Purpose: To construct a preoperative nomogram to differentiate invasive pulmonary adenocarcinomas (IPAs) from preinvasive lesions in patients with solitary pure ground-glass nodules (GGN). Methods: A primary cohort of patients with pathologically confirmed pulmonary solitary pure GGN after surgery were retrospectively studied at five institutions from January 2009 to September 2015. Half of the patients were randomly selected and assigned to a model-development cohort, and the remaining patients were assigned to a validation cohort. A nomogram predicting the invasive extent of the solitary GGNs was constructed based on the independent risk factors. Predictive performance was evaluated by concordance index (C-index) and calibration curve. Results: Out of 898 cases included in the study, 501 (55.8%) were preinvasive lesions and 397 (44.2%) were IPAs. In the univariate analysis, lesion size (p < 0.001), lesion margin (p = 0.041), lesion shape (p < 0.001), mean computed tomography (CT) value (p = 0.018), presence of pleural indentation (p = 0.017), and smoking status (p = 0.014) were significantly associated with invasive extent. In multivariate analysis, lesion size (p < 0.001), lesion margin (p = 0.042), lesion shape (p < 0.001), mean CT value (p = 0.014), presence of pleural indentation (p = 0.026), and smoking status (p = 0.004) remained the predictive factors of invasive extent. A nomogram was developed and validation results showed a C-index of 0.94, demonstrating excellent concordance between predicted and observed results. Conclusions: We established and validated a novel nomogram that can identify IPAs from preinvasive lesions in patients with solitary pure GGN.

A novel, simple and sensitive method based on the use of a PRiME (process, robustness, improvements, matrix effects, ease of use) pass-through cleanup procedure combined with ultra-fast liquid chromatography-tandem quadrupole mass spectrometry (UFLC-MS/MS) for the determination of 4 AFs, i.e., AFB1, AFB2, AFG1 and AFG2 in human serum, was developed and validated. The effective PRiME pass-through cleanup procedure using the Oasis PRiME HLB cartridge was optimized for the extraction of AFB1, AFB2, AFG1 and AFG2 in human serum. The obtained results demonstrated the higher extraction capacity of the PRiME pass-through cleanup procedure with recoveries between 92.1% and 102%. The limits of detection (LODs) for AFB1, AFB2, AFG1 and AFG2 were 0.048 μg L-1, 0.069 μg L-1, 0.043 μg L-1 and 0.14 μg L-1, respectively. Validation results on linearity, specificity, accuracy and precision, as well as on application to the analysis of AFs in 10 liver cancer volunteers demonstrated the applicability to clinical studies. © The Royal Society of Chemistry 2016.

Hydrogen sulfide (H2S), produced endogenously by the activation of two major H2S-generating enzymes (cystathionine β-synthase and cystathionine γ-lyase), plays important regulatory roles in different physiologic and pathologic conditions. The abnormal metabolism of H2S is associated with fibrosis pathogenesis, causing damage in structure and function of different organs. A number of in vivo and in vitro studies have shown that both endogenous H2S level and the expressions of H2S-generating enzymes in plasma and tissues are significantly downregulated during fibrosis. Supplement with exogenous H2S mitigates the severity of fibrosis in various experimental animal models. The protective role of H2S in the development of fibrosis is primarily attributed to its antioxidation, antiapoptosis, anti-inflammation, proangiogenesis, and inhibition of fibroblasts activities. Future studies might focus on the potential to intervene fibrosis by targeting the pathway of endogenous H2S-producing enzymes and H2S itself. © 2015 Shufang Zhang et al.

Objective: To compare the effect of different antigen retrieval methods of immunohistochemistry in detection of p16 and cyclin D1 in cervical squamous epithelial lesions. Methods: Expression of p16 and cyclin D1 was detected by immunohistochemistry in 45 tissue specimens of cervical squamous epithelial lesions. Four antigen retrieval methods were applied in immunohistochemistry. Groups A and B use EDTA as repairing fluid, with B using pressure cooker for 1.5 min; Groups C and D use citrate buffer (CB) and pressure cooker for 1.5 min and 3 min respectively. Wilcoxon nonparametric test was used for comparison. Results: There was no significant difference in p16 detection between methods A and B or methods C and D(P>0.05). There were significant differences in p16 expression in lesions of both CIN class I and II between methods B and C(P=0.001; P=0.028). There was no difference in the expression of cyclin D1 among 4 antigen repair methods. The positive staining was located in the cervical basal nuclei, a small number of cervical columnar epithelial cells and all levels of cervical lesions near the basal cell area; but the express strength of cyclin D1 when using EDTA was stronger than that using CB. Staining was the strongest in method B with intense background staining and generally colored cytoplasm. In method A positive signal was slightly diminished, contrast was marked and endochylema was occasionally stained. Conclusion: Immunohistochemical staining of p16 in cervical lesions by using CB for antigen retrieval may reflect the evolution of lesions more clearly.

