, the world’s first statue of the late Steve Jobs has today been unveiled in Budapest, Hungary. It’s made completely of bronze and stands nearby the entrance of architectural software maker Graphisoft’s headquarters. Jobs seems to be presenting a new product on stage, or possibly waving as he was known to do upon exiting the stage. The statue was commissioned by Graphisoft founder and chairman Gabor Bojar and sculpted by Hungarian sculptor Erno Toth. Graphisoft and Apple go way back, when Jobs first took a liking to Graphisoft’s ArchiCAD software in 1984 and threw Apple’s support behind it. “Apple’s support included cash and computers at a time when GRAPHISOFT was a small company with limited resources, working within the economic and political confines of what was, at the time, communist Hungary,” Gabor Bojar said. This is only the first in what should be many Steve Jobs tribute statues, and hopefully we’ll get one stateside pretty soon.
