Background Aconitum plants have been widely used in China for thousands of years. Recent evidences indicate that aconitine, the main active ingredient of Aconitum, has immunomodulatory properties that might be useful for treating autoimmune diseases, such as rheumatoid arthritis. In this study, we conducted a pilot study to explore the effect and mechanisms of aconitine on the treatment of systemic lupus erythematosus. Methods A pristane-induced murine model was used. The pristane-induced mice were treated with aconitine (25, 75 μg kg−1 d−1, po) for 9 weeks. Every three weeks, proteinuria was detected to monitor the kidney damage and blood was collected to measure serum levels of autoantibodies, besides the kidney pathological examination. The major B cell activating factor and major pro-inflammatory mediators, PGE2, IL-17a and IL-6, were also detected. Results We found that aconitine significantly improved the mouse health, decreased the elevated blood leukocyte counts, reduced the serum level of anti-double-stranded DNA (anti-dsDNA) antibody, greatly ameliorated renal histopathologic damage and reduced IgG deposit in glomerular. Furtherly, the levels of PGE2, IL-17a and IL-6, were found to have decreased in aconitine treated mice. Conclusion We have demonstrated that aconitine can inhibit the progression of disease and ameliorate the pathologic lesion of systemic lupus erythematosus. © 2017 The Authors

A series of polylactic acid (PLA) based nanocomposite fibrous membranes, including neat PLA, PLA/hydroxyapatite (HA) and PLA/HA/graphene oxide (GO), were fabricated via electrospinning method. The morphology and composition were investigated by scanning electron microscopy (SEM), X-ray diffraction (XRD) and Fourier transform infrared spectroscopy (FTIR) respectively. The thermal stability was determined by thermogravimetric analysis (TGA). To estimate the cytocompatibility of asprepared PLA/HA/GO fibrous membrane, MC3T3-E1 cells were cultured, and the corresponding cell adhesion and differentiation capability were investigated by fluorescence microscopy, SEM and MTT test. The electrospun ternary PLA/HA/GO membrane exhibited three-dimensional fibrous structure with relatively rough surface morphology, which made itself ideal for cell attachment and proliferation in bone tissue regeneration. The fluorescence microscopy, SEM and MTT test confirmed that the PLA/HA/GO nanocomposite fibrous membrane created a proper environment for the seeding and proliferation of MC3T3-E1 cells. © 2012 The Author(s).

There are a variety of ways for isolation and selective cultivation of retinal microvascular endothelial cells in vitro, in which the newly reported tissue culture with collagenase digestion method are widely used for simple, economical, high purity and good activity of obtained endothelial cells. As a result of the limitation of person eye donor, researchers often use rat retinal microvascular endothelial cells that are highly homologous to human beings to experiment in vitro. In this paper, the methods of culturing retinal microvascular endothelial cells in vitro will be elaborated from the aspects of animal species, added factors, cell purification and identification in vitro. At the same time, the advantages and disadvantages of these methods in the process of operation are briefly described, which can provide a reference for the researchers to select suitable cultivation methods. The QUOROM and PRISMA statements were published in 1999 and 2009, respectively, to improve the consistency of reporting systematic reviews (SRs)/meta-analyses (MAs) of clinical trials. However, not all SRs/MAs adhere completely to these important standards. In particular, it is not clear how well SRs/MAs of acupuncture studies adhere to reporting standards and which reporting criteria are generally ignored in these analyses.To evaluate reporting quality in SRs/MAs of acupuncture studies.We performed a literature search for studies published prior to 2014 using the following public archives: PubMed, EMBASE, Web of Science, the Cochrane Database of Systematic Reviews (CDSR), the Chinese Biomedical Literature Database (CBM), the Traditional Chinese Medicine (TCM) database, the Chinese Journal Full-text Database (CJFD), the Chinese Scientific Journal Full-text Database (CSJD), and the Wanfang database. Data were extracted into pre-prepared Excel data-extraction forms. Reporting quality was assessed based on the PRISMA checklist (27 items).Of 476 appropriate SRs/MAs identified in our search, 203, 227, and 46 were published in Chinese journals, international journals, and the Cochrane Database, respectively. In 476 SRs/MAs, only 3 reported the information completely. By contrast, approximately 4.93% (1/203), 8.81% (2/227) and 0.00% (0/46) SRs/Mas reported less than 10 items in Chinese journals, international journals and CDSR, respectively. In general, the least frequently reported items (reported50%) in SRs/MAs were protocol and registration, risk of bias across studies, and additional analyses in both methods and results sections.SRs/MAs of acupuncture studies have not comprehensively reported information recommended in the PRISMA statement. Our study underscores that, in addition to focusing on careful study design and performance, attention should be paid to comprehensive reporting standards in SRs/MAs on acupuncture studies.