Objective: To analyze the prevalence, anatomical features, as well as variance of the sternalis muscle in the Chinese population using multi-detector computed tomography (MDCT). Methods: We retrospectively reviewed 6,000 adult axial MDCT images to determine the overall and gender prevalence of the sternalis muscles. We also analyzed the side prevalence and anatomical features, including shape, size, location and course. Results: The sternalis muscle was present in 347 (5.8 %) of 6,000 adults. This muscle was more common in males (6.0 %, 187/3091) than in females (5.5 %, 160/2909). Among the 347 adults, 118 (34.0 %) had bilateral sternalis muscles; 148 (42.7 %) had right sternalis muscles; and 81 (23.3 %) had left sternalis muscles. The sternalis muscle was either flat or nodular and located superficial to the major pectoral muscles on CT axial transverse images. According to the muscle morphology and course, we classified sternalis muscles as three different types and nine subtypes. The muscles appeared with a single head and single belly in 58.5 %, double or multiple heads in 18.1 %, and double or multiple bellies in 23.4 %. The mean length, width and thickness were 111.1 ± 33.0, 17.7 ± 9.9 and 4.1 ± 1.7 mm measured on MDCT. Conclusion: The sternalis muscle was highly prevalent in normal Chinese adults. MDCT is an effective method to demonstrate this muscle in vivo. © 2013 Springer-Verlag.

Aim: To investigate the effects of early enteral nutrition on nutritional indicators and immune function in coma patients with brain trauma.Methods: Sixty coma patients with traumatic brain injury coma treated at our hospital from January 2013 to December 2013 were divided into either a study group or a control group, with 30 patients in each group. The control group received parenteral nutrition support and treatment, and the study group was given early enteral nutrition care. Nutritional indicators and immune function were compared between the two groups of patients.Results: After nursing case, albumin, prealbumin and hemoglobin levels were significantly lower in the control group than in the study group (34.7 mg/dL ± 5.7 mg/dL vs 41.7 mg/dL ± 6.3 mg/dL, 125.7 pg/dL ± 11.7 pg/dL vs 204.4 pg/dL ± 13.6 pg/dL, 114.7 mg/dL ± 17.8 mg/ dL vs 126.7 mg/dL ± 18.4 mg/dL, P < 0.05), and CD4+, CD4+/CD8+, IgA, IgG, and IgM were significantly better in the study group than in the control group (P < 0.05).Conclusion: Early implementation of enteral nutrition support and care is feasible in coma patients with traumatic brain injury, and can better improve the nutritional indicators and immune function. © 2014 Baishideng Publishing Group Inc. Diabetic cardiomyopathy is a severe complication of diabetes mellitus (DM). The goal of current work was to study the effects of sinomenine on streptozotocin-induced cardiomyopathy in rats. DM in rats was induced by intraperitoneal injection of streptozotocin. Cardiac function was evaluated by measuring left ventricle end-diastolic diameter, left ventricle end-systolic diameter and ejection fraction. Cardiac inflammation was evaluated by the degree of infiltration of T lymphocytes and the levels of pro-inflammatory cytokines. Sinomenine attenuated diabetic symptoms without affecting plasma glucose. Cardiac dysfunction in the sinomenine-treated diabetic rats was significantly improved, as reflected by decreased levels of left ventricle end-diastolic diameter, left ventricle end systolic diameter and an increased level of ejection fraction. Sinomenine observably reduced cardiomyocyte hypertrophy in DM rats. Moreover, sinomenine reduced infiltration of CD3+ and CD68+ positive cells and decreased the levels of tumor necrosis factor-, interlukin-1 and interlukin-6. Finally, sinomenine-treated rats showed a reduced expression of NF-B and an increased expression of IB in the myocardium compared with the myocardium of untreated diabetic rats. Our results indicate sinomenine significantly improves cardiac function in diabetic rats, which may be attributed to the deactivation of NF-B and the blockade of inflammatory cytokine-mediated immune reactions.

To investigate the changes in lymphocyte subsets that are caused by infection with different pathogens in children with hand, foot, and mouth disease.T lymphocyte subsets were measured in the patients peripheral blood, and serum, throat swab, and fecal samples were tested for enterovirus.Fecal and throat swab samples exhibited similar positive detection rates, and were significantly more likely to be positive, compared to serum samples (P<0.01). The EV71-positive group exhibited significantly lower CD4+TM cell counts (QR: 1.058), compared to the CD4+TM cell counts in the CoxA16-positive group (QR: 1.391; P<0.05).Throat swab and fecal samples exhibited significantly higher positive detection rates, compared to serum samples. In addition, EV71-infected children exhibited significantly lower CD4+ T-cell counts, compared to CoxA16-infected children, which suggests that EV71 infection may be associated with a poorer prognosis.
