The present application provides a composition comprising periodates or persulfates for extracting nucleic acid from a microorganism, and a corresponding method of using the composition for nucleic acid extraction. The composition and method are particularly useful in extracting nucleic acid from microorganisms that are generally resistant to standard nucleic extraction techniques, such as, one or more species of the Mycobacterium genus, one or more species of the M. tuberculosis complex, MDR strains of M. tuberculosis, one or more species of Clostridium, one or more species of Bacillus, and other microorganisms with hardy cell walls.

The present invention relates to compositions and methods for preserving and extracting nucleic acids from saliva. The compositions include a chelating agent, a denaturing agent, buffers to maintain the pH of the composition within ranges desirable for DNA and/or RNA. The compositions may also include a reducing agent and/or antimicrobial agent. The invention extends to methods of using the compositions of the invention to preserve and isolate nucleic acids from saliva as well as to containers for the compositions of the invention.

Methods, compositions, and kits for stabilizing both human and microbial deoxyribonucleic acid (DNA) present in complex biological samples, such as feces, are disclosed. In particular, aqueous compositions for stabilizing DNA contained in biological samples at ambient temperature are disclosed, together with associated methods and kits using same. In one aspect, the compositions comprise a chelating agent present at a concentration of at least about 150 m M, and the composition has a pH of at least about 9.5.

The present invention generally relates to a sample receiving device for releasably storing a substance. The sample receiving device includes a lid having a reservoir for retaining the substance, and a pierceable barrier sealing the substance within the reservoir; and b) a funnel for receiving a sample and configured for closure by the lid. The funnel is configured for releasable attachment to a sample receptacle such that a sample can be provided to the funnel and travel through the channel in the funnel into the sample receptacle. Further, the funnel includes one or more cutting ribs for cutting the pierceable barrier such that upon cutting of the pierceable barrier the substance is released from the reservoir, flows through the channel in the funnel and into the sample receptacle to be mixed with the sample. The present invention also provides a kit for collecting and storing biomolecules.

The present application provides a sample receiving device comprising a vial, a receptacle in communication with the vial for receiving the sample, and a cap comprising a pusher, the pusher for engaging with the sample in the receptacle. The receptacle comprises a disrupting means for disrupting the sample when the pusher engages with the sample in the receptacle and expels the disrupted sample into the vial. Typically, the device can be used for collecting fecal samples. A method of preserving a biomolecule in the device using a biomolecule preserving composition is also provided.

The present application is to provide a composition and method for stabilizing and maintaining the viability of hardy microorganisms from sample collection to downstream analysis. In particular, there is a method for preserving viable hardy bacteria, such as Mycobacteria, Bacillus anthracis, or Clostridium difficile, in a biological sample, comprising contacting the biological sample with a stabilization composition, wherein the stabilization composition comprises a chelating agent, a denaturing, a salt and has a p H between about 6 and about 11.

The present invention provides a container system for releasably storing a substance. The container system includes a vial having a sample storage chamber and a piercing member for piercing is membrane in the lid, which membrane seals a substance within a reservoir in the lid until the membrane is pierced by the piercing member. The container system optionally includes a funnel. There is also provided a method and kit for use of such a container system.

The present invention provides a composition and method for stabilizing ribonucleic acid (RNA) from biological samples such that the ribonucleic acid within the sample remains stable at room temperature. The composition comprises an anionic detergent and a buffering agent at a pH of about 5 to about 8.2 and is used in methods for extracting and storing ribonucleic acid from the biological sample.
