The electronic circular dichroism (ECD) spectra of two sesquiterpenoids (1 and 2) related to oplopanone, obtained from a methanolic extract of the plant Serphidium stenocephalum (Artemisia stenocephala), were measured and reproduced by means of time-dependent density functional theory (TDDFT) calculations, establishing their absolute configuration. The application of ketone octant rule for carbonyl n-π* ECD band to compounds 1 and 2, which include an acyclic carbonyl group, was critically assessed. The peculiar oplopanone skeleton makes a straightforward application of the octant rule impossible, because of the uncertainty related to the shape of the so-called third nodal surface separating front and back octants. The various group contributions to the carbonyl n-π* ECD band were estimated with TDDFT calculations on selected molecular models obtained by consecutive dissections from 1. Studies on the aerial parts of Buddleja crispa yielded 13 known compounds, nonyl benzoate, hexyl p-hydroxy-cinnamate, ginipin, gardiol, 1-heptacosanol, steroidal galactoside (22 R)-stigmasta-7,9 (11)-dien-22 β-ol-3α-O- β- D-galactopyranoside, 3-methoxy benzoic acid, β-sitosterol and ursolic acid. Besides this two iridoid galactosides buddlejosides A, buddlejosides B and a benzofuran-type sesquiterpene buddlejone have been isolated from the ETOAC fraction of B. crispa. Together with the above compounds, methyl benzoate (1) and 3-methoxy-4-hydroxy benzoic acid (2) were also isolated. Compound 2 (C8H8O4) was identified by comparison of its data with those reported earlier, which was originally isolated from Onosma hispidum, and this is the first report of its isolation from this species. For compounds 1 and 2, the total alcoholic soluble extract, methanol soluble, chloroform soluble, ethyl acetate soluble and petroleum ether soluble extract of the aerial parts of B. crispa were screened for nematicidal activity against nematodes of freshly hatched second-stage juveniles of Meloidogyne incognita (root-knot nematode), exhibiting 92%, 40%, 88%, 83%, 82% and 50% mortality, respectively, of eloids M. incognita at 0.5% concentration. Compound 1 was more potent than the nematicide Azadirachta indica at the same concentration. Negative results were obtained for the nematicidal activity of petroleum ether extract of B. crispa leaves. © 2010 Taylor & Francis.

Objective: To determine the association of ABCB1polymorphism G2677T with anti-emetic efficacy in patients treated with ondansetron for preventing postoperative nausea and vomiting. Study Design: A clinical trial. Place and Duration of Study: Combined Military Hospital, Rawalpindi and Institute of Biomedical and Genetic Engineering, Islamabad, from 2012 to 2013. Methodology: Four mg ondansetron was administered intravenously 30 minutes before the end of surgery. A total of 246 patients with the complaints of nausea and vomiting and 244 patients without nausea and vomiting were analyzed for G2677T polymorphism using PCR-RFLP method. Results were described as frequency percentages and chi-square test with significance at p < 0.05. Results: The patients with TT genotype had significantly lower incidence of postoperative nausea and vomiting during the first 2 hours (p < 0.001) and between 2 - 24 hours after surgery as compared to other genotypes (p < 0.001). The patients with GG genotypes had significantly higher incidence of this complaint (p=0.014). Conclusion: Polymorphism of ABCB1 has an association with responsiveness for ondansetron. There is a role for genetics in the management of PONV.

To investigate the frequency of the single nucleotide polymorphism C1236T in exon 12 of the ABCB1 gene in Pakistani population and to compare it with published data on Asian and Caucasian populations.Across-sectional observational study.Combined Military Hospital, Rawalpindi and Institute of Biomedical and Genetic Engineering (IBGE), Islamabad, from August 2012 to May 2013.C1236T polymorphism was investigated in 426 Pakistani subjects. The frequency was compared with the published data on other Asian and Caucasian populations.The frequencies of ABCB1 C1236T were 16.4% for CC, 44.1% for CT and 39.4% for TT. Pakistanis differed significantly from all the European populations compared in the distribution of the TT genotype of C1236TABCB1 (p < 0.05). The Pakistani population also differed significantly from some of the European populations in the distribution of CC and CT genotype (p < 0.05).There was significant difference in the genotype frequency of the ABCB1 gene compared to other populations. This study has provided a framework for future pharmacogenetic and pharmacokinetic studies on this polymorphic variant of ABCB1 gene in the Pakistani population.

