Background: von Willebrand factor (vWF) is a potent regulator of angiogenesis, tumor growth, and metastasis. Yet, the expression pattern of vWF in human gastric cancer (GC) tissues and its relation to clinicopathological features of these cases remains unknown. Methods: Tumor and 5-cm adjacent non-tumoral parenchyma specimens were collected from 99 patients with GC (early stages I/II and late stages III/IV), and normal specimens were collected from 32 healthy controls (reference group). Plasma vWF antigen (vWF:Ag) and vWF activity were assessed by ELISA. The role of vascular endothelial growth factor (VEGF) in differential vWF expression was investigated using cultured human umbilical vein endothelial cells (HUVECs). vWF and VEGF protein and mRNA expression levels were investigated by qRT-PCR, western blotting and immunohistochemistry (IHC) respectively. The correlation of IHC-detected vWF expression with patient clinicopathological characteristics was analyzed. Results: Compared to the reference group, the patients with late GC showed significantly higher levels of vWF:Ag (72% (21-115) vs. 101% (40-136)) and vWF activity (62% (20-112) vs. 117% (33-169)) (both P < 0.001). The GC tumor tissues also showed higher vWF mRNA and protein levels than the adjacent non-tumoral parenchyma. Patients at late GC stage had significantly higher median number of vWF-positive cells than patients at early GC stage (P < 0.05). VEGF induced vWF mRNA and protein expression in HUVECs in dose- and time-dependent manners. Patients with late GC stage also had significantly higher serum VEGF than patients at early GC stage (23 ± 26 vs. 10 ± 12 pg/mL, P < 0.01). Most of the undifferentiated GC tumor tissues at late disease stage exhibited strong VEGF and VEGFR2 protein staining, which co-localized with the vWF protein staining pattern. Conclusions: GC-related plasma vWF:Ag and vWF activity levels become substantially elevated in the late stage of disease. The higher mRNA and protein expression of vWF in GC tumor stroma may be regulated by the VEGF-VEGFR2 signaling pathway in vitro and may contribute to GC progression in vivo. © 2015 Yang et al.; licensee BioMed Central. Background/Aim: To assess the status of 25-hydroxyvitamin D [25(OH)D] in pregnant women in their second and third trimesters, who reside and work indoors in Guiyang, China. Subjects and methods: A total of 311 pregnant women in their 12th to 40th gestational week were engaged in employment located indoors in the urban area of Guiyang and completed a questionnaire on living habits. Levels of serum 25(OH)D were measured from fasting venous blood by liquid chromatography-mass spectrometry (LC-MS/MS). Levels of 25(OH)D were classified as vitamin D deficient [25(OH)D<20 ng/ml], insufficient [20 ng/ml≤25(OH)D<30 ng/ml], or sufficient [25(OH)D≥30 ng/ml]. Results: The mean serum level of 25(OH)D was 14.69±6.81 ng/ml. Vitamin D deficiency, insufficiency and sufficiency were found in 260 (83.6%), 39 (12.5%), and 12 (3.9%) women, respectively. The mean level of 25(OH)D in the third trimester was significantly higher than in the second trimester (p<0.001). The mean 25(OH)D level in summer (June, July, August) was significantly higher than in the other seasons (p<0.001). The 25(OH)D level in pregnant women compliant with pre-natal calcium or multi-vitamin supplements was higher than in those not taking supplements (p<0.001). Conclusions: These results suggest that pregnant women residing in Guiyang urban area and working indoors are at high risk of vitamin D insufficiency, particularly during spring, winter, and autumn, regardless of use of pre-natal calcium and multivitamins. Appropriate vitamin D supplementation is necessary to improve maternal vitamin D nutrition. © 2013, Editrice Kurtis. Source

Background: The increase of higher order aberration may be an important cause of blurred, bad night vision and glare after cataract and intraocular lens (IOL) implantation surgery. However, seldom study on the relationship between higher order aberration and contrast sensitivity after cataract surgery is published. Objective: The study aimed to investigate the correlation of high-order aberrations with contrast sensitivity (CS) and glare contrast sensitivity (GS) after phacoemulsification and IOL implantation. Methods: One hundred and twenty nine eyes received phacoemulsification and IOL implantation for age-related cataract with the post-operation vision ≥ 0.8 were collected in this study. The uncorrect vision, best correct vision, the opacification of lens, intraocular pressue were examined in 3 months after surgery. The high-order aberrations such as general high-order aberrations, spherical aberration, and coma by Zywave wavefront instrument were detected. The correlation of higher order abbration and contrast sensitivity, glare contrast sensitivity was analyzed by Spearman rank correlation analysis. Written informed consent was obtained from subjects before any medical process. Results: The positive correlations were found between total higher order abbration and CS at 12 c/d (r = 0.23, P < 0.05) or Z400 and CS at 3 c/d (r = 0.17, P = 0.05). The negative correlations were seen between Z440 and CS at 9 c/d (r = -0.21, P < 0.05) or Z550 and CS at 12 c/d (r = -0.18, P < 0.05). In additional, the positive correlations were found between Z441 and GS at 9 c/d (r = 0.19, P < 0.05), and the negative correlations were found between Z331 and GS at 6 c/d (r = -0.18, P < 0.05), Z420 and GS at 9 c/d (r = -0.25, P < 0.05), Z420 and GS at 12 c/d (r = -0.21, P < 0.05), Z440 and GS at 9 c/d (r = -0.21, P < 0.05), Z440 and GS at 12 c/d (r = -0.23, P < 0.05), Z510 and GS at 9 c/d (r = -0.18, P < 0.05), Z530 and GS at 12 c/d (r = -0.23, P < 0.05). The 4-order high-order aberrations relevant to GS mainly focused on middle and high spatial frequency bands. There were only scattered correlations between aberrations and CS. Conclusion: The changes of high-order aberrations do not influent CS after phacoemulsification and IOL implantation surgery, but it influent the GS. Source
