The male-specific, human Y-chromosomal short tandem repeats (Y-STRs) are very useful in forensic analysis and human evolution studies. The authors report two sexual crime cases in which the perpetrators were successfully traced using Y-haplotype screening of local patrilineages followed by autosomal STR typing. First, several main local patrilineages from local cases were investigated using Y-STR haplotyping, aimed to find the pedigrees whose haplotypes were identical or similar to those of the crime scene samples. Then, several key suspects were defined from the screened pedigrees, and autosomal STR typing was performed to identify the perpetrator of the crime. The application of Y-haplotype screening of local patrilineages followed by autosomal STR typing in these two cases demonstrates its usefulness for solving sexually related crimes in certain populations. © 2011 American Academy of Forensic Sciences.

A reliable and sensitive liquid chromatography-ion trap-time of flight tandem mass spectrometric assay (LCMS-IT-TOF) coupled with accelerated solvent extraction (ASE) was developed to identify and quantify six typical heterocyclic aromatic amines (HCAs), including 2-amino-3-methylimidazo[4,5-f]quinoline (IQ), 2-amino-3,4-dimethylimidazo[4,5-f]quinoline (MeIQ), 2-amino-3,8-dimethylimidazo[4,5-f]quinoxaline (MeIQx), 2-amino-1-methyl-6-phenylimidazo[4,5-b]pyridine (PhIP), 2-amino-9H-pyrido[2,3-b]indole (AαC) and 2-amino-3-methyl-9H-pyrido[2,3-b]indole (MeAαC) in cooked meat products. The effects of various experimental factors on separation and detection were investigated, and the fragmentation patterns of six HCAs were discussed. The method has shown high reproducibility with intra-day and inter-day precision (RSD, %) less than 6.16% across three quality control levels for the six analytes. The assay was linear over the concentration range of 10 to 1000 μg L-1 for IQ, MeIQ and 5 to 500 μg L-1 for MeIQx, PhIP, AαC and MeAαC (r2 ≥ 0.996). The experimental results showed that the proposed method can be used successfully to identify and determine six typical HCAs at ultra-trace levels (μg kg-1) in cooked meat products. © 2015 The Royal Society of Chemistry.

Objective: To explore the effectiveness of direct amplification for the STR analysis of cartilage, and to accelerate the effectiveness of disaster victim identification. Methods: Eighty-eight cartilage samples were directly amplified by PowerPlex® 21 kit, and the results of genotyping were compared with that obtained by the magnetic beads method. Results: In 88 cartilage samples, the STR genotypes were successfully detected from 84 samples by direct amplification and magnetic beads method, and both the results of genotyping by two method were consistent. Conclusion: Direct amplification with PowerPlex® 21 kit can be used for STR genotyping of cartilages. This method is operated easily and promptly, which has a potential application in the individual identification of mass disasters. © 2017 by the Editorial Department of Journal of Forensic Medicine.

Morphological changes of the sternum occured regularly with the increase of age in the process of development and degradation. A great deal of researches of relation between morphological changes of the sternum and age have been done by domestic and foreign scholars and some valuable data which laid the foundation for the threedimensional image reconstruction of the sternum for the study on the morphological changes were accumulated. The introduction of 3-dimensional reformation technology is expected to lead to breakthrough in sternum age estimation of living person.

In this paper, immunosensors based on a self-assembly modified (SAM) Au electrode for the specific detection of two illicit drugs, morphine (MO) and methamphetamine (MA), are reported. Using 3-mercaptopropionic acid SAM and activation by 1-ethyl-3-(3-dimethylaminoprophyl) carbodiimide (EDC)/n-hydroxysulfsuccinimide (NHS) with an adequate quantity of the antibody of MO or MA immobilized onto the surface to act as the sensing host, the sensors specifically respond to their respective target based on specific covalent bonding. Electrochemical impedance spectroscopy (EIS) is used as the read-out signal. The value of the electron transfer resistance (Ret) responded to the concentration of drug in spiked blood samples with an ultra-low detection limit and satisfactory recovery. In conclusion, the developed sensors are demonstrated to be more advantageous compared to those previously reported in terms of sensitivity. © 2014 Elsevier B.V. All rights reserved.

Objective: To provide reference for the determination of postmortem time according to the law of seasonal succession of necrophagous fly in Qingdao region. Methods: The outdoor place was set in the field and the pig carcasses was continuously put for observation from 1st, April, 2012 to 1st, April, 2013. The pig carcasses were put at 6:00 of the first day of each month, and the occurrence of necrophagous fly was observed 20min after, then was observed every one hour till 18:00 of this day; 3 days after, the observation was carried out at 7:00, 12:00 and 18:00 respectively. The insects appeared around the pig carcasses were collected and indentified, the sorts, phase and amount were record. Results: The result showed that the necrophagous flies can attack the pig carcasses from April to November; there are 23 species of necrophagous flies that belong to 3 families were record. Among these flies, Lucilia sericata, Chrysomyia megacephala and L. illustris took the first three positions for the amount, with the proportion of 40.71% 31.95% and 7.12% respectively. The amount of L. sericata, and C. megacephala appeared at noon were obviously greater than that appeared at dusk. The amount of different sorts of flies appeared at noon were all greater than that appeared at morning and dusk through the whole year. The difference was significant at first ten days of April, while not significant at July and August. Most necrophagous flies appeared at the "fresh stage" and "swelling stage" of the pig carcasses. Conclusion: The law of seasonal succession of necrophagous flies indicated by our experiment can provide reference for the determination of postmortem time in Qingdao region, and can provide support to the construction of regional database of necrophagous flies.

Objective: To analyze suicide scene, damage morphology and case investigation of mountain type scenic spot for giving the references to determine the nature of cliff suicide cases. Methods: The suicide cases collected from 2002 to 2012 in scenic spot of Mount Huangshan. The age, gender, native place, case location, damage inspection and behavior were analyzed. Results: In the 52 suicide cases, the suicide rate of male was higher than that of female. The numbers from other provinces were higher than that of local province. The age was mainly range from 19 to 50. The time of suicide cases mostly happened between 16:00 to 24:00. The major damage was compound injury with varying degrees of traumatic brain injury, organic injury of pleuroperitoneal cavity and surface bruise and scratch. Conclusion: In order to determine the nature of cliff suicide cases, it needs to work synthetically in the investigation of crime scene and interview. © 2015 by the Editorial Department of Journal of Forensic Medicine.
