Background Endothelin A (ET-A) receptor antagonists including zibotentan and atrasentan, have been suggested as a treatment for castration-resistant prostate cancer (CRPC). Our aim was to conduct a meta-analysis and indirect comparison to assess the efficacy and safety of ET-A receptor antagonists for treatment of CRPC. Methods We systematically searched PubMed, EMBASE, the Cochrane Library, and Web of Science from inception to November 2014 to identify randomized controlled trials (RCTs) which assessed ET-A receptor antagonists for treatment of CRPC. Meta-analysis was conducted by STATA version 12.0 software. Results Eight RCTs were identified, involving 6,065 patients. The results of direct comparison showed that compared with placebo, there was no statistically significant difference in the improvement of progression-free survival (PFS), overall survival (OS), time to disease progression (TTP), and total adverse events (AEs) with ET-A receptor antagonist treatment for CRPC. The results of ET-A receptor antagonists plus docetaxel versus docetaxel alone were similar. The indirect comparisons showed that there were no significant differences between zibotentan plus docetaxel versus atrasentan plus docetaxel when compared with docetaxel alone or zibotentan versus atrasenta compared with placebo in the improvement of PFS, OS, TTP, and total adverse events. Conclusions There were no significant benefits for ET-A receptor antagonists with or without docetaxel in the improvement of PFS, OS, TTP, and overall AEs. And there were no significant differences between zibotentan and atrasentan. Objective To investigate the awareness situation on the ARRIVE guideline and the Gold Standard Publication Checklist (GSPC) of animal experiments in researchers in animal experiments field in Lanzhou city, in order to improve the promotion of the two reporting guidelines in China. Methods A self-designed questionnaire was used to investigate the clinical graduate students and teachers in medical college in Lanzhou city. The investigation contents mainly included the basic information of the respondents, the awareness situation on the ARRIVE guideline, GSPC and other medical reporting guidelines. SPSS 21.0 software was used for data analysis. Results A total of 329 questionnaires (40 were from teachers and 289 were from graduate students) were issued, of which, 287 questionnaires were effective. The results showed that the awareness rate on the ARRIVE guideline and GSPC in clinical graduate students and teachers in medical college in Lanzhou city were 11.8% and 12.5%, respectively, and there was no significant difference between students and teachers in awareness rate (P=0.903). The survey approaches, the age, education, job, and the organization of the respondents were all not the influence factors of awareness rate (P>0.05). The respondents knew about the reporting guidelines mainly through the website (33.4%), related studies (21.2%) and academic reports (17.4%). Conclusion The awareness rate on the ARRIVE guideline and GSPC is relative low in researchers in animal experiments field in Lanzhou city, and it needed to take purposeful measures to promote and popularize them. © 2015 Editorial Board of Chin J Evid-based Med. To study whether the effects of Leflunomide were different in treating patients with rheumatoid arthritis (RA) of different Chinese medical syndrome patterns. Totally 150 RA patients were recruited and assigned to 5 Chinese medical syndrome types, i.e., the heat-dampness blocking collateral type, cold-dampness blocking collateral type, Shen-qi deficiency cold type, Gan-Shen yin deficiency type, and stagnant blood blocking collateral type according to Chinese medical syndrome typing standards. They were treated with Leflunomide, 3 months as one therapeutic course. The parameters including numbers of joint tenderness and swelling, morning stiffness time, scores estimated by Visual Analog Scale (VAS), as well as laboratory indices involving rheumatoid factor (RF), erythrocyte sedimentation rate (ESR), C-reactive protein (CRP), American College of Rheumatology 20% improvement (ACR20), and American College of Rheumatology 50% improvement (ACR50) were observed before and after treatment, and statistically analyzed. After treatment the numbers of joint tenderness, numbers of joint swelling, VAS scores, ESR, CRP, and RF all decreased, showing statistical difference when compared with those before treatment (P < 0.05). The morning stiffness time was shortened in the heat-dampness blocking collateral type, cold-dampness blocking collateral type, and stagnant blood blocking collateral type, showing statistical difference (P < 0.05). Of them, the numbers of joint tenderness, the numbers of joint swelling, the morning stiffness time, RF, VAS scores, and the improvement of the total effective rate were obviously better in the heat-dampness blocking collateral type, cold-dampness blocking collateral type, and stagnant blood blocking collateral type than in the Shen-qi deficiency cold type and Gan-Shen yin deficiency type, showing statistical difference (P < 0.05). Leflunomide showed significant effects in treating RA. Of them, its effects were obviously better in the heat-dampness blocking collateral type, cold-dampness blocking collateral type, and stagnant blood blocking collateral type than in the Shen-qi deficiency cold type and Gan-Shen yin deficiency type. BACKGROUND: In principle, fracture treatment mainly focuses on bone remodeling from the biological and mechanical aspects to combine mechanical fixation with biological fixation as well as combine intramedullary fixation with extramedullary fixation. OBJECTIVE: To introduce the principle development of fracture treatment and bone remodeling. METHODS: A computer-based search of CNKI (2006-01/2011-05) and PubMed (2006-01/2011-05) for bone remodeling articles using the keywords of "bone structure, rebuild, principle" in Chinese and English, respectively. RESULTS AND CONCLUSION: In 30 collected articles, 14 were included in result analysis. The treatment concept of fractures has been gradually changed from mechanical fixation system to biological fixation system. Fracture treatment research focuses on the balance between stable fixation and intact soft tissues, and the design of implants and instruments is significantly improved. In recent years, there are digitized virtual human technology, bio-mechanics finite element theory, trauma-control theory, pharmacological research for osteoporosis, and tissue engineering theory and technique to repair defects. In general, the concept of bone remodeling is to choose a most reasonable patient-centered way to guide and promote bone healing and functional recovery at the macro-and micro-level. Source