To study whether the effects of Leflunomide were different in treating patients with rheumatoid arthritis (RA) of different Chinese medical syndrome patterns. Totally 150 RA patients were recruited and assigned to 5 Chinese medical syndrome types, i.e., the heat-dampness blocking collateral type, cold-dampness blocking collateral type, Shen-qi deficiency cold type, Gan-Shen yin deficiency type, and stagnant blood blocking collateral type according to Chinese medical syndrome typing standards. They were treated with Leflunomide, 3 months as one therapeutic course. The parameters including numbers of joint tenderness and swelling, morning stiffness time, scores estimated by Visual Analog Scale (VAS), as well as laboratory indices involving rheumatoid factor (RF), erythrocyte sedimentation rate (ESR), C-reactive protein (CRP), American College of Rheumatology 20% improvement (ACR20), and American College of Rheumatology 50% improvement (ACR50) were observed before and after treatment, and statistically analyzed. After treatment the numbers of joint tenderness, numbers of joint swelling, VAS scores, ESR, CRP, and RF all decreased, showing statistical difference when compared with those before treatment (P < 0.05). The morning stiffness time was shortened in the heat-dampness blocking collateral type, cold-dampness blocking collateral type, and stagnant blood blocking collateral type, showing statistical difference (P < 0.05). Of them, the numbers of joint tenderness, the numbers of joint swelling, the morning stiffness time, RF, VAS scores, and the improvement of the total effective rate were obviously better in the heat-dampness blocking collateral type, cold-dampness blocking collateral type, and stagnant blood blocking collateral type than in the Shen-qi deficiency cold type and Gan-Shen yin deficiency type, showing statistical difference (P < 0.05). Leflunomide showed significant effects in treating RA. Of them, its effects were obviously better in the heat-dampness blocking collateral type, cold-dampness blocking collateral type, and stagnant blood blocking collateral type than in the Shen-qi deficiency cold type and Gan-Shen yin deficiency type.

Inadequate venous drainage can lead to congestion and necrosis of flaps used in the repair of defects, thereby elevating the risk of flap failure. In this study, we sought to test the hypothesis that the venous drainage was better in flow-through flaps than in conventional dorsalis pedis free flaps used in the repair of dorsal foot defects. In this retrospective study, we investigated the data of 14 patients who underwent repair with flow-through flaps (n = 7) or conventional flaps (n = 7) for dorsal foot defects, between January 2007 and December 2013. The defects ranged from 6.2 × 11 cm to 9.5 × 16 cm in size. The donor sites were resurfaced using full-thickness free-skin grafts, and after transfer, the flaps were evaluated for postoperative congestion, surviving area, and sensory function. The results showed that the operative time was significantly longer for flow-through flaps than for conventional flaps (6.4 ± 1.7 h vs. 4.3 ± 1.2 h, P = 0.020), mainly due to additional dissection of the first dorsal metatarsal artery required in the case of the former. Necrosis was observed in the case of 4 conventional flaps, but not in the case of flow-through flaps. The flow-through flaps showed significantly lower incidence of congestion and higher survival area proportion than the conventional flaps (P < 0.05). The flow-through dorsalis pedis flaps have the advantages of lower incidence of necrosis and congestion and better survival over the conventional flaps in the repair of dorsal foot defects, and absence of additional morbidities, but required a longer operative time than conventional flaps. © 2015, E-Century Publishing Corporation. Adjustable spine prone frame is used in posterior lumbar surgery, during operation, X-ray can shoot fast and internal fixation implants can fix accurately, the vacant abdominal ensure normal respiratory and circulatory function, and the lumbar curvature can adjust conveniently. It meets the surgical position and care requirements, can prevent the complications effectively.

Gastric cancer remains highly prevalent, but treatment options are limited. Natural products have proved to be a rich source of anticancer drugs. Chrysosplenium nudicaule Ledeb. (Saxifragaceae) is a perennial herb that grows in the highlands of China. It has been used as a traditional Chinese medicine to treat digestive diseases for hundreds of years. Recent studies revealed that this herb had anticancer activity, and the flavonoids were speculated to be the effective components. 6,7,3-Trimethoxy-3,5,4-trihydroxy flavone (TTF) and 5,4-dihydroxy-3,6,3trimethoxy-flavone-7-O--d-glucoside (DTFG) are flavonoid compounds isolated from Chrysosplenium nudicaule.This study examined the effect of TTF and DTFG on SGC-7901 human stomach cancer cell in vitro to determine the anticancer and induction of apoptosis properties of TTF.The proliferation of cells treated with 32, 16, 8, 4, and 2g/mL of TTF or DTFG for 24, 48, and 72h was assessed by the MTT assay. After being treated with TTF, the apoptosis of SGC-7901 cells was assessed by acridine orange staining, ultrastructure, electrophoresis of DNA fragmentation, and flow cytometry.Results indicated that TTF inhibited the growth of cancer cells with an IC50 value of 8.33g/mL after 72h incubation. However, DTFG showed no inhibitory effect on the growth of the cancer cell. Further studies on TTF also confirmed that it was able to induce apoptosis of SGC-7901 cells at a concentration as low as 4g/mL.The apoptotic effect of TTF makes it a promising candidate for future chemotherapeutic application in treating stomach cancer.

BACKGROUND: In principle, fracture treatment mainly focuses on bone remodeling from the biological and mechanical aspects to combine mechanical fixation with biological fixation as well as combine intramedullary fixation with extramedullary fixation. OBJECTIVE: To introduce the principle development of fracture treatment and bone remodeling. METHODS: A computer-based search of CNKI (2006-01/2011-05) and PubMed (2006-01/2011-05) for bone remodeling articles using the keywords of "bone structure, rebuild, principle" in Chinese and English, respectively. RESULTS AND CONCLUSION: In 30 collected articles, 14 were included in result analysis. The treatment concept of fractures has been gradually changed from mechanical fixation system to biological fixation system. Fracture treatment research focuses on the balance between stable fixation and intact soft tissues, and the design of implants and instruments is significantly improved. In recent years, there are digitized virtual human technology, bio-mechanics finite element theory, trauma-control theory, pharmacological research for osteoporosis, and tissue engineering theory and technique to repair defects. In general, the concept of bone remodeling is to choose a most reasonable patient-centered way to guide and promote bone healing and functional recovery at the macro-and micro-level.