Objective: To determine the effect of smoking on the post-operative nausea and vomiting (PONV) as well as severity of pain in the post-operative period. Methodology: A total of 147 patients undergoing elective laparoscopic cholecystectomy under general anesthesia were divided into 2 groups of non-smokers (n=75) and smokers (n=72). In the first 24 hours after surgery the frequency of PONV and the severity of pain on Visual Analogue Score were assessed. Results: The group of non-smokers had statistically more nausea and vomiting (n=59, 78.6%) as compared to that of smokers (n=20, 27.7%) [p<0.05]. However there was no significant difference in the maximum pain scores in both the groups (p>0.05). Conclusion: We conclude that frequency of PONV is less in smokers as compared to non-smokers.

We use a distributed modeling approach to investigate the spatial and quantitative distribution of incoming global solar radiation (GSR) falling over the complex terrain using remote sensing (RS) and geographical information system (GIS). To apply distributed model, we use SRTM digital elevation models (DEM) for impact assessment of slope, aspect orientation and correcting possible sunshine duration over the inclined surface. Similarly, to evaluate the impact of cloud cover on incoming global solar radiation amount and spatial distribution, we use MODIS Level-2 (L2) cloud fraction products and observational data of cloud cover from 21 meteorological observatories in Pakistan. As an input for incoming GSR model, the direct, diffused, and terrain reflected components of GSR were employed. The model results show clear quantitative influences of local topography based on slope angles and cloud cover variations on the distribution of GSR in the North and Northwest of Pakistan. Similarly, slope and aspect influences are evident over the mountainous areas in Himalaya, Karakorum and Hindukush Ranges in North and Suleiman Ranges in Northwest. Furthermore, terrain effect for eastern and western slopes is not significant; the slopes receive almost the same radiation due to the orientation of topography and latitudinal consistency, but inter-shielding effect in eastern-western oreintation is more prominent compared to the one in northern-southern. © 2015 IEEE.

Vitex agnus-castus Linn. (Verbenaceae) is locally known as "Hub-el-faked" and "Sumbhalu-ke-bij". It is abundantly found in Baluchistan (Pakistan) and extensively used for medicinal purpose. Methyl 4-hydroxybenzoate (1) is a very well known compound, was isolated from the aerial parts of Vitex agnus-castus. Methyl 4-hydroxybenzoate (1) was found to be a moderate immunomodulatory and non-cytotoxic activities. The activity was tested in vitro for chemiluminescence, chemotaxis, T-cell proliferation and cytotoxicity. Methyl 4-hydroxybenzoate (1) exhibited a moderate inhibitory effect on oxidative burst of neutrophils in a dose dependent manner. It was found to have a moderate suppressive effect on the chemotaxic action at lower concentrations on fMLP (10 -8 M) stimulated neutrophils. It also showed a suppressive effect on PHA stimulated T-cell (PMBC).

Two new acylated flavonol glycosides, 3-O-{[2-O-β-D-glucopyranosyl]-3- [O-β-D-glucopyranosyl]-4-[(6-O-p-coumaroyl)-O-β-D-glucopyranosyl]} -α-L-rhamnopyranosyl-kaempferol 7-O-α-L-rhamnopyranoside and 3-O-{2-[(6-O-p-coumaroyl)-O-β-Dglucopyranosyl]-3-[O-β-D- glucopyranosyl]-4-[(6-O-p-coumaroyl)-O-β-D-glucopyranosyl]} -α-L-rhamnopyranosyl-kaempferol 7-O-α-L-rhamnopyranoside, trivially named as brauhenefloroside E (1) and F (2), respectively, were isolated from the fruits of Stocksia brauhica and their structures were elucidated using spectroscopic methods, including 2DNMRexperiments.
