Hydrogen sulfide (H2S), produced endogenously by the activation of two major H2S-generating enzymes (cystathionine β-synthase and cystathionine γ-lyase), plays important regulatory roles in different physiologic and pathologic conditions. The abnormal metabolism of H2S is associated with fibrosis pathogenesis, causing damage in structure and function of different organs. A number of in vivo and in vitro studies have shown that both endogenous H2S level and the expressions of H2S-generating enzymes in plasma and tissues are significantly downregulated during fibrosis. Supplement with exogenous H2S mitigates the severity of fibrosis in various experimental animal models. The protective role of H2S in the development of fibrosis is primarily attributed to its antioxidation, antiapoptosis, anti-inflammation, proangiogenesis, and inhibition of fibroblasts activities. Future studies might focus on the potential to intervene fibrosis by targeting the pathway of endogenous H2S-producing enzymes and H2S itself. © 2015 Shufang Zhang et al.

Objective: To analyze the prevalence, anatomical features, as well as variance of the sternalis muscle in the Chinese population using multi-detector computed tomography (MDCT). Methods: We retrospectively reviewed 6,000 adult axial MDCT images to determine the overall and gender prevalence of the sternalis muscles. We also analyzed the side prevalence and anatomical features, including shape, size, location and course. Results: The sternalis muscle was present in 347 (5.8 %) of 6,000 adults. This muscle was more common in males (6.0 %, 187/3091) than in females (5.5 %, 160/2909). Among the 347 adults, 118 (34.0 %) had bilateral sternalis muscles; 148 (42.7 %) had right sternalis muscles; and 81 (23.3 %) had left sternalis muscles. The sternalis muscle was either flat or nodular and located superficial to the major pectoral muscles on CT axial transverse images. According to the muscle morphology and course, we classified sternalis muscles as three different types and nine subtypes. The muscles appeared with a single head and single belly in 58.5 %, double or multiple heads in 18.1 %, and double or multiple bellies in 23.4 %. The mean length, width and thickness were 111.1 ± 33.0, 17.7 ± 9.9 and 4.1 ± 1.7 mm measured on MDCT. Conclusion: The sternalis muscle was highly prevalent in normal Chinese adults. MDCT is an effective method to demonstrate this muscle in vivo. © 2013 Springer-Verlag.

OBJECTIVE: To investigate the expression of Caspase-9 in thyroid carcinoma and thyroid benign tumor and to analyze clinical signficance. METHODS: The expression of Caspase-9 protein was detected with immunohistochemical Enision TM plus non-hiotin technique in 65 cases of thyroid carcinoma and 40 cases of thyroid benign tumor as well as normal tissues. RESULTS: The positive expression rates of Caspase-9 protein in thyroid carcinoma, thyroid benign tumor and normal tissues were 21.5%(14/65), 50.0%(20/40), 85.0%(34/40). There was significantly different among three groups (χ2=7.65, P<0.01). The expression of Caspase-9 were positively related to AJCC clinical stage, differentiation degree, lymph node metascasis and the degree of infiltration (χ2 were 6.39, 7.08, 8.17 and 9.25, all P<0.01). CONCLUSION: The low expression of Caspase-9 protein may play important roles in the carcinogenesis of thyroid carcinoma and it is involved in the metastasis and infiltration.

Objective: To compare the effect of different antigen retrieval methods of immunohistochemistry in detection of p16 and cyclin D1 in cervical squamous epithelial lesions. Methods: Expression of p16 and cyclin D1 was detected by immunohistochemistry in 45 tissue specimens of cervical squamous epithelial lesions. Four antigen retrieval methods were applied in immunohistochemistry. Groups A and B use EDTA as repairing fluid, with B using pressure cooker for 1.5 min; Groups C and D use citrate buffer (CB) and pressure cooker for 1.5 min and 3 min respectively. Wilcoxon nonparametric test was used for comparison. Results: There was no significant difference in p16 detection between methods A and B or methods C and D(P>0.05). There were significant differences in p16 expression in lesions of both CIN class I and II between methods B and C(P=0.001; P=0.028). There was no difference in the expression of cyclin D1 among 4 antigen repair methods. The positive staining was located in the cervical basal nuclei, a small number of cervical columnar epithelial cells and all levels of cervical lesions near the basal cell area; but the express strength of cyclin D1 when using EDTA was stronger than that using CB. Staining was the strongest in method B with intense background staining and generally colored cytoplasm. In method A positive signal was slightly diminished, contrast was marked and endochylema was occasionally stained. Conclusion: Immunohistochemical staining of p16 in cervical lesions by using CB for antigen retrieval may reflect the evolution of lesions more clearly.

A novel, simple and sensitive method based on the use of a PRiME (process, robustness, improvements, matrix effects, ease of use) pass-through cleanup procedure combined with ultra-fast liquid chromatography-tandem quadrupole mass spectrometry (UFLC-MS/MS) for the determination of 4 AFs, i.e., AFB1, AFB2, AFG1 and AFG2 in human serum, was developed and validated. The effective PRiME pass-through cleanup procedure using the Oasis PRiME HLB cartridge was optimized for the extraction of AFB1, AFB2, AFG1 and AFG2 in human serum. The obtained results demonstrated the higher extraction capacity of the PRiME pass-through cleanup procedure with recoveries between 92.1% and 102%. The limits of detection (LODs) for AFB1, AFB2, AFG1 and AFG2 were 0.048 μg L-1, 0.069 μg L-1, 0.043 μg L-1 and 0.14 μg L-1, respectively. Validation results on linearity, specificity, accuracy and precision, as well as on application to the analysis of AFs in 10 liver cancer volunteers demonstrated the applicability to clinical studies. © The Royal Society of Chemistry 2016.
