To ascertain the genetic basis of a paroxysmal nocturnal hemoglobinuria (PNH) case without somatic mutations in PIGA, we performed deep next-generation sequencing on all exons of known genes of the glycosylphosphatidylinositol (GPI) anchor synthesis pathway. We identified a heterozygous germline splice site mutation in PIGT and a somatic 8-MB deletion in granulocytes affecting the other copy of PIGT. PIGA is essential for GPI anchor synthesis, whereas PIGT is essential for attachment of the preassembled GPI anchor to proteins. Although a single mutation event in the X-chromosomal gene PIGA is known to cause GPI-anchored protein deficiency, 2 such hits are required in the autosomal gene PIGT. Our data indicate that PNH can occur even in the presence of fully assembled GPI if its transfer to proteins is defective in hematopoietic stem cells. Source

The present invention is directed to a method of diagnosis and prognostication of a hepatobiliary tumor, the method comprising the step of determining expression of MACC1 polypeptide or of a nucleic acid encoding said MACC1 polypeptide in a biological sample.

The present invention relates to a phosphate adsorbent on the basis of maghemite or maghemite/magnetite comprising (i) an iron oxide core comprising a crystal structure of inverse spinel iron oxide, (ii) a coating selected from monomeric carbohydrates, in particular monosaccharides or disaccharides, alditols, or mixtures thereof, and/or (iii) a pharmaceutical excipient selected from polymeric carbohydrates, wherein the phosphate adsorbent has the form of nanoparticles with a particle size of the iron oxide core (i) of less than 20 nm. The present invention further relates to a method for the production of a phosphate adsorbent on the basis of maghemite or maghemite/magnetite, to pharmaceutical compositions comprising the phosphate adsorbent, and to medical uses thereof, especially for the prevention and/or treatment of hyperphosphatemia.

The present invention relates to S-enantiomerically enriched compositions of beta blockers and uses thereof, including uses of the beta blocker compositions for treating amyotrophic lateral sclerosis. The beta blocker compositions can also be used for preventing loss of lean mass, preventing body weight loss in subjects, improving quality of life in subjects, and prolonging survival in amyotrophic lateral sclerosis patients. The beta blocker can be oxprenolol or a pharmaceutically acceptable salt thereof.
