The need to screen the kinetic characteristics of pharmaceuticals in early development stages is justified. The advantages and limitations of cassette dosing in pharmacokinetic investigations are discussed. The dose and the presence or absence of pharmaceutical interactions are shown to be the critical factors affecting the reliability of pharmacokinetic parameters assessed by the cassette method. The hypothesis that toxicokinetic cassette screening of compounds capable of interacting is appropriate for describing the slow elimination phase was verified by comparing lewisite excretion parameters after injection into laboratory animals of lewisite and a mustard-gas—lewisite mixture. © 2016 Springer Science+Business Media New York

Objective. Analysis of factors affecting results of inferior vena cava (IVC) thrombectomy in patients with renal cell carcinoma. Materials and methods. Nephrectomy with thrombectomy from IVC was performed in 53 patients (31 (58.5 %) males and 22 (41.5 %) females) with extended thrombosis. Patient age was 35-76 years (mean age 58.7 ± 10.1 years). In 36 (68 %) patients stage T3b was determined (mean thrombus length 5.28 ± 0.32 cm), in 17 (32 %) patients - stage T3c (mean thrombus length - 13.00 ± 0.71 cm). Postoperative complications were evaluated in accordance with the Clavien-Dindo classification. Calculations were performed using statistics software SPSS Statistics 16. Results. No intraoperative complications were observed, mean blood loss was 714 (200-4000) ml. Use of the Cell Saver system allowed to perform reinfusion in the majority of patients. Blood transfusion of 200-500 ml was necessary in 15 patients. In 32 (60.3 %) patients no complications were observed. Grade I complications were observed in 2 (3.7 %) patients, grade II - in 15 (28.3 %) patients, IIIA grade - in 2 (3.7 %) patients, and grade IIIB - in 1 (1.8 %) patient; there wasn't any grade IV complications. Two (3.8 %) patients died of pulmonary artery thromboembolism. Correlation analysis has shown a high dependency of complications on thrombus size (r = 0.7) and blood loss volume (r = 0.6), and low dependency on tumor size (r = 0.44); complications weren't affected by patients' age (r = 0.1) or Karnofsky status (r = 0.0). Median overall survival was 48 months, 3-year survival was 57.1 %, median progression-free survival was 46 months, 3-year progression-free survival was 56.8 %; 90-day morbidity was absent. Three-year overall survival for pN0 was significantly higher than for pN1 (69 % vs. 25 %; p = 0.014). There was no difference in terms of survival between patients with supradiaphragmatic and subdiaphragmatic thrombi (p = 0.14). Conclusion. Results of our study show that surgical treatment of patients with renal cell carcinoma and extended IVC thrombosis is effective.

The results of the development of a medical-purpose extraction 188Re generator and of 188Re radiopharmaceuticals are presented. The main advantage of this generator is the possibility of using as starting material tungsten oxide of natural isotope composition, irradiated in a reactor with medium neutron flux [(1.0–1.4) × 1014n cm–2 s–1]. Separation of the parent 188W and daughter 188Re was performed in a centrifugal semicounter-current contactor developed at the Institute of Physical Chemistry and Electrochemistry, Russian Academy of Sciences. The 188Re yield is about 90%, and the radiochemical purity (Na188ReO4 content) is ≥95%. The content of the 188W impurity is less than 1 × 10–3% relative to the 188Re activity, and the content of other radionuclides is less than 1 × 10–4%. The resulting Na188ReO4 solution was registered as “Active pharmaceutical substance” and was used in the development and subsequent preclinical trials of several radiopharmaceuticals for radionuclide therapy of pain syndrome accompanying the development of cancer metastases into the skeleton, and also for radiosynovectomy. © 2016, Pleiades Publishing, Inc.

Methylation of CpG-islands in promoter regions as well as interaction of miRNAs with messenger RNAs of target genes are related to multilayer mechanisms regulating gene expression. The goal of this study was to assess a possibility for miRNA gene methylation to influence indirectly activation of their target genes in lung tumors. By using a unified collection of samples of non-small cell lung cancer, it was demonstrated that elevated levels of mRNA for RHOA and NKIRAS1 genes were significantly (Spearman rank correlation, P < 10-11) associated both with loss of methylation in their CpG-islands and methylation in a number of miRNA genes, which, according to the miRWalk database, were predicted to possess regulatory functions. Novel potential regulatory miRNAs for RHOA (miR-9-1/-3, -34b/c, -129-2, -125b-1, -375, -1258) and NKIRAS1 (miR-34b/c, -129-2, -125b-1, -193a, -124a-1/-2/-3, -212, -132) genes in lung cancer were identified. © 2015 Pleiades Publishing, Ltd.

It has been established that intraperitoneal bolus administration of S-nitrosoglutathione (GS-NO) (12.5 μmoles/kg; 10 injections in 10 days), beginning with day 4 after transplantation of two 2-mm autologous fragments of endometrial tissue onto the inner surface of the abdominal wall of rats with surgically induced (experimenta) endometriosis failed to prevent further growth of endometrioid (EMT) and additive tumors, while treatment of animals with dinitrosyl iron complexes (DNIC) with glutathione (12.5 μmoles/kg, 10 injections in 10 days) suppressed tumor growth virtually completely. The histological analysis of EMT samples of GS-NO-treated rats revealed pathological changes characteristic of control (non-treated with GS-NO or DNIC) rats with experimental endometriosis. EPR studies established the presence of the active form of ribonucleotide reductase, a specific marker for rapidly proliferating tumors, in EMT samples of both control and GS-NO-treated animals. Noteworthy, in small-size EMT and adjacent tissues of DNIC-treated rats the active form of ribonucleotide reductase and pathological changes were not found. © 2014 Elsevier B.V.

Dinitrosyl iron complexes (DNIC) with glutathione exert a cytotoxic effect on endometrioid tumours in rats with surgically induced experimental endometriosis. Intraperitoneal treatment of rats (Group 1) with DNIC (12.5 μmoles/kg, daily, for 12 days), beginning with day 4 after the surgical operation (implantation of two 2 mm-thick uterine fragments onto the abdominal wall) followed by 14-day keeping of animals on a standard feeding schedule (without medication) resulted in complete inhibition of the growth of endometrioid implants (EMI) in the majority of experimental animals. The ratio of mean EMI volumes in control and experimental rats of Group 1 was 14:1. In Group 2 rats, the use of a similar treatment protocol 4 weeks after surgery changed this ratio to 1.4:1. Noteworthy, the decrease of this ratio was irrelevant to deceleration of EMI growth at later periods after surgery. The histopathological analysis of EMI samples from experimental rats of Group 2 demonstrated complete disappearance of endometrial cysts suggesting a cytotoxic effect of DNIC on the tumours. The data obtained demonstrate that DNIC with glutathione and, probably, with other thiol-containing ligands hold considerable promise in the design of drugs for treating endometriosis in female patients. © 2014 Elsevier B.V.

The cell wall of Gram-negative bacteria plays a crucial role in the bacteria resistance to certain antimicrobials and its adaptation to the host environment. The first line of cell defense is its outer membrane composed of glycerophos-pholipids in the inner monolayer, lipopolysaccharides (LPS) in the outer monolayer and integral proteins. To understand the details of the outer membrane structure we build a molecular model of the Ra-LPS/LPS bilayer containing 8 β-barrels of the OmpA protein. We pay particular attention to the structural evolution of the outermost part of the LPS molecules (O-antigen) and analyze its conformational behavior using different computational criteria. © 2015 IEEE.

We studied the effects of tactivin and splenic polypeptides on learning and memory of thymectomized animals. In 3-week rats, thymectomy blocked active avoidance conditioning. Injections of tactivin (0.5 mg/kg) during 1 month after surgery restored learning capacity; splenic polypeptides were ineffective.
